In vitro morphogenesis and cell suspension culture establishment in Piper solmsianum C. DC. (Piperaceae) Acta Botanica

Piper solmsianum is a shrub from Southeast Brazil in which many biologically active compounds were identified. The aim of this work was to establish a cell suspension culture system for this species. With this in mind, petiole and leaf explants obtained from in vitro plantlets were cultured in the presence of different plant growth regulator combinations (IAA, NAA, 2,4-D and BA). Root and indirect shoot adventitious formation, detected by histological analysis, was observed. Polyamine patterns in haploid and diploid tobacco tissues and in vitro cultures BABT

The aim of this work was to determine PAs levels in pith tissues and callus cultures from haploid and diploid tobacco plants, explanted from the apical and basal regions of the stem. These explants were cultured in an RM-64 medium supplied with IAA and kinetin, under light or in the dark, during successive subcultures. PAs levels followed a basipetal decrease in diploid and an increase in haploid, pith tissues. A similar pattern of total PAs (free + conjugated) was observed for the callus of diploid and haploid plants maintained in the light, and for the haploid callus in the dark, whereas the diploid callus in the dark showed a constant increase in total PAs levels until the end of culture. ABSTRACT The use of dark septate fungi (DSE) to promote plant growth can be beneficial to agriculture, and these organisms are important allies in the search for sustainable agriculture practices. This study investigates the contribution of dark septate fungi to the absorption of nutrients by rice plants and their ensuing growth. Four dark septate fungi isolates that were identified by Internal transcribed spacer phylogeny were inoculated in rice seeds (Cv. Piauí). The resulting root colonization was estimated and the kinetic parameters Vmax and Km were calculated from the nitrate contents of the nutrient solution. Proteomics is a branch of molecular biology and biotechnology that has undergone considerable development in the post-genomic era. The aim of the present work was to characterize changes in the protein profile throughout seed development in O. catharinensis, a recalcitrant species, by two-dimensional gel electrophoresis. Protein extraction was undertaken by using a thiourea/urea buffer, followed by a precipitation step with 10% TCA. Comparative analysis during seed development showed that a large number of proteins were exclusively detected in each developmental stage. The cotyledonary stage, which represents the transition phase between embryogenesis and the beginning of metabolism related to maturation, presents the highest number of stage-specific spots. Micropropagação do porta-enxerto de macieira 'Seleção 69' tolerante à podridão do colo (Phytophthora cactorum) Ciência Rural

A utilização de porta-enxertos anões, tolerantes a problemas fitossanitários da cultura da macieira, como a podridão do colo (Phytophthora cactorum), pode aumentar o rendimento e reduzir os custos de produção. Com o presente trabalho, objetivou-se estabelecer um protocolo de micropropagação do porta-enxerto 'Seleção 69', tolerante à podridão do colo. Ápices meristemáticos foram cultivados in vitro em meio de cultura MS, suplementado com 4,4mM de 6-Benzilaminopurina (BAP), 2,5mM de Ácido indolbutírico (AIB), 0,3mM de Ácido giberélico (GA3), 3% de sacarose e 0,6% de ágar.
