Validation d’un procédé automatisé d’extraction sur phase solide de glandes digestives de moules pour l’identification et la quantification des dinophysistoxines en LC/ESI/SM2 par piégeage d’ions quadripolaire ArchiMer

This report presents a new extraction method of the dinophysistoxins (DTXs), confirmed by quantification using high-performance liquid chromatography coupled to mass spectrometry with an ion trap and electro spray interface (HPLC/ESI/MS2). The method originality consists on the adaptation of DTXs basic extraction procedure (liquid/ liquid) to a solid phase extraction (SPE) via a robotic station: ASPEC XLi The parameters of the automatization procedure were optimized to obtain the best DTXs recovery rate. Modélisation de la détoxication de mollusques bivalves contenant des phycotoxines paralysantes ou diarrhéiques ArchiMer

The purpose of this study is to implement models to describe detoxification kinetics of Pacific oysters Crassostrea gigas and Blue mussels Mytilus edulis containing paralytic and diarrheic phycotoxins respectively. First, experimental trials made it possible to complement data from the literature regarding the understanding of processes involved in mollusc detoxification. In particular, they pointed out a difference in the mollusc capacity to degrade cellwalls of phycotoxin-producing dinoflagellates, according to their en cystement capacities. It was observed that paralytic phycotoxins reached the various oyster tissues whereas diarrheic toxins were mainly sequestred in the digestive gland of mussels. Cytodétection des phycotoxines diarrhéiques - Une alternative au test de toxicité aiguë sur souris pour la surveillance de la salubrité des coquillages ArchiMer

The cytodetection test of diarrheic phycotoxin KB-cells (DRAME test) was developed to ultimately provide a substitute for the acute toxicity mouse-test currently used by IFREMER's phytoplankton monitoring network (REPHY). The cyctodetection test relies on the cell morphological changes induced by the phosphorylation of cytoskeleton proteins resulting from the inhibition of cell protein-phosphatases by okadaic-type toxins. The method is specific, sensitive and fast with results given on the same day. In addition, the method has been shown to produce a good correlation with the acute toxicity mouse-test as weil as with HPLC analyses of diarrheic phycotoxins. Cette étude s’est inscrite dans un contexte d’évaluation de la salubrité des mollusques bivalves destinés à la consommation humaine. Ces organismes peuvent en effet accumuler des toxines, en particulier de microalgues, en concentrations importantes, pouvant induire des intoxications chez les personnes les consommant. Afin d’assurer la protection des consommateurs, les toxines réglementées sont recherchées dans les coquillages par techniques physico-chimiques. En parallèle, les toxines émergentes, potentiellement produites par d’autres organismes que les micro-algues telles que les micromycètes, peuvent être detectées par l’utilisation du test de toxicité aigüe sur souris. Un nouvel outil dans l'identification et la quantification des dinophysistoxines (DTXs) : le couplage Chromatographie Liquide Haute Performance / Spectrométrie de masse par piégeage d'ions quadripôlaire et interface d'électro- nébulisation (CLHP/SM2) ArchiMer

This report presents a new method for detection and confirmed quantification of dinophysistoxins (DTXs) using high-performance liquid chromatography coupled to mass spectrometry with an ion trap and électrospray interface (HPLC/ESI/MS2). The parameters of the ESI source and ion trap spectrum analyser were optimised to provide detection of DTXs with maximum sensitivity. These improvements were obtained after reverse-phase separation on a C18 column, with a first elution to 0.1% TFA (75:25, v/v) in isocratic acetonitrile/water mode without flow split at a column flow rate of 200 µl/min for 15 min. En cas de fermeture de zone de production pour cause de présence d'une espèce de micro algues toxique, une mesure prévoit pour les professionnels de la conchyliculture l'interdiction générale d'utilisation de l'eau de mer déjà pompée dans un bassin insubmersible. C'est dans ce contexte que la DGAL a sollicité l'appui du LER BO pour la mise en place d'une étude dans des conditions professionnelles. The context of this study was the assessment of the safety of edible bivalve molluscs. Bivalve can indeed accumulate high concentrations of phycotoxins into their hepatopancreas, potentially leading to human intoxication outbreaks after consumption of such contaminated bivalves. In order to ensure the protection of shellfish consumers, regulated toxins are monitored using physico-chemical techniques. Etude de pré-validation du test de cytodétection des toxines diarrhéiques (acide okadaïque) dans les coquillages contaminés ArchiMer

A cytodetection test of diarrhoeic phycotoxins was developped by both IFREMER and Nantes University. Its principle is based on cell morphological changes of KB strain, induced by inhibition of protein-phosphatases involved in cytoskeleton building. This test has been shown sensitive, fast and specific for protein phosphatase inhibitors like diarrhoeic toxins (specialy okadaic acid in France).
