Summer mortality associated with juveniles of the oyster Crassostrea gigas is the result of a complex interaction between the host, pathogens and environment. Divergent selection based on summer survival have been applied to produce families with resistant and susceptible progeny. The French project "Morest", coordinated by IFREMER, aims to understand the causes of the summer mortalities in Crassostrea gigas. In 2001, three sets of families were bred following a nested half-sib mating design. 17 half-sib families (HSF) were obtained in this first generation (G1) and reared in 3 sites. Significant differences in survival were observed among HSF, and some HSF showed high levels of mortality in all sites, clearly indicating a genetic basis for survival. This study investigated the potential of RNA interference, which is technically challenging in bivalve mollusc species, to assess gene function in the oyster Crassostrea gigas. We designed dsRNA targeting the oyster vasa-like gene (Oyvlg), specifically expressed in oyster germ cells. In vivo injection of oyvl-dsRNA into the gonad provokes a knockdown phenotype corresponding to germ cell underproliferation and prematurely arrested meiosis througout the organ. AMP-activated protein kinase α (AMPKα) is a key regulator of energy balance in many model species during hypoxia. In a marine bivalve, the Pacific oyster Crassostrea gigas, we analyzed the protein content of adductor muscle in response to hypoxia during 6 h. In both smooth and striated muscles, the amount of full-length AMP-activated protein kinase α (AMPKα) remained unchanged during hypoxia. The concentration and spatial distribution of microplastics in the Bay of Brest (Brittany, France) was investigated in two surveys. Surface water and sediment were sampled at nine locations in areas characterized by contrasting anthropic pressures, riverine influences or water mixing. Microplastics were categorized by their polymer type and size class. Microplastic contamination in surface water and sediment was dominated by polyethylene fragments (PE, 53–67%) followed by polypropylene (PP, 16–30%) and polystyrene (PS, 16–17%) microparticles. We investigated the role of oyster gonadal TGF beta (og-TGF beta) in the reproduction of Crassostrea gigas, using an in vivo RNA interference approach. We designed double-stranded RNA targeting og-TGF beta, which is specifically expressed in the somatic cells surrounding germ cells in the gonad of both male and female oysters. In the oyster Crassostrea gigas consumption-related traits, amylase properties and growth were found to be linked through genotypes that differed for polymorphism in the two amylase genes AMYA and AMYB. Modulation of AMYA mRNA level had already been observed in response to food availability, whereas the functional role of AMYB was still unknown. Field characterization of summer mortality was performed in France in the frame of the Morest project. Natural and hatchery spat were compared between three oyster production areas in France. Regardless of the natural or hatchery origin, oysters died during the reproduction period after temperature reaches 19 degree C. Thus, in southern areas, temperature accelerated gametogenesis of small spat (10mm) as well as adults, and mortality appeared for the two age classes. Through differential screening between oyster families selected for high and low summer survival, we have characterized a new transforming growth factor-beta (TGF-beta) superfamily member. This novel factor, named oyster-gonadal-TGF beta-like (og-TGF beta-like), is synthesized as a 307 amino acid precursor and displays 6 of the 7 characteristic cysteine residues of the C-terminal, mature peptide.
