Quality of cocoa (Theobroma cacao L.) DNA from foliar tissue at different stages of development Acta Agron. (Palmira)
Henao Ramírez,Ana María; Salazar Duque,Héctor Jaime; Urrea Trujillo,Aura Inés.
Abstract Theobroma cacao L. and its products are consumed worldwide. Those products are of great research interest due to antioxidant properties of some of their polyphenolic constituents. The amount of these polyphenols and polysaccharides has shown that can interfere with the high quality and quantity of nucleic acids for molecular research. Therefore, cocoa DNA extraction protocols can require a large amount of plant material and optimization time according with plant material source. The aim of this study was to evaluate the quality and quantity of DNA isolated from field plant leaves at different developmental stages from TSH565 genotype using different DNA extraction protocols. In addition, DNA extraction protocol was evaluated for small amount of...

Ex situ potential conservation of ipecacuanha ( Psychotria ipecacuanha (Brot.) Stokes . ) , a critically endangered medicinal plant species Acta Agron. (Palmira)
Urrea Trujillo,Aura Inés; Botero Giraldo,Catalina; Naranjo Gómez,Esther Julia.
Abstract Psychotria ipecacuanha is a medicinal plant endangered in Colombia, due to the indiscriminate exploitation suffered several decades ago for its therapeutic importance. Its great medicinal value is due to emetine production and its derivatives, mainly in the roots. Given these concerns, the aim of this research was to evaluate different parameters for in vitro conservation of P. ipecacuanha by a limited plant growth, using as in vitro explants plant nodes. Plant growth response to 18, 23 and 25°C with and without addition to MS basal culture medium 15 and 30 gL-1 of mannitol osmotic regulator was evaluated. Registration of plant survival data (%), plant height (cm), plant development of axillary shoots, number of leaves, callus formation and vigour...

In vitro propagation of avocado (Persea americana Mill.) cv. Hass through morphogenesis Acta Agron. (Palmira)
Restrepo Osorio,Catalina; Gómez Velásquez,Felipe Andrés; Gil Correal,Alejandro; Torres Bonilla,Javier Mauricio; Urrea Trujillo,Aura Inés.
Abstract Given the importance and potential development of avocado (Persea americana Mill.) cultivation in Colombia and its problems for an efficient field cultivation, which includes the genetic variability of propagated material and adverse phytosanitary conditions, the aim of this study was to develop a method for in vitro clonal propagation for avocado cv. Hass. In vitro introduction of apical explants and nodal segments was carried out from grafted parent plants. Etiolation pretreatment effect on the number of shoots formed and in vitro phenolization was evaluated in grafted parent plants and in explants under in vitro conditions, subject to photoperiod (12/12 hours) and darkness. Darkness conditions reduced significantly plant phenolization, and...
