Utilização de penas de galinha para produção de queratinase por Aspergillus carbonarius PAB

O objetivo deste trabalho foi estudar a produção de queratinase por Aspergillus carbonarius URM 1546, tendo-se como substrato penas de galinha, por meio de planejamento fatorial completo 2³. Todos os parâmetros estudados e as interações de segunda ordem foram estatisticamente significativos. A maior atividade queratinolítica (48,9 U mL-1) foi obtida com 120 rpm, 0,5% (p/v) de penas de galinha e sete dias de cultivo.

Avaliação de variáveis que influenciam a hidrólise enzimática da caseína do leite de cabra Moxotó PAB

O objetivo deste trabalho foi avaliar os efeitos da temperatura, pH, relação enzima:substrato (E:S) e tempo de reação sobre a hidrólise enzimática da caseína do leite de cabra Moxotó, com uso de diferentes enzimas proteolíticas. A hidrólise enzimática da caseína caprina foi realizada com uso de tripsina, pepsina e papaína. Para cada enzima, foram utilizados dois níveis de cada variável, na avaliação de seus efeitos sobre a hidrólise da caseína, em um fatorial completo 2(4). Os produtos da hidrólise foram visualizados em eletroforese SDS-PAGE. Production of extracellular lipase by the phytopathogenic fungus Fusarium solani FS1 Rev. Microbiol.

A Brazilian strain of Fusarium solani was tested for extracellular lipase production in peptone-olive oil medium. The fungus produced 10,500 U.l-1 of lipase after 72 hours of cultivation at 25oC in shake-flask at 120 rpm in a medium containing 3% (w/v) peptone plus 0.5% (v/v) olive oil. Glucose (1% w/v) was found to inhibit the inductive effect of olive oil. Peptone concentrations below 3% (w/v) resulted in a reduced lipase production while increased olive oil concentration (above 0.5%) did not further stimulate lipase production. The optimum lipase activity was achieved at pH 8.6 and 30oC and a good enzyme stability (80% activity retention) was observed at pH ranging from 7.6 to 8.6, and the activity rapidly dropped at temperatures above 50oC. Purification of plasmid (pVaxLacZ) by hydrophobic interaction chromatography BABT

This paper describes a method for the plasmid DNA purification, which includes an ammonium sulphate precipitation, followed by hydrophobic interaction chromatography (HIC) using Phenyl Sepharose 6 Fast Flow (low sub). The use of HIC took advantage of the more hydrophobic character of single stranded nucleic acid impurities as compared with double-stranded plasmid DNA.

Comparative performance of microbial lipases immobilized on magnetic polysiloxane polyvinyl alcohol particles BABT

Microbial lipase from Mucor miehei and Candida rugosa were immobilized by covalent binding onto magnetized polysiloxane polyvinyl alcohol particles (POS-PVA). The resulting immobilized derivatives were evaluated in aqueous solution (olive oil hydrolysis) and organic solvent (butyl butyrate synthesis). Higher catalytic activities were found when the coupling procedure was made with M. miehei lipase. Immobilized M. miehei lipase also showed a better operational stability and a higher half-life than C. rugosa lipase after the successive batches of esterification. Aspergillus niveus Blochwitz 4128URM: new source for inulinase production BABT

Aspergillus niveus Blochwitz 4128 URM isolated from sunflower rhizosphere demonstrated a new source of inulinase. The enzyme was produced in culture medium containing inulin as substrate in the concentrations: 10, 15 and 20g L-1. Maximum enzyme activity was obtained in medium containing 20g L-1 inulin. The enzyme was partially purified using ammonium sulphate precipitation, followed by ion charge (DE-32) and molecular exclusion (Sephadex) chromatography. The results showed the optimal pH and temperature of inulinase from crude extract were 4.0 and 4.8 and 45ºC, respectively. The enzyme was purified 34.65 fold with yield of 53.63%. A. niveus 4128URM can be used in the inulinase production with use in the food industries.

Immobilization of lipase from Fusarium solani FS1 BJM

Lipase from Fusarium solani FS1 was immobilized by covalent attachment to polyacrylamide beads and onto magnetized Dacron, retaining 12% and 97% of activity, respectively. Lipase was also entrapped within polyacrylamide beads, retaining 53% of activity. Investigations of the kinetic characteristics of the immobilized derivatives using triolein as substrate showed that lipase immobilized onto polyacrilamide beads and Dacron did not follow Michaelis-Menten kinetics.

PARTIAL CHARACTERIZATION OF PROTEASES FROM STREPTOMYCES CLAVULIGERUS USING AN INEXPENSIVE MEDIUM BJM

The partial characterization of extracellular proteases from Streptomyces clavuligerus NRRL 3585 and 644 mutant was investigated. The enzyme production was carried out in batch fermentation using soy bean filtrate as nitrogen source. Maximum activity was obtained after 96h of fermentation with an initial pH of 7.0. The enzyme was partially purified by ammonium sulphate precipitation. Enzymes from the two strains retained 37% of their initial activities at pH 8.0 after 2 h incubation at 25ºC. Enzyme half-life at pH 8.0 and 60ºC was 40.30 and 53.32 min, respectively for both strains (partially purified extract). Protoplast formation and regeneration from Streptomyces clavuligerus NRRL 3585 and clavulanic acid production BJM

Protoplasts of the wild type Streptomyces clavuligerus NRRL 3585 (ATCC 27064) were formed from spores cultures obtained in the lag, exponential and stationary growth phases by using 0.5% glycine in the culture medium. The protoplasts were obtained by treatment of the cells with lysozyme (EC-3.2.1.17) 40,000 U (1mg/mL), in an osmotic solution for 90 min at 28ºC. The frequency of regenerated protoplasts in the lag phase was 1.7x10³ CFU/mL (28.97%), in the beginning of the exponential phase 0.4x10² CFU/mL (31.67%), in the exponential growth phase 2.5x10³ CFU/mL (46.30%) and 1.0x10(5) CFU/mL in stationary phase (48.45%). O objetivo deste estudo foi identificar as proteínas do plasma seminal de caprinos da raça Alpina Americana criados na região Nordeste do Brasil que estão relacionadas ao índice pluviométrico e à qualidade do sêmen. O sêmen foi obtido pelo método de vagina artificial a partir de três reprodutores e foi avaliado quanto aos parâmetros macroscópicos e microscópicos. O perfil de proteínas do plasma seminal foi realizado por eletroforese bidimensional. Os parâmetros volume do sêmen, integridade do acrossoma e proteínas totais evidenciaram diferença significativa (P<0,05) entre os períodos de alto (1,7mL, 90,3% e 372µg mL-1, respectivamente) e baixo (1,2mL, 80,3% e 494µg mL-1, respectivamente) índice pluviométrico.
