RESPONSES OF COWPEA CULTIVARS TO SALINE STRESS 196
Romanoski, Vanessa; de Moura Souza Filho, Paulo Roberto; Badaró Pimentel, Adérico Júnior.
The cowpea [Vigna unguiculata (L.) Walp] is widely cultivated in Northeast Brazil and stands out for its economic and social importance. Part of the cultivation area is affected by salt stress, which affects the performance of the crop. The effects of salinity can be mitigated with the development of tolerant cultivars. The objective of this work was to evaluate the behavior of cowpea cultivars under different levels of saline stress in the germination and emergence phases of the crop. Two trials evaluating the germination and emergence phases were conducted, under controlled conditions, in a factorial scheme with 11 cultivars and five solution of NaCl with concentrations ranging from 0 to 120 mM. There was a marked difference in the response of the...

Sustainable production of bioactive alkaloids in Psychotria L. of southern Brazil: propagation and elicitation strategies Acta Botanica
Magedans,Yve Verônica da Silva; Rodrigues-Corrêa,Kelly Cristine da Silva; Costa,Cibele Tesser da; Matsuura,Hélio Nitta; Fett-Neto,Arthur Germano.
ABSTRACT Psychotria is the largest genus in Rubiaceae. South American species of the genus are promising sources of natural products, mostly due to bioactive monoterpene indole alkaloids they accumulate. These alkaloids can have analgesic, antimutagenic, and antioxidant activities in different experimental models, among other pharmacological properties of interest. Propagation of genotypes with relevant pharmaceutical interest is important for obtaining natural products in a sustainable and standardized fashion. Besides the clonal propagation of elite individuals, the alkaloid content of Psychotria spp. can also be increased by applying moderate stressors or stress-signaling molecules. This review explores advances in research on methods for plant...

Hydrogen sulfide: a new endogenous player in an old mechanism of plant tolerance to high salinity Acta Botanica
da-Silva,Cristiane J.; Modolo,Luzia V..
ABSTRACT High salinity affects plants due to stimulation of osmotic stress. Cell signaling triggered by nitric oxide (NO) and hydrogen sulfide (H2S) activates a cascade of biochemical events that culminate in plant tolerance to abiotic and biotic stresses. For instance, the NO/H2S-stimulated biochemical events that occur in plants during response to high salinity include the control of reactive oxygen species, activation of antioxidant system, accumulation of osmoprotectants in cytosol, induction of K+ uptake and Na+ cell extrusion or its vacuolar compartmentation among others. This review is a compilation of what we have learned in the last 10 years about NO participation during cell signaling in response to high salinity as well as the role of H2S, a new...

Menezes, Cicero Beserra de; Lima, Geisiane Matos; Marucci, Rosangela Cristina; Bernardino, Karine Costa; Santos, Crislene Vieira dos; Julio, Marcos Paulo Mingote; Schaffert, Robert Eugene.
Sorghum (Sorghum bicolor L. Moench) is one of the most important cereal crops in the world. In Brazil, the acreage of grain sorghum during off-season is quite expansive. Most of this area is the Cerrado, a Brazilian biome that is similar to a Savannah and is characterized by high acidity and soluble aluminum at toxic levels for plants. The aluminum acts as a limiting factor in achieving high yields. The purpose of this work was to phenotype sorghum hybrids for aluminum tolerance. Eighteen hybrids were evaluated in a nutrient solution containing {0} or {27} µM Al3+. The work was carried out in a growth chamber at the Embrapa Maize and Sorghum, from April 4 to May 30, 2014. The lines ATF 13B (susceptible) and ATF 14B (tolerant) were used as check cultivars....

Expression of genes related to soil flooding tolerance in soybeans Agronomy
Casarotto, Gabriele; Kaspary, Tiago Edu; Cutti, Luan; Thomas, André Luis; Barbosa Neto, Jose Fernandes.
The flooded environment brings about injuries to soybeans that vary depending on the adaptation ability of the genotype. Oxygen deprivation promotes the induction of the expression of genes related to glycolysis and fermentation pathways to maintain energy metabolism and, in addition to reducing-power consuming processes, act in the formation of adaptive structures and the maintenance of the redox status of the plant. The aim of this work was to evaluate the relative expression of genes related to soil flooding response in two contrasting soybean cultivars. Soybean plants of the sensitive (BRS 154) and tolerant (I27) cultivars at the V1 development stage were submitted to the flooding and control conditions (without flooding) for 0, 24, 48, and 96 hours....

Physiological analysis and gene expression analysis of ZmDBP3, ZmALDH9, ZmAN13, and ZmDREB2A in maize lines Agronomy
Marques, Thaís Lima; Pinho, Renzo Garcia Von; Pinho, Édila Vilela de Resende Von; Paniago, Bruno da Costa; Freitas, Natália Chagas; Santos, Heloisa Oliveira dos.
The objective of this study was to evaluate gene expression related to water deficit tolerance in maize lines. For this, lines previously classified as tolerant (91-T and 32-T) and non-tolerant (24-NT and 57-NT) to water deficit were used. The seeds of the four lines were evaluated for emergence and emergence speed index, and the seedlings were evaluated for root and shoot length under two conditions of water availability (70 and 10% substrate water retention capacity). In transcript analysis, the expression of several genes associated with water deficit tolerance, ZmDBP3, ZmALDH9, ZmAN13, and ZmDREB2A, was evaluated by qRT-PCR for the 91-T and 57-NT lines. It can be concluded that soil water deficiency did not reduce root development. However, the shoot...

High temperatures on root growth and lignification of transgenic glyphosate-resistant soybean Agrociencia
Zonetti,P. da Costa; Suzuki,L. Sayuri; Bonini,E. Aparecida; Ferrarese,M. L. Lucio; Ferrarese-Filho,Osvaldo.
Glyphosate resistant transgenic soybean [Glycine max (L.) Mettill] expresses a glyphosate insensitive EPSPS (5-enolpyruvylshikimate-3-phosphate synthase). This enzyme is involved in important secondary metabolism pathways, including lignin biosynthesis. Thus, differences in lignin content and growth between susceptible (OC14) and glyphosate-resistant soybean may be observed. The objective of this study was to evaluate differences between growth and lignin content of roots in transgenic and OC14 soybean cultivated at high temperatures. Seeds from the OC14 soybean and its transgenic cultivar, CD213RR, were germinated at 25, 27.5, 30 and 32.5 °C. After 3 d, seedlings were cultivated in Hoagland half-strength nutrient solution during 12 h photoperiods and at...

Effects of hormonal priming on seed germination of pigeon pea under cadmium stress Anais da ABC (AABC)
SNEIDERIS,LARISSA C.; GAVASSI,MARINA A.; CAMPOS,MARCELO L.; D'AMICO-DAMIÃO,VICTOR; CARVALHO,ROGÉRIO F..
In this work we investigated whether priming with auxin, cytokinin, gibberellin, abscisic acid and ethylene, alters the physiological responses of seeds of pigeon pea germinated under water and cadmium stress. Seeds treated with water or non-treated seeds were used as control. Although compared to non-treated seeds we found that the hormone treatments improve the germination of pigeon pea under cadmium stress, however, these treatments did not differ from water. However, we also observed a trend of tolerance to the effects of cadmium in the presence of ethylene, suggesting that the use of this hormone may be an efficient method to overcome seed germination under metal stress.

Salinity induced anatomical and morphological changes in Chloris gayana Kunth roots Biocell
Céccoli,Gabriel; Ramos,Julio C.; Ortega,Leandro I.; Acosta,Juan M.; Perreta,Mariel G..
Chloris gayana Kunth is a grass species valuable as forage which was introduced into Argentina to be used as pasture in saline soils of subtropical and warm-temperate zones, given its good adaptability to drought, salinity and mild freezing. However, its tolerance varies according to the cultivar. In tetraploid cultivars, important reductions in yield have been observed. Here, a study of the variations produced on the root and stem system by salinity at different NaCl concentrations (0, 150 y 250 mM) was performed in the Boma cultivar, with the aim of determining the anatomical and morphological alterations produced by the salt excess. Plants cultivated with the highest level of salinity showed, in the whole, significant differences in the measured...

The role of phytochromes in cadmium stress responses in tomato Bragantia
Gaion,Lucas Aparecido; Lorevice,Paulo Guilherme; Monteiro,Carolina Cristina; Gavassi,Marina Alves; D’Amico-Damião,Victor; Gratão,Priscila Lupino; Gasparino,Eduardo Custódio; Carvalho,Rogério Falleiros.
ABSTRACT It is well known that phytochromes mediate a wide range of photomorphogenic processes in plants. In addition, many studies have demonstrated the involvement of phytochromes as part of abiotic stress signaling responses. However, little is known about cadmium (Cd) stress regulation by phytochromes. Thus, in this study, we used the phyA (far red-insensitive; fri), phyB1 (temporary redinsensitive; tri) and phyB2 (phyB2) tomato (Solanum lycopersicum L.) mutants to investigate the roles of these three phytochromes on Cd stress responses. The plants were grown over a 21-d period in the presence of Cd. We evaluated plant growth, Cd and chlorophyll content and anatomical changes in the leaves. The results indicated that all genotypes were affected by Cd...

Rice genotypes for drought tolerance: morphological and transcriptional evaluation of auxin-related genes Bragantia
Madabula,Frederico Pedro; Santos,Railson Schreinert dos; Machado,Nata; Pegoraro,Camila; Kruger,Mariana Madruga; Maia,Luciano Carlos da; Sousa,Rogerio Oliveira de; Oliveira,Antonio Costa de.
ABSTRACT Drought stress affects crop quality and productivity. The challenge of increasing food availability for a growing worldwide demand relies on the development of tolerant cultivars that will need to be adapted to arid and semi-arid areas. In order to help the understanding of rice response to stress, the phenotypic response of 6 Brazilian rice cultivars and 2 different crosses between them were characterized under drought conditions. Since gene regulation is an important part of root morphological responses to stressful conditions, 4 genes related to auxin response and root modifications (OsGNOM1 CRL4, OsIAA1, OsCAND1 and OsRAA1) were evaluated. The expression of these genes was analyzed in stressed rice using public available microarray data and...

Silva,Daiana Alves da; Esteves,Jose Antonio de Fatima; Gonçalves,João Guilherme Ribeiro; Azevedo,Cleber Vinícius Giaretta; Ribeiro,Tamires; Chiorato,Alisson Fernando; Carbonell,Sérgio Augusto Morais.
ABSTRACT Common bean is one of the most important legumes in Latin America, mostly grown in soils with low phosphorus (P) availability. Thus, this study aimed to evaluate the responses of 20 bean genotypes to P deficiency. The experiment was a completely randomized design in a 2 × 20 factorial arrangement; the first factor consisted of P levels and the second factor, of 20 bean genotypes, with six replications. The substrate was a Red Eutrophic Oxisol with low P content. For application of the P treatments, it was applied simple superphosphate, consisting of two levels: restrictive and control, with the application of 45 and 90 kg∙ha–1 of P2O5, respectively. At 28 days, we observed the first symptoms of nutrient deficiency, with the decrease in the...

Index selection of tropical maize genotypes for nitrogen use efficiency Bragantia
Granato,Ítalo Stefanine Correia; Bermudez,Felipe Pereira; Reis,Gabriel Gonçalves dos; Dovale,Julio César; Miranda,Glauco Vieira; Fritsche-Neto,Roberto.
Nitrogen (N) limitation in maize crops is related to the fact that the efficiency of nitrogen fertilization in maize does not exceed 50%, primarily due to volatilization, denitrification and soil leaching. Therefore, the development of new nitrogen use efficient (NUE) cultivars is necessary. The aim of the present study was to develop indices for the accurate selection of NUE maize genotypes for use in conditions of both high and low N availability. The experiment was conducted in a greenhouse (20º45'14"S; 42º52'53"W) at the Federal University of Viçosa during October 2010. A total of 39 experimental hybrid combinations and 14 maize lines differing in NUE were evaluated under two N availability conditions. We determined the relative importance of the...

Phytoene Synthase Gene ( PSY ) from Sweet Potato ( Ipomoea batatas Lam . ) Enhances Tolerance to Abiotic Stress BABT
Shao,Huanhuan; Yong,Bin; Xu,Pan; Zheng,Haiyan; Liao,Ruoxing; Wang,Xiaoyan; Li,Xinyu; Zhang,Li; Shen,Jiabin.
ABSTRACT Phytoene synthase (PSY) is the rate-limiting enzyme for carotenoid biosynthesis. To date, several studies focused on PSY genes in the context of abiotic stress responses. In this study, two phytoene synthase encoding genes, IbPSY1 and IbPSY2, were identified from a published transcriptome and bioinformatic analysis showed that they shared conserved domains with phytoene synthases from other plants. The IbPSY1 gene was cloned and carefully characterized. Digital gene expression profiling (DGE) showed that the highest transcription level of IbPSY1 was in young leaves, and the lowest level was in stems. In vivo expression levels of IbPSY1 under abiotic stress were observed to be highest in stems at day 11. Over-expression of IbPSY1 in Escherichia...

Methodology adjustments for organic acid tolerance studies in oat under hydroponic systems BABT
Kopp,Mauricio Marini; Luz,Viviane Kopp da; Souza,Velci Queiróz de; Coimbra,Jefferson Luis Meirelles; Sousa,Rogério Oliveira de; Carvalho,Fernando Irajá Félix de; Oliveira,Antonio Costa de.
The occurrence of anaerobic conditions in hydromorphic soils favors the development of anaerobic microorganisms that produce phytotoxic substances representing primarily by organic acids. The selection of promising oat (Avena sativa L.) genotypes for use in those situations requires field evaluations that can be cumbersome, making hydroponics a viable alternative. The objective of this work was to adjust a methodology to use in studies of tolerance to organic acids in oat under hydroponic systems. For such goal, the best germination system was determined in order to reduce the seedling initial establishment effects under hydroponics, the ideal concentration for screening genotypes and the best variable for stress evaluation. It was found that the most...
