Background The quality of wheat grain depends on several characteristics, among which the composition of high molecular weight glutenin subunits, encoded by Glu-1 loci, are the most important. Application of biotechnological tools to accelerate the attainment of homozygous lines may influence the proportion of segregated genotypes. The objective was to determine, whether the selection pressure generated by the methods based on in vitro cultures, may cause a loss of genotypes with desirable Glu-1 alleles. Methods of gynogenesis, androgenesis and polyploidy induction were developed with the aim of genetic improvement in the rainbow trout, the Pacific oyster, the Manila clam and the black scallop. In the trout: · haploid androgenesis was induced by means of a pressure shock applied on diploid eggs, probably at the time of karyogamy. Diploid androgenetic individuals were directly produced from eggs fertilized with sperm from tetraploid male. · a heat shock treatment effective for suppressing the first egg mitosis was developed. This treatment allows the production of homozygous gynogenetic lines (checked using enzyme electrophoresis technique) from eggs inseminated with ultraviolet irradiated sperm. Towards the development of clonal lines in the European sea bass (Dicentrarchus labrax L.): application of uniparental reproduction techniques with an insight into sea bass eggs ArchiMer

Clonal lines are a powerful scientific tool for improved genetic characterization of organisms used in research. Inbred fish lines can be produced in only two generations using uniparental reproduction techniques. Androgenesis, achieved with variable success in several freshwater species, has been attempted in the European sea bass (Dicentrarchus labrax L), a marine fish of commercial and scientific interest. The low yields of progenies inheriting only the paternal genome after UV-irradiation of eggs led to considerations on the occurrence of UV screening compounds in pelagic eggs. Mycosporine-like amino acids and gadusol were found in many marine and freshwater organisms, but their occurrence in fish eggs was not clearly related to a behavioral pattern...

Androgenic response of Brazilian wheat genotypes to different pretreatments of spikes and to a gelling agent PAB

Abstract The objective of this work was to analyze the androgenic response of Brazilian wheat genotypes to different pretreatments of the spikes, prior to the culture of isolated microspores, and to the effect of a gelling agent in the induction culture medium. Five genotypes were evaluated for embryo formation, green plant regeneration, and spontaneous chromosome duplication. Wheat spikes were subjected to two pretreatments: cold, at 4ºC for 21 days; and 2-hydroxynicotinic acid, at 32ºC for two days. Culture media were evaluated with or without Ficoll as a gelling agent. Oat (Avena spp.) is poorly responsive to the haplodiploidization process, which leads to the production of homozygous lines in one step, increasing breeding efficiency. Androgenetic haploids in small grain cereal crops are obtained from microspores cultured at the mononucleate stage, which can be identified by the size of anthers. In the last three decades, research on soybean microspore embryogenesis was restricted to anther culture, which presents limitations such as the small number of responsive microspores and the high embryogenic potential of sporophytic tissues. Therefore, a sequence of studies was performed to establish appropriate conditions for the isolation and culture of soybean microspores and pollen grains as an alternative to anther culture. First, a pollen and microspore isolation technique was developed using floral buds from four soybean cultivars (Bragg, IAS 5, MG/BR-46 Conquista and BRSMT Uirapuru). Effect of temperature shock on soybean microspore embryogenesis BABT

In this study the effect of cold and heat shock on androgenetic induction in soybean anther culture was tested. Anthers of soybean were subjected to 4 and 33ºC, while the anthers used for control were maintained at 25ºC. Cytological analysis done during the 30 days of culture showed that frequencies of symmetrical binucleated and multicellular pollen grain did not differ among temperature treatments. Multicellular pollen grain could be formed by symmetrical division, as well as by assymetrical division. In relation to the embryo induction, the results of the treatments did not differ too. These results suggested that these treatments did not induce a sporophytic pathway in soybean.

Morphological markers for microspore developmental stage in maize BABT

The use of maize in anther culture has been limited because only few genotypes presented a high androgenetic potential. Obtaining the proper stage of microspore development at culture initiation is one of the most crucial factors for success in the androgenesis. For Brazilian maize genotypes there are no studies reporting a correlation between cytological features and morphological parameters. In this study, morphological parameters were recorded and associated with cytological specific stages of the the microsporogenesis in two Brazilian maize genotypes that were sowed in different places (field and growing chamber).
