The potential of an Aspergillus fumigatus Brazilian strain to produce antimicrobial secondary metabolites BJM
Furtado,Niege Araçari Jacometti Cardoso; Fonseca,Maria José Vieira; Bastos,Jairo Kenupp.
During the course of screening for biologically leading active natural products from fungi, a strain of Aspergillus fumigatus was assessed for antimicrobial activity production. Four parameters consisting of medium culture, pH, temperature and time of fermentation were investigated. The extracts from the fungus cultures displayed different TLC profiles and produced, at least, one inhibition zone in the bioautography assays. Overlapping inhibition zones in the range of Rf 0.09 to 0.65 against Kocuria rhizophila were detected in the bioautography assay of the chloroform extract obtained from Jackson's medium culture, initial pH of 6.0 and incubation at 40ºC for 144 hours. However, the obtained extracts by developing the fungus culture in Vogel's medium...

Anti-inflammatory and toxicity studies of atranorin extracted from Cladina kalbii Ahti in rodents BJPS
Melo,Marcelia Garcez Dória de; Araújo,Adriano Antunes de Souza; Serafini,Mairim Russo; Carvalho,Larissa Feitosa; Bezerra,Marília Santos; Ramos,Cledison Santos; Bonjardim,Leonardo Rigoldi; Albuquerque-Júnior,Ricardo Luiz Cavalcanti; Lima,Julianeli Tolentino; Siqueira,Rosana Souza; Fortes,Vanessa Silveira; Fonseca,Maria José Vieira; Quintans-Júnior,Lucindo José.
Atranorin (ATR) is the main compound from the lichen Cladina kalbii Ahti, which grows in the arid regions of northeastern Brazil. This study was conducted to evaluate the anti-inflammatory and toxicological properties of ATR. To evaluate anti-inflammatory properties, paw edema was induced by injecting 0.1 mL of carrageenan into the subplantar region of the right hind paw of rats, and leukocyte migration was induced by injection of 500 µL of carrageenan into the peritoneal cavity of mice. In addition, we determined ATR cytotoxicity in L929 cells by MTT assay and acute (5 g/kg-single dose) and subchronic (50 mg/kg-30 days) toxicity tests in Wistar rats. The results showed that ATR (100 mg/kg and 200 mg/kg) exhibited significant anti-inflammatory activity...

Antioxidant activity and physical-chemical properties of spray and spouted bed dried extracts of Bauhinia forficata BJPS
Souza,Claudia Regina Fernandes; Georgetti,Sandra Regina; Salvador,Marcos José; Fonseca,Maria José Vieira; Oliveira,Wanderley Pereira.
Two distinct drying methods (spouted bed and spray drying) were used for production of dried extracts of Bauhinia forficata Link (Leguminosae, Caesalpinoideae). High-quality powder products in terms of physical and chemical properties were obtained. HPLC fingerprints revealed that the chromatographic profiles of flavonoid compounds present in the dried extract did not change significantly, due to drying. In general, the spouted bed drying caused a degradation of total flavonoids than was lower than that of the spray drying. Antioxidant properties of the dried extract, examined by their radical scavenging activity using 2,2-diphenyl-1-picrylhydrazyl radical (DPPH•) and inhibition of lipid peroxidation induced by Fe+2 assays (LPO), confirmed their...

Assessment of in vitro methodologies to determine topical and transdermal delivery of the flavonoid quercetin BJPS
Vicentini,Fabiana Testa Moura de Carvalho; Georgetti,Sandra Regina; Bentley,Maria Vitória Lopes Badra; Fonseca,Maria José Vieira.
To be effective against the oxidative damages induced by UVB irradiation in the skin, the drug needs to release from the formulation in which it was incorporated and reach the skin layers where the ROS are generated. Thus, it is very important the development of a robust and sensitive methodology to extract and quantify in different skin layers the antioxidant agent delivered from topical formulations. Therefore, in the present work suitable methods to extract and quantify quercetin in skin samples and receptor phase after in vitro penetration studies were developed. The results demonstrated that the recovery from two different layers of skin, the SC and [E+D], using two different methods of quantification (DPPH• assay and HPLC, respectively), was 93.8 %...
