MICROBIOLOGICAL ACTIVITY DURING THE COMPOSTING OF WASTES FROM BROILER PRODUCTIVE CHAIN REA
Bernardi,Francieli H.; Costa,Mônica S. S. de M.; Costa,Luiz A. de M.; Damaceno,Felippe M.; Chiarelotto,Maico.
ABSTRACT The quality of the carbon used as bulking agent in the composting process directly affects the dynamics of the waste degradation, which is governed by an active and diversified microbiota. The monitoring of the basal respiration and enzymatic activities during the composting of broiler agro-industrial wastes with different carbon sources as bulking agents was the aim of this study. The microbiological activities were more intense in the thermophilic phase of the process. The dynamics of the enzymatic activities were affected by the different carbon sources used as bulking agent in the composition of the windrows. When the milled sugarcane bagasse was used as carbon source, the windrow presented the highest amount of P in its constitution and...

Ma, Hongming; Cahu, Chantal; Zambonino, Jose-luis; Yu, Hairui; Duan, Qingyuan; Le Gall, Marie-madeleine; Mai, Kangsen.
Large yellow croaker is an important marine fish species for aquaculture in China. Larval rearing of this species is still based on live prey feeding. The aim of this study was to describe the onset and development of the main digestive enzymes during larval development of this species, in order to obtain essential data for the formulation of a compound diet adapted to larvae. Enzymatic assays were conducted from day I post hatching to day 40 in larvae fed the following live prey sequence: rotifers, Artemia nauplii, copepods, and then frozen copepods. Assays were done on whole larvae until day 19, then on dissected larvae and on purified brush border membranes of intestine. Pancreatic enzymes, amylase, and trypsin, showed high activities from early stages,...

Influence of partial substitution of dietary fish meal on the activity of digestive enzymes in the intestinal brush border membrane of gilthead sea bream, Sparus aurata and goldfish, Carassius auratus ArchiMer
Silva, Flavia; Nicoli, Jacques R.; Zambonino, Jose-luis; Le Gall, Marie-madeleine; Kaushik, Sadasivam; Gatesoupe, François-joel.
The sustainable composition of diets of high nutritional value is of the utmost importance for intensive aquaculture. Digestion and absorption of nutrients depends on the activity of the digestive enzymes, in particular those located in the brush border membrane of enterocytes, which are responsible for the final stages of breaking down and absorption of nutrients. In the present study, the substitution of fish meal by lupin or rapeseed meal in the diet was evaluated on gilthead sea bream (Sparus aurata) and goldfish (Carassius auratus). The objectives were to compare the activities of intestinal brush border enzymes in both species fed the control and experimental diets. When gilthead sea bream were fed the vegetable diets, significantly lower activities...

Characterization of a highly thermostable alkaline phosphatase from the euryarchaeon Pyrococcus abyssi ArchiMer
Zappa, Sebastien; Rolland, Jean-luc; Flament, Didier; Gueguen, Yannick; Boudrant, Joseph; Dietrich, Jacques.
This work reports the first isolation and characterization of an alkaline phosphatase (AP) from a hyperthermophilic archaeon. An AP gene from Pyrococcus abyssi, a euryarchaeon isolated from a deep-sea hydrothermal vent, was cloned and the enzyme expressed in Escherichia coli. Analysis of the sequence showed conservation of the active site and structural elements of the E. coli AP. The recombinant AP was purified and characterized. Monomeric and homodimeric active forms were detected, with a monomer molecular mass of 54 kDa. Apparent optimum pH and temperature were estimated at 11.0 and 70 degreesC, respectively. Thus far, P. abyssi AP has been demonstrated to be the most thermostable AP, with half-lives at 100 and 105 degreesC of 18 and 5 h, respectively....

Growth and phosphorus uptake by the toxic dinoflagellate Alexandrium catenella (dinophyceae) in response to phosphate limitation ArchiMer
Jauzein, Cecile; Labry, Claire; Youenou, Agnes; Quere, Julien; Delmas, Daniel; Collos, Yves.
Alexandrium catenella (Whedon et Kof.) Balech has exhibited seasonal recurrent blooms in the Thau lagoon (South of France) since first reported in 1995. Its appearance followed a strong decrease (90%) in phosphate (PO(4)3-) concentrations in this environment over the 1970-1995 period. To determine if this dinoflagellate species has a competitive advantage in PO(4)3--limited conditions in terms of nutrient acquisition, semicontinuous cultures were carried out to characterize phosphorus (P) uptake by A. catenella cells along a P-limitation gradient using different dilution rates (DRs). Use of both inorganic and organic P was investigated from measurements of 33PO(4)3- uptake and alkaline phosphatase activity (APA), respectively. P status was estimated from...

Intraspecific variation in alkaline phosphatase activity in Phaeodactylum tricornutum (Bacillariophyceae, Bohlin) Acta Botanica
Lima,Domênica Teixeira de; Piedras,Fernanda Reinhardt; Nogueira,Flávia Previero; Richard,Eduardo da Cunha; Tocci,Bianca Reis Castaldi; Alba,Jose Juan Barrera; Moser,Gleyci Aparecida Oliveira.
ABSTRACT To describe potential intraspecific variation in phosphorus incorporation in two strains of Phaeodactylum tricornutum (Bohlin), Ub3 and Ub7, alkaline phosphatase (AP) activity was evaluated via enzyme-labeled fluorescence assay. Analysis using the probe ELF-97(r) provides individual evaluation, and therefore can determine the nutritional status of inorganic phosphorus in phytoplanktonic cells. Bioassays compared the control treatment to both phosphate-enriched and phosphate-depleted treatments by varying only the phosphate concentration in the media. The P. tricornutum strains exhibited differences in their development when incubated in the phosphate-enriched media. The development of the Ub7 strain differed by exhibiting "luxury uptake" and...

Application of Scharer's quantitative method for the determination of residual alkaline phosphatase activity in standard Minas Arq. Bras. Med. Vet. Zootec.
Soares,C.F.; Fonseca,L.M.; Leite,M.O.; Oliveira,M.C.P.P..
Milk pasteurization is a critical issue in the dairy industry, and failures in this process can affect final product safety. Scharer's enzymatic method is still traditionally used to verify pasteurization efficiency compliance, and it is based on screening for residual alkaline phosphatase in milk. Although several methods are used to quantify enzymatic activity to assess milk pasteurization efficiency, there is a small amount of published data regarding the use of these methods to quantify alkaline phosphatase in cheese. In this study, the Scharer's modified method was used to determine the levels of residual alkaline phosphatase in standard minas cheese, before and after 20 days of ripening. The cheeses were made using raw or pasteurized milk with the...

Tridax procumbens flavonoids: a prospective bioactive compound increased osteoblast differentiation and trabecular bone formation Biol. Res.
Mamun,Md. Abdullah Al; Hosen,Mohammad Jakir; Khatun,Amina; Alam,M. Masihul; Al-Bari,Md. Abdul Alim.
Abstract Background The Tridax procumbens extracts (TPE) are known for their ethno-medicinal properties to increase osteogenic functioning in mesenchymal stem cells. Recently, we found that the T. procumbens flavonoids (TPF) significantly suppressed the RANKL-induced osteoclasts differentiation and bone resorption. The TPF also promoted osteoblasts differentiation and bone formation demonstrated by increasing bone formation markers in cultured mouse primary osteoblasts. However, the effects of the TPF on in vivo bone formation remain unclear. In this study, we investigated the effects of the TPF on in vivo bone formation, injected the TPF (20 mg/kg) twice a day in the low calcium diet mice and killed them after 21 day. Radiographic and histomorphometric...

Contribution of matrix vesicles and alkaline phosphatase to ectopic bone formation BJMBR
Ciancaglini,P.; Simão,A.M.S.; Camolezi,F.L.; Millán,J.L.; Pizauro,J.M..
Endochondral calcification involves the participation of matrix vesicles (MVs), but it remains unclear whether calcification ectopically induced by implants of demineralized bone matrix also proceeds via MVs. Ectopic bone formation was induced by implanting rat demineralized diaphyseal bone matrix into the dorsal subcutaneous tissue of Wistar rats and was examined histologically and biochemically. Budding of MVs from chondrocytes was observed to serve as nucleation sites for mineralization during induced ectopic osteogenesis, presenting a diameter with Gaussian distribution with a median of 306 ± 103 nm. While the role of tissue-nonspecific alkaline phosphatase (TNAP) during mineralization involves hydrolysis of inorganic pyrophosphate (PPi), it is unclear...
