Tratamento térmico e químico para controle da atividade da poligalacturonase no albedo do maracujá PAB

Resumo: O objetivo deste trabalho foi avaliar tratamentos térmicos e químicos com Ca2+, no controle da atividade da poligalacturonase e de seus efeitos no rendimento de extração e no grau de esterificação da pectina do albedo da casca de maracujá-amarelo (Passiflora edulis). Utilizaram-se cascas de frutos com coloração verde-clara, as quais foram lavadas, sanitizadas, trituradas e deixadas em repouso em água, para suspensão do albedo. O albedo foi recolhido e submetido à temperatura de 90oC a 30, 60 e 120 min, ou a concentrações de cálcio de 0,01 e 1% durante 1 hora de agitação de 400 rpm, em temperatura ambiente. Atividade da pectina metilesterase e da β-galactosidase durante o amadurecimento do mamão cv. golden Rev. Bras. Frutic.

Este trabalho foi realizado com o objetivo de investigar o comportamento das enzimas pectina metilesterase (PME) e β-galactosidase (β-Gal) durante o amadurecimento do mamão cv.'Golden'. Mamões com 15% de cor amarela foram estocados em câmara de refrigeração a 20ºC e 85-95 % UR, por um período de 11 dias. Durante este período de estocagem, foi determinada a cor da casca, firmeza e pH da polpa dos frutos, bem como a atividade das enzimas PME e β-Gal. A atividade da PME, observada ao longo do amadurecimento do mamão, indicou que esta enzima participou do processo de hidrólise da parede celular, principalmente no início do amadurecimento dos frutos. Este trabalho foi realizado com o objetivo de avaliar o efeito de atmosferas controladas contendo diferentes concentrações de oxigênio sobre a atividade das enzimas β-galactosidase e pectina metilesterase, e sobre a cor da casca e a firmeza da polpa de mamões 'Golden'. Os frutos foram mantidos por 36 dias, nas seguintes atmosferas controladas: 1% de O2 e 0,03% CO2 com adsorvedor de etileno, 3% de O2 e 0,03% de CO2 com adsorvedor de etileno, 5% O2 e 0,03% de CO2 com adsorvedor de etileno e atmosfera ambiente sem adsorvedor de etileno. Injúria pelo frio em frutos de mamoeiro (Carica papaya L.) cv 'Golden' Rev. Bras. Frutic.

Os sintomas da injúria pelo frio em frutos de mamoeiro cv 'Golden' foram investigados neste trabalho.Os frutos apresentando de 10% a 15% de coloração amarela na casca, foram classificados na linha de operação de embalagem da Caliman Agrícola S.A (Linhares-ES) e após receberam tratamentos térmicos e químicos. Os frutos foram embalados com filmes plásticos e estocados a 6ºC e 13ºC (85-95% UR) em uma incubadora (BOD) por 30 dias. Em intervalos de tempo definidos, seis frutos foram analisados quanto à firmeza, mudança de cor e aparência. Production and properties of Xylanase from thermophilic Bacillus sp. BABT

An aerobic, thermophilic, xylanolytic bacterium was isolated from local soil. The results of 16S rRNA sequence comparisons indicated that the isolate was closely related to Bacillus caldoxylolyticus and Bacillus sp strain AK1. These organisms exhibited 94% levels of ribossomal DNA sequence homology. Studies on the xylanase characterisation from liquid cultures grown on beechwood xylan revealed that the enzyme retained 100% of activity for 2 hours at temperatures ranging from 30 to 50º C, while at 60, 70 and 100º C, 10%, 11% and 29% of the original activities were lost, respectively. Effect of the medium composition on formation of amylase by Bacillus sp BABT

Studies on the alpha -amylase synthesis was carried out with a moderately thermophilic, facultatively anaerobic Bacillus sp, isolated from soil samples. The cells were cultivated in a complex medium containing soluble starch or maltose as carbon source. The levels of the alpha -amylaseactivity detected in culture supernatants varied greatly with the type of carbon source used. Maltose, soluble starch and citrate stimulated alpha -amylaseformation. Addition of exogenous glucose repressed formation of alpha -amylase, demonstrating that a classical glucose effect was operative in this organism. Isolation, properties and kinetics of growth of a thermophilic Bacillus BJM

A strictly aerobic, thermophilic, spore-forming bacteria was isolated from a soil sample collected in Campos dos Goytacazes City, Rio de Janeiro, Brazil. The cells of this organism were Gram-positive, catalase positive, actively motile, 1.2 µm wide and 5.3 µm long. Growth occurred at pH values ranging from 6.5 to 9.0, and optimum growth occurred at about pH 7.0. The optimum temperature for growth was around 55ºC, and the upper temperature limit for growth was around 70ºC. Yeast extract stimulated growth on glucose and was obligately required at supraoptimal temperatures (65ºC). Effect of the culture conditions on the production of an extracellular protease by thermophilic Bacillus sp and some properties of the enzymatic activity BJM

Protease production by thermophilic Bacillus sp strain SMIA-2 cultivated in liquid cultures containing 1% maltose as a carbon source and supplemented with whey protein (0.1%) and corn steep liquor (0.3%) reached a maximum at 14 h, with levels of 42 U/mg protein. The microorganism was capable of utilizing a wide range of carbon sources, but protease activity varied according the carbon source. Starch and maltose were the best carbon sources in the present study for protease secretion, while lactose and sucrose were less effective. Increasing maltose concentration in the medium until 1%, improved the growth of the organism and the enzyme activity. This study aimed at evaluating the bacteriological effects of the treatment of sheep meat contaminated with total coliforms, coliforms at 45 °C and Salmonella spp. by using irradiation at doses of 3 kGy and 5 kGy. Thirty sheep meat samples were collected from animals located in Rio de Janeiro State, Brazil, and then grouped in three lots including 10 samples: non-irradiated (control); irradiated with 3 kGy; and irradiated with 5 kGy. Exposure to gamma radiation in a 137Cs source-driven irradiating facility was perfomed at the Nuclear Defense Section of the Brazilian Army Technological Center (CTEx) in Rio de Janeiro. Studies on the stability of protease from Bacillus sp. and its compatibility with commercial detergent BJM

Enzymes, and particularly proteases, have become an important and indispensable part of industrial processes such as laundry detergents, pharmaceuticals and food products. Detergents such as Tide®, Ariel® and Biz® contain proteolytic enzymes, most of them produced by members of the genus Bacillus. This paper describes the compatibility of protease produced by the thermophilic Bacillus sp, with commercial laundry detergent. Stability studies indicated that this enzyme retained about 95% and 74% of its maximum activity after 1h at 60ºC in the presence of glycine in combination with MnSO4 and CaCl2, respectively. No inhibitory effect was observed at 1.0-5.0 mM of EDTA. Triton X-100 inhibited the enzyme in all the concentrations tested. The enzyme was unstable...

Production and properties of an extracellular protease from thermophilic Bacillus sp BJM

Protease production by thermophilic Bacillus sp strain SMIA-2 cultivated in liquid cultures containing trisodium citrate reached a maximum in 9h, with levels of 1.93U/mg protein. The microorganism utilized several carbon sources for the production of protease. Starch was the best substrate, followed by trisodium citrate, citric acid and sucrose. Among the various organic and inorganic nitrogen sources, ammonium nitrate was found to be the best. Studies on the protease characterization revealed that the optimum temperature of this enzyme was 60ºC. alpha-amylase (1,4-alpha-D-glucan glucanohydrolase, EC 3.2.1.1) production by thermophilic Bacillus sp strain SMIA-2 cultivated in liquid media containing soluble starch reached a maximum at 48h, with levels of 57U/mL. Studies on the a-amylase characterization revealed that the optimum temperature for activity was 70ºC. The enzyme was stable for 2h at 50ºC, while at 60ºC, 70ºC and 90ºC, 4%, 13% and 38% of the original activities were lost, respectively. The optimum pH of the enzyme was 7.5. Culture conditions for the production of thermostable amylase by Bacillus sp BJM

Studies on the alpha-amylase production were carried out with a bacterial strain isolated from a soil sample. The cells were cultivated in a mineral medium containing soluble starch as sole carbon source. The addition of calcium (10 mM) or peptone (1%) and yeast extract (0.5%) to the mineral medium shortened the lag period and improved the growth and alpha-amylase synthesis. The addition of glucose to the culture diminished greatly the synthesis of alpha-amylase, demonstrating that a classical glucose effect is operative in this organism. alpha-Amylase production by thermophilic Bacillus sp strain SMIA-2 cultivated in liquid cultures containing soluble starch as a carbon source and supplemented with 0.05% whey protein and 0.2% peptone reached a maximum activity at 32 h, with levels of 37 U/mL. Studies on the amylase characterization revealed that the optimum temperature of this enzyme was 90ºC. The enzyme was stable for 1 h at temperatures ranging from 40-50ºC while at 90ºC, 66% of its maximum activity was lost.
