Cloning of endothelin-1 (ET-1) from European sea bass (Dicentrarchus labrax) and its gene expression analysis in larvae with retinoic acid-induced malformations ArchiMer
Geay, Florian; Darias, Maria J.; Santigosa, E.; Desbruyeres, Elisabeth; Quazuguel, Patrick; Zambonino, Jose-luis; Cahu, Chantal; Mazurais, David.
It is known in vertebrates that endothelin-1 (ET-1) plays a key role ill morphogenesis whose expression level greatly influences the development of craniofacial malformations. In the Present study, the complete cDNA fragment encoding a precursor of endothelin-1, the preproendothelin-1 (PPET1), was cloned by RACE-PCR from European sea bass. The cDNA encoded a 199 amino acid polypeptide that was composed of the "mature" and "big" ET-1. Relative ET-1 expression levels were investigated in European sea bass larvae fed microparticulate diets containing the standard amount (0.08 g retinol/kg diet: group N) or all excess of retinoic acid which induces skeletal malformations (0.5 g retinol/kg diet: group RA). Real-time reverse transcriptor polymerase chain...

Mazurais, David; Coves, Denis; Papandroulakis, Nikos; Ortega, Aurelio; Desbruyeres, Elisabeth; Huelvan, Christine; Le Gall, Marie-madeleine; De La Gandara, Fernando; Cahu, Chantal.
The aim of this study was to determine whether mortality observed during the larval development of reared bluefin tuna (Thunnus thynnus) could be related to improper expression profiles of key genes involved in digestive or antioxidant response capabilities. Tuna larvae were sampled at hatching, 2, 5, 10, 15 and 20 dph (days post hatching) for the relative quantification of transcripts encoded by genes involved in digestive [trypsinogen 1 (TRYP1), alpha-amylase (AMY), aminopeptidase N (ANPEP)] and antioxidant [catalase (CAT)] functions. The levels of expression of ANPEP related to the development and maturation of intestinal function increased from 5 to 20 dph. Furthermore, AMY and TRYP1 genes, which are pancreatic enzymes implicated in carbohydrate and...

Identification of Hypoxia-Regulated Genes in the Liver of Common Sole (Solea solea ) Fed Different Dietary Lipid Contents ArchiMer
Mazurais, David; Ferraresso, Serena; Gatta, Pier Paolo; Desbruyeres, Elisabeth; Severe, Armelle; Corporeau, Charlotte; Claireaux, Guy; Bargelloni, Luca; Zambonino, Jose-luis.
Coastal systems could be affected by hypoxic events brought about by global change. These areas are essential nursery habitats for several fish species including the common sole (Solea solea L.). Tolerance of fish to hypoxia depends on species and also on their physiological condition and nutritional status. Indeed, high dietary lipid content has been recently shown to negatively impact the resistance of sole to a severe hypoxic challenge. In order to study the molecular mechanisms involved in the early response to hypoxic stress, the present work examined the hepatic transcriptome in common sole fed diets with low and high lipid content, exposed to severe hypoxia. The activity of AMP-activated protein kinase (AMPK) was also investigated through the...

Impact de la pisciculture marine intensive sur l'environnement : contribution à la modélisation de l'utilisation de l'azote par le turbot ArchiMer
Dosdat, Antoine; Tetu, N.; Metailler, Robert; Huelvan, Christine; Desbruyeres, Elisabeth; Le Delliou, Herve.

The effects of dietary marine protein hydrolysates on the development of sea bass larvae, Dicentrarchus labrax, and associated microbiota ArchiMer
Delcroix, Jeremy; Gatesoupe, F.j.; Desbruyeres, Elisabeth; Huelvan, Christine; Le Delliou, Herve; Le Gall, Marie-madeleine; Quazuguel, Patrick; Mazurais, David; Zambonino-infante, Jose-luis.
Protein hydrolysate is an essential component of dry starter diets for fish larvae, as promoting healthy development. Peptides are also suitable substrates for many intestinal microbes. Five experimental diets were compared to a control diet (CONT) supplemented with a commercial fish protein hydrolysate. Each diet contained one marine protein hydrolysate, which differed by the proportion of di- and tri-peptides, and by raw materials. Two diets (HYD4 and HYD5) stimulated larval growth compared with CONT. Two other diets (HYD1 and HYD2) yielded inferior growth. HYD1 was detrimental for survival, which was associated with an up-regulation of genes involved in inflammation and antioxidative responses, while the gene coding for osteocalcin was down-regulated in...

Comparison of brown trout (Salmo trutta) reared in fresh water and sea water to freshwater rainbow trout (Oncorhynchus mykiss): II. Phosphorus balance ArchiMer
Dosdat, Antoine; Metailler, Robert; Desbruyeres, Elisabeth; Huelvan, Christine.
Brown trout and rainbow trout (average weight 100 g) were reared in fresh water at 12 degrees C under the same conditions before transferring brown trout to sea water, in order to compare phosphorus utilisation in both species. Apparent phosphorus availability, orthophosphate excretion and phosphorus accretion in the fish were directly determined. Thus, actual phosphorus mass balance was built. Rainbow trout raised in fresh water had a higher phosphorus retention coefficient (maximum 50 %) than brown trout reared in fresh water (maximum 45 %). Transferring brown trout to sea water induced a reduction in phosphorus retention (maximum 39 %), Orthophosphate excretion, ranging 7-20 mg phosphorus per kg wet weight per day, represented 10-20 % of ingested...

Comparison of brown trout (Salmo trutta) reared in fresh water and sea water to freshwater rainbow trout (Oncorhynchus mykiss) .1. Growth and nitrogen balance ArchiMer
Dosdat, Antoine; Metailler, Robert; Desbruyeres, Elisabeth; Huelvan, Christine.
Brown trout and rainbow trout (average weight 100 g) were reared in fresh water at 12 degrees C under the same conditions before transfer of brown trout to sea water, in order to compare nitrogen utilisation in the two species. Apparent protein digestibility (ADC), nitrogen (ammonia and urea) excretion, protein productive value (PPV) and actual observed nitro en mass balance were determined. Rainbow trout raised in fresh water had a higher growth rate (1.1 vs 0.8%. d(-1)), better food conversion ratio (0.7 vs 1.0), better ADC (91 vs 85%) and PPV (45 vs 35%) and lower ammonia excretion rates than brown trout reared in fresh water. Transferring brown trout to sea water induced lower PPV (30%) and ammonia and urea excretion. Salinity did not modify metabolic...

Dietary vitamin D-3 affects digestive system ontogenesis and ossification in European sea bass (Dicentrachus labrax, Linnaeus, 1758) ArchiMer
Darias, Maria J.; Mazurais, David; Koumoundouros, Giorgos; Glynatsi, Nomiki; Christodoulopoulou, Stavroula; Huelvan, Christine; Desbruyeres, Elisabeth; Le Gall, Marie-madeleine; Quazuguel, Patrick; Cahu, Chantal; Zambonino-infante, Jose L..
The influence of dietary vitamin D-3 (VD3) on survival, growth and morphogenesis during the larval development of European sea bass (Dicentrarchus labrax) was evaluated until 45 days post hatching. Diets contained 4% of the standard vitamin mix (VM) recommended by the National Research Council (NRC) and incorporated 0, 19.2, 38.4, or 140 IU of VD3 per gram of diet to give VD-0, VD-1, VD-2 and VD-3 dietary treatments, respectively. The present study revealed for the first time an impact of dietary VD3 on the sea bass digestive system ontogenesis that consequently conditioned the ossification process and morphogenesis. All dietary VD3 levels were in the "adequate range" based on larval survival. Nevertheless, growth, intestinal maturation and ossification at...
