Natural abundance of 13C in tropical grasses from the INPA, Instituto Nacional de Pesquisas da Amazônia, herbarium Rev. Bras. Bot.
MEDINA,ERNESTO; MARTINELLI,LUIZ ANTONIO; BARBOSA,EDELCILIO; VICTORIA,REYNALDO LUIZ.
The stable carbon isotopic composition of 165 grass species was determined with the objective of verifying their photosynthetic pathway (C3 and C4). The samples, taken from the INPA herbarium, were mainly collected in the North of Brazil. Approximately 60% of the species proved to be of the C4 type, with <FONT FACE="Symbol">d</font> 13C values ranging from -13.6 to -9.5‰, while the remainder 40% belonged to the C3 type, with values ranging from -34.7 to -23.4‰. This relatively high proportion of C3 species is probably due to the high relative humidity of the sites where the species were collected.

BRS 177,...

Nitrate levels and stages of growth in hypernodulating mutants of Lupinus albus. II. Enzymatic activity and transport of N in the xylem sap Rev. Microbiol.
Burity,Hélio Almeida; Chamber-Perez,Manuel; Lyra,Maria do Carmo Catanho Pereira de; Figueiredo,Márcia do Vale Barreto.
The enzymatic study and transport of N in the xylem sap was carried out with a view to observing the influence of different nitrate levels and growth stages of the plant in chemically treated mutants of Lupinus albus. Several stresses induce a reduction in plant growth, resulting in the accumulation of free amino acids, amides or ureides, not only in the shoot, but also in the roots and nodules. Although enzyme activity is decisive in avoiding products that inhibit nitrogenase by ammonium, little is known about the mechanism by which the xylem carries these products. However, this process may be the key to the function of avoiding the accumulation of amino acids in the cells of infected nodules. The behaviour of the enzymes nitrate reductase (NR),...

Bacteria isolated from a sugarcane agroecosystem: their potential production of polyhydroxyalcanoates and resistance to antibiotics Rev. Microbiol.
Lima,Teresa Cristina S. de; Grisi,Breno M.; Bonato,M. Christina M..
In this investigation, a sugarcane agroecosystem at a coastal tableland, in the northeast of Brazil, was screened to obtain bacteria strains able to synthesize poly-<FONT FACE="Symbol">b</font>-hydroxyalkanoates (PHA), using sucrose as the main carbon source. The potential to synthesize PHA was tested qualitatively by Sudan Black staining of colonies growing in different carbon sources: sucrose, glucose, fructose, propionate and cellulose. In a typical sugarcane crop management system, the plantation is burned before harvesting and vinasse, a byproduct of alcohol production, is used in a fertirrigation system causing, probably, selective pressures on the microbiota of natural environments. Eightytwo bacteria strains, belonging to 16 different...

Optimized Fast-FISH with a-satellite probes: acceleration by microwave activation BJMBR
It has been shown for several DNA probes that the recently introduced Fast-FISH (fluorescence in situ hybridization) technique is well suited for quantitative microscopy. For highly repetitive DNA probes the hybridization (renaturation) time and the number of subsequent washing steps were reduced considerably by omitting denaturing chemical agents (e.g., formamide). The appropriate hybridization temperature and time allow a clear discrimination between major and minor binding sites by quantitative fluorescence microscopy. The well-defined physical conditions for hybridization permit automatization of the procedure, e.g., by a programmable thermal cycler. Here, we present optimized conditions for a commercially available X-specific <FONT...

The role of IL-12 in experimental Trypanosoma cruzi infection BJMBR
Silva,J.S.; Aliberti,J.C.S.; Martins,G.A.; Souza,M.A.; Souto,J.T.; Pádua,M.A..
Host resistance to Trypanosoma cruzi infection is dependent on both natural and acquired immune responses. During the early acute phase of infection in mice, natural killer (NK) cell-derived IFN-<FONT FACE="Symbol">g</font> is involved in controlling intracellular parasite replication, mainly through the induction of nitric oxide biosynthesis by activated macrophages. We have shown that IL-12, a powerful inducer of IFN-<FONT FACE="Symbol">g</font> production by NK cells, is synthesized soon after trypomastigote-macrophage interaction. The role of IL-12 in the control of T. cruzi infection in vivo was determined by treating infected mice with anti-IL-12 monoclonal antibody (mAb) and analyzing both parasitemia and mortality during the...

An apparently paradoxical role for IFN-<FONT FACE="Symbol">g</font> in human Chagas' disease was observed when studying the pattern of cytokine production by peripheral blood mononuclear cells (PBMC) obtained from two groups of chagasic patients after specific stimulation with Trypanosoma cruzi-derived antigens. The groups studied were 1) patients treated with benznidazole during the acute phase of Trypanosoma cruzi infection and 2) chronically infected untreated patients. In the treated group, higher levels of IFN-<FONT FACE="Symbol">g</font> were produced by PBMC from individuals cured after treatment when compared to non-cured patients. The role of different cytokines in the peripheral blood mononuclear cell (PBMC) proliferative response and in in vitro granuloma formation was evaluated in a cross-sectional study with patients with the different clinical forms and phases of Schistosoma mansoni infection, as well as a group of individuals "naturally" resistant to infection named normal endemic (NE). The blockage of IL-4 and IL-5 using anti-IL-4 and anti-IL-5 antibodies significantly reduced the PBMC proliferative response to soluble egg (SEA) and adult worm (SWAP) antigens in acute (ACT), chronic intestinal (INT) and hepatosplenic (HS) patients. Similar results were obtained in the in vitro granuloma formation. Blockage of IL-10 had no significant effect on either assay using PBMC from ACT...

Immunologic patterns associated with cure in human American cutaneous leishmaniasis BJMBR
Coutinho,S.G.; Da-Cruz,A.M.; Bertho,A.L.; Santiago,M.A.; De-Luca,P..
Patients with American cutaneous leishmaniasis were studied before therapy (active lesion) and at the end of therapy (cured patients). Assays of lymphocyte proliferative responses of peripheral blood mononuclear cells induced in vitro by Leishmania braziliensis promastigote antigens (Lb) were performed. Antigen-stimulated cells were harvested for CD4 and CD8 phenotype analysis and the levels of gamma interferon (IFN-<FONT FACE="Symbol">g</font>) and interleukin 4 (IL-4) produced were also determined in the culture supernatants. Two different patterns of Lb-induced T cell responses were observed: a) predominance of responding CD4+ cells and mixed type 1 and type 2 cytokine production (IFN-<FONT FACE="Symbol">g</font> and IL-4) during...

Eosinophil-active chemokines: assessment of in vivo activity BJMBR
Teixeira,M.M..
The selective recruitment of eosinophils in tissue is a striking feature of allergic diseases. Recently, a family of chemoattractant molecules, namely chemokines, has been described which potently activates eosinophil function in vitro. We have developed a murine model of eosinophil recruitment to compare the relative potency and efficacy of chemokines in vivo. Of the chemokines tested, only eotaxin and MIP-1<FONT FACE="Symbol">a</font> induced significant accumulation of eosinophils in vivo, but eotaxin was more effective than MIP-1<FONT FACE="Symbol">a</font>. Chemokines, especially eotaxin acting via the CCR-3 receptor, may have a fundamental role in determining selective eosinophil recruitment in vivo

Gastroduodenal resistance and neural mechanisms involved in saline flow decrease elicited by acute blood volume expansion in anesthetized rats BJMBR
Graça,J.R.V.; Gondim,F. de-A.A.; Cavalcante,D.I.M.; Xavier-Neto,J.; Messias,E.L.M.; Rego,M.C.V.; Marques,J.A.P.; Santos,A.A.; Rola,F.H..
We have previously demonstrated that blood volume (BV) expansion decreases saline flow through the gastroduodenal (GD) segment in anesthetized rats (Xavier-Neto J, dos Santos AA &amp; Rola FH (1990) Gut, 31: 1006-1010). The present study attempts to identify the site(s) of resistance and neural mechanisms involved in this phenomenon. Male Wistar rats (N = 97, 200-300 g) were surgically manipulated to create four gut circuits: GD, gastric, pyloric and duodenal. These circuits were perfused under barostatically controlled pressure (4 cmH2O). Steady-state changes in flow were taken to reflect modifications in circuit resistances during three periods of time: normovolemic control (20 min), expansion (10-15 min), and expanded (30 min). Perfusion flow rates...

Cytokine profiles during experimental Chagas' disease BJMBR
Morato,M.J.F.; Colley,D.G.; Powell,M.R..
People infected with Trypanosoma cruzi remain so for life, yet only 30-40% of these individuals develop characteristic chagasic cardiomyopathies. Similarly, when infected with the Brazilian strain of T. cruzi, DBA/2 mice develop severe cardiac damage while B10.D2 mice do not. To better understand the immunological parameters that may be involved in the disease process, we have used this murine model (DBA/2 vs B10.D2) and compared the changes in cytokine production during the course of infection with T. cruzi. Concanavalin A (Con A) stimulation of spleen cells harvested during the acute phase (day 30) resulted in similarly high levels of IFN-<FONT FACE="Symbol">g</font> in both mouse strains. Interaction between NO and oxytocin: Influence on LHRH release BJMBR
Nitric oxide synthase (NOS)-containing neurons have been localized in various parts of the CNS. These neurons occur in the hypothalamus, mostly in the paraventricular and supraoptic nuclei and their axons project to the neural lobe of the pituitary gland. We have found that nitric oxide (NO) controls luteinizing hormone-releasing hormone (LHRH) release from the hypothalamus acting as a signal transducer in norepinephrine (NE)-induced LHRH release. LHRH not only releases LH from the pituitary but also induces sexual behavior. On the other hand, it is known that oxytocin also stimulates mating behavior and there is some evidence that oxytocin can increase NE release. Therefore, it occurred to us that oxytocin may also stimulate LHRH release via NE and NO. To...

Multifactorial control of water and saline intake: role of a2-adrenoceptors BJMBR
Water and saline intake is controlled by several mechanisms activated during dehydration. Some mechanisms, such as the production of angiotensin II and unloading of cardiovascular receptors, activate both behaviors, while others, such as the increase in blood osmolality or sodium concentration, activate water, but inhibit saline intake. Aldosterone probably activates only saline intake. Clonidine, an <FONT FACE="Symbol">a</font>2-adrenergic agonist, inhibits water and saline intake induced by these mechanisms. One model to describe the interactions between these multiple mechanisms is a wire-block diagram, where the brain circuit that controls each intake is represented by a summing point of its respective inhibiting and activating factors.
