Characterization of the acetohydroxyacid synthase multigene family in the tetraploide plant Chenopodium quinoa Electron. J. Biotechnol.
Mestanza,Camilo; Riegel,Ricardo; Silva,Herman; Vásquez,Santiago C.
Background Currently, the technology called Clearfield® is used in the development of crops resistant to herbicides that inhibit the enzyme acetohydroxy acid synthase (AHAS, EC 2.2.1.6). AHAS is the first enzyme of the biosynthetic pathway that produces the branched-chain of the essential amino acids valine, leucine, and isoleucine. Therefore, multiple copies of the AHAS gene might be of interest for breeding programs targeting herbicide resistance. In this work, the characterization of the AHAS gene was accomplished for the Chenopodium quinoa Regalona-Baer cultivar. Cloning, sequencing, and Southern blotting were conducted to determine the number of gene copies. Results The presence of multiple copies of the AHAS gene as has been shown previously in...

Resistance of Raphanus raphanistrum to the herbicide metsulfuron-methyl Planta Daninha
Costa,L.O.; Rizzardi,M.A..
The ALS-inhibiting herbicides, especially metsulfuron-methyl, are widely used for weed control, mainly wheat and barley in southern Brazil. Raphanus raphanistrum is a major weed of winter crops. However, in recent years, R.raphanistrum, after being treated with metsulfuron, has shown no symptoms of toxicity, possibly due to herbicide resistance. Aiming to evaluate the existence of R.raphanistrum biotypes resistant to metsulfuron, an experiment was conducted in a greenhouse, in a completely randomized design with four replications. The plots consisted of pots with six plants. The treatments consisted of the interaction of resistant R. raphanistrum (biotype R) and susceptible R. raphanistrum (biotypes S) with ten doses of the herbicide (0.0; 0.6; 1.2; 2.4;...

Globe fringerush (Fimbristylis miliacea) cross resistance to als-inhibitor herbicides under field conditions in irrigated rice in the south of Brazil Planta Daninha
Schaedler,C.E.; Noldin,J.A.; Eberhardt,D.S.; Agostinetto,D.; Burgos,N.R..
ALS-inhibiting herbicides usually provide adequate weed control in irrigated rice fields. After consecutive years of use, the Cyperaceae species, globe fringerush (Fimbristylis miliacea) began to show resistance to ALS (acetolactate synthase) inhibitors. Globe fringerush is one of the most problematic herbicide-resistant weeds in irrigated rice in the state of Santa Catarina in the South of Brazil. The objective of this research was to examine cross resistance of globe fringerush to ALS inhibitors, under field conditions. Two experiments were conducted in a rice field naturally infested with ALS-resistant globe fringerush in Santa Catarina, in the 2008/09 and 2009/10 cropping seasons. The experimental units were arranged in randomized complete block...

Resistence of Radish Biotypes to Iodosulfuron and Alternative Control Planta Daninha
CECHIN,J.; VARGAS,L.; AGOSTINETTO,D.; ZIMMER,V.; PERTILE,M.; GARCIA,J.R..
The repetitive use of iodosulfuron for the control of weeds in winter cereals in the south of Brazil has favored the emergence of resistant Raphanus sativus biotypes. The objective of this study was to evaluate: the response of Raphanus sativus biotypes susceptible and resistant to different dosages of iodosulfuron; the control of biotypes with alternative registered herbicides for the control of the species in crops of wheat, corn and soybean; and the existence of cross-resistance of the biotypes. Thus, four experiments were done in a greenhouse, with a completely randomized design and four replicates. The experimental units were composed of vases with a volumetric capacity of 0.75 L filled with substrate, containing a plant each. For the dose-response...

Resistance detection of blackjack to ALS inhibitors by in vitro plant growth method Planta Daninha
Tessaro,Daniela; Benneman,Daiane; T. Perboni,Lais; R. Garcia,Jéssica; M. Teixeira,Marlon; Agostinetto,Dirceu.
Abstract Background: The resistance of weeds to herbicides is a problem that has been increasingly studied because of its great importance in agriculture and rapid growth of this phenomenon worldwide. The quick detection of resistant plants is extremely important for resistance management. Weed resistance to herbicides can be detected through field, greenhouse and laboratory tests. A fast way of detecting resistance is using tissue culture method, where is possible to use resistant plant clones and obtain results faster than whole-plant method in greenhouse. Objective: The aims of the research were to detect the resistance of blackjack to herbicides inhibitors of the acetolactate synthase (ALS), addition in vitro cultivation; determine the herbicide...

Resistance Mapping of the Genus Cyperus in Rio Grande do Sul and Selection Pressure Analysis Planta Daninha
ULGUIM,A.R.; SILVA,B.M.; AGOSTINETTO,D.; AVILA NETO,R.C.; ZANDONÁ,R.R..
ABSTRACT: Among the species of the genus Cyperus occurring in paddy rice, C. esculentus, C. difformis and C. iria are the most important, and the latter two are reported as resistant to herbicides in Rio Grande do Sul (RS). The aim of this work was to characterize and mapping the occurrence of resistant plants of the genus Cyperus in areas of paddy rice in RS State, and to analyze the agronomic factors that interfere in the resistance selection. Seeds from individual plants were harvested in the 2014/15 crop, characterized as control failures by the application of acetolactate synthase inhibitors (ALS), totaling 43 samples. At the time of collection, the grower was inquired on aspects related to the management of weeds in the crop. Seeds were germinated in...

Activity of Antioxidant Enzymes in Euphorbia heterophylla Biotypes and their Relation to Cross Resistance to ALS and Protox Inhibitors Planta Daninha
XAVIER,E.; TREZZI,M.M.; OLIVEIRA,M.C.; VIDAL,R.A.; BRUSAMARELLO,A.P..
ABSTRACT: The characteristics of multiple resistance in Euphorbia heterophylla biotypes to herbicides that are inhibitors of ALS (Acetolactate synthase) and PPO (Protoporphyrinogen oxidase) and their responsible mechanisms are still not completely elucidated. The objectives of this study were to identify cross-resistance to herbicides from different chemical groups of ALS inhibitors (imidazolinones, sulfonylureas, pyrimidyl benzoates and sulfonanilides) and also PPO inhibitors (diphenylethers, phthalamides, oxadiazoles, triazolinones and pyrimidinediones) in E. heterophylla biotypes with multiple resistance to these herbicides; to analyze whether the antioxidant enzymes superoxide dismutase (SOD) and peroxidase (POD) constitute mechanisms that are...

Effects of monosulfuron-ester on metabolic processes of nitrogen-fixing cyanobacteria Anabaena flos-aquae and Anabaena azotica BJM
Shen,Jian Ying; Liao,Jin Zhi; Guo,Li Li; Su,Rui Fang.
Abstract Presence of the relatively new sulfonylurea herbicide monosulfuron-ester at 0.03-300 nmol/L affected the growth of two non-target nitrogen-fixing cyanobacteria (Anabaena flos-aquae and Anabaena azotica) and substantially inhibited in vitro Acetolactate synthase activity, with IC50 of 3.3 and 101.3 nmol/L for A. flos-aquae and A. azotica, respectively. Presenting in 30-300 nmol/L, it inhibited protein synthesis of the cyanobacteria with less amino acids produced as its concentration increased. Our findings support the view that monosulfuron-ester toxicity in both nitrogen-fixing cyanobacteria is due to its interference with protein metabolism via inhibition of branch-chain amino acid biosynthesis, and particularly Acetolactate synthase activity.
