Investigating the roles of chemical factors stimulating and inhibiting sperm motility is required to understand the mechanisms of spermatozoa movement. In this study, we described the composition of the seminal fluid (osmotic pressure, pH, and ions) and investigated the roles of these factors and salinity in initiating spermatozoa movement in the Pacific oyster. The acidic pH of the gonad (5.82 ± 0.22) maintained sperm in the quiescent stage and initiation of flagellar movement was triggered by a sudden increase of spermatozoa external pH (pHe) when released in seawater (SW). At pH 6.4, percentage of motile spermatozoa was three times higher when they were activated in SW containing 30 mM NH4Cl, which alkalinizes internal pH (pHi) of spermatozoa, compared... |

The pollack Pollachius pollachius is a commercially important fish, considered as a candidate for aquaculture in France. Rearing attempts are promising (Suquet et al., 1996), and more information is required about its reproduction, to contribute to a better management of breeders for this species. This study was designed to examine the first maturity in pollack and the effects of conditions such as origin and delayed reproduction season, on gonad maturation, reproductive and growth performances of adults. |

Four‐year‐old pollack Pollachius pollachius L., previously fed on dry pellets since their juvenile stage, were divided into three experimental groups two months prior their first spawning season. They were fed a commercial broodstock pellet enriched with 6% added oils, either: (1) capelin oil (control), (2) capelin oil plus arachidonic acid, or (3) DHA rich tuna oil. Spawning performance was determined in each group and egg lipids were analysed. During the vitellogenic period, the estradiol levels in plasma increased with oocyte enlargement, indicating that captivity and pelleted feed did not affect reproductive capacity. Females from each group spawned spontaneously between February and May. Egg production per kg of female was highest in the control... |

P>The present study aimed to establish some basic characteristics of Atlantic bluefin tuna sperm from captive mature males, treated or untreated by gonadotropin releasing hormones agonist (GnRHa). Intratesticular milt was collected from treated and untreated fish (mean weight +/- SD: 122.9 +/- 29.2 kg, n = 21). There was no significant effect of GnRHa treatment on GSI (1.33 +/- 0.70%, n = 21) or on sperm concentration (3.8 +/- 1.3 x 1010 spermatozoa ml-1, n = 21) estimated by optical density at 260 nm. Similarly, the percentage of motile spermatozoa measured at 30 s post activation: activating medium (AM seawater containing 10 mg ml-1 BSA) was not significantly different between control and GnRHa implanted males. On the other hand, a significantly... |

In the Pacific oyster, spermatozoa are characterized by a remarkably long movement phase (i.e., over 24 h) sustained by a capacity to maintain intracellular ATP level. To gain information on oxidative phosphorylation (OXPHOS) functionality during the motility phase of Pacific oyster spermatozoa, we studied 1) changes in spermatozoal mitochondrial activity, that is, mitochondrial membrane potential (MMP), and intracellular ATP content in relation to motion parameters and 2) the involvement of OXPHOS for spermatozoal movement using carbonyl cyanide m-chlorophenyl hydrazone (CCCP). The percentage of motile spermatozoa decreased over a 24 h movement period. MMP increased steadily during the first 9 h of the movement phase and was subsequently maintained at a... |

The king scallop Pecten maximus is a high valuable species of great interest in Europe for both fishery and aquaculture. Notably, there has been an increased investment to produce seed for enhancement programmes of wild scallop populations. However, hatchery production is a relatively new industry and it is still underdeveloped. Major hurdles are spawning control and gamete quality. In the present study, a total of 14 scallops were sampled in the bay of Brest (Brittany, France) to compare transcriptomic profiles of mature oocytes collected by spawning induction or by stripping. To reach such a goal, a microarray analysis was performed by using a custom 8x60K oligonucleotide microarray representing 45,488 unique scallop contigs. First we identified genes... |

The biology of gametes and larvae in brooding mollusc species remains little explored. The aim of this study is to present the results obtained in 2016, on biological characteristics of gametes and larvae of European flat oyster. Measurements were carried out on 50 oysters, collected by divers in the Rade de Brest (Finistère, France), during the spawning period of this species. Gonad pH is acidic (6.58±0.54). After the dissociation of spermatozeugmata, an acellular structure to which the spermatozoa are clustered, the percentage of motile spermatozoa ranged from 20 to 34% and the Velocity of the Average Path from 70 to 85µm/s. Spermatozeugmata were shed after serotonin injection. Oocytes (mean diameter: 129.4±19.1µm) were rarely observed (2... |

In order to improve oyster culture, many research programs try and propose new methods aiming at improving genetically the product performances and quality (selection, polyploidisation). As for terrestrial animal production, the continuity of the different works as well as the spreading of the genetical progress would be made easier by the development of conservation techniques of the selected genotypes. Cryopreservation, which is often used in selection programs of terrestrial animals or fish farming species, is not always available for bivalves. This technique would enable the conservation of the gametes of individuals selected for their high growth potential, their resistance to diseases and/or their quality for a smaller cost and less risks than the... |

In production operations the application of daily examination of each female of broodstock should be a determinant progress in hatchery management in terms of fry output. The eggs expelled shortly after ovulation by means of a systematic fivefold a week stripping attempt of females, presented a high viability rate (72%) compared to that of eggs collected twice a week (46%) whereas the treatments did not have any effect on female fecundity. Increase of the stripping rhythm brought the hatching rates and broodstock fertilities respectively from 11% to 35% and from 20,000 to 65,000 larvae/kg of spawning female. The enhancement of egg quality and of the resulting larva production is discussed. |

Pacific oyster Crassostrea gigas is one major species for aquaculture, and the development of breeding programs and the need for preservation of wild stock genetic resources prompted the need for larvae cryopreservation. The objective of the present study was to choose the most reliable protocol from several existing publications, to test its biological and practical limitations, and to adapt it to hatchery conditions. The selected protocol was characterized by a very slow freezing rate without seeding, and by the use of ethylene glycol and sucrose as cryoprotectant. The best survivals after thawing and rearing up to 48 h post fertilization (hpf) were obtained with larvae that were frozen at late trochophore (20 hpf) and early-D (24 hpf) stages. Increasing... |

This paper presents some features of turbot sperm and compares them with data reported in the literature on other freshwater and marine fish. In captivity, turbot spermiation period lasts 6 months. When compared to other fish species, turbot males are characterized by low values of maximum gonadosomatic index (0.6-0.8%) and are poor sperm producers in terms of volume (0.2-2.2 ml), concentration (0.7-11.0 x 10(9) spermatozoa/ml) and total sperm number released per stripping (0.2-12.0 x 10(9) spermatozoa). Compared to most freshwater species, the duration of cell movement is long (1-17 min). The spermatozoon of turbot can be considered to be of a primitive type. A very high protein content (8.8 mg/ml) is found in the seminal fluid of turbot. Increasing... |

A basic cryopreservation protocol for great scallop sperm was recently published, with which thawed sperm survival remained limited to 25% of control values (Suquet et al. 2014). We designed here a set of five experiments to test ways of improving thawed sperm survival and storage by studying the effects of (1) cryoprotectant concentration; (2) sugar incorporation in the extender; (3) egg yolk addition to the extender; (4) sperm dilution rate; and (5) storage capacity of thawed sperm at 4 degrees C. A sixth experiment was then performed to compare rearing performances of larvae produced using thawed sperm from the optimal protocol defined in the present study with those produced using fresh sperm. During the first experiment, a significantly higher... |

Controlled reproduction of the European flat oyster requires the development of tools adapted to this species, including a practical anaesthesia and gonad sampling protocol to facilitate sex determination and the verification of gametogenesis. Three replicate groups of 10 oysters (mean weight +/- SD: 29.9 +/- 8.5 g) were anaesthetised in 5 L containers using magnesium chloride designed either for laboratory (Flucka (R)) or agricultural (Dead Sea Work (R): DSW) use, at two concentrations (50 or 72 g L-1). No significant differences were observed in the percentages of oysters anaesthetised or subsequent oyster mortality with the different anaesthetics or concentrations, but increasing water temperature from 14.9 to 18.8 degrees C significantly increased the... |

P>The objective of this study was to complement published hake (Merlucius merlucius) sperm motility characteristics by obtention of data more specifically dependent on flagella: beat frequency, flagellar wave amplitude, wave length, number of waves along the length of the flagellum and curvature of the wave. The changes of these parameters are described in relation to the time period elapsed since activation are described using high resolution video images of flagella while quantifying the progression during the swimming period from the activity initiation to full cessation of movement several minutes later. The main characteristic of the swimming period of hake spermatozoa shows a progressive decrease of forward displacement resulting from a cumulative... |

Sperm characteristics of scallops have not been well described in the scientific literature. The effects of sperm release technique (thermal shock versus serotonin injection), of sperm collection technique (testis sampling versus serotonin injection), of sperm sampling location along the genital tract, of in vitro sperm maturation, and of time post activation on scallop sperm characteristics were assessed in the present work. Whatever sperm release technique used, no significant differences were observed regarding the percentage of motile spermatozoa and the velocity of the average path (YAP). Compared to testicular sperm, a higher percentage of motile spermatozoa, YAP and intracellular adenosine triphosphate (ATP) content were observed for sperm shed... |
