Background The quality of wheat grain depends on several characteristics, among which the composition of high molecular weight glutenin subunits, encoded by Glu-1 loci, are the most important. Application of biotechnological tools to accelerate the attainment of homozygous lines may influence the proportion of segregated genotypes. The objective was to determine, whether the selection pressure generated by the methods based on in vitro cultures, may cause a loss of genotypes with desirable Glu-1 alleles. Abstract The objective of this work was to analyze the androgenic response of Brazilian wheat genotypes to different pretreatments of the spikes, prior to the culture of isolated microspores, and to the effect of a gelling agent in the induction culture medium. Five genotypes were evaluated for embryo formation, green plant regeneration, and spontaneous chromosome duplication. Wheat spikes were subjected to two pretreatments: cold, at 4ºC for 21 days; and 2-hydroxynicotinic acid, at 32ºC for two days. Culture media were evaluated with or without Ficoll as a gelling agent. Oat (Avena spp.) is poorly responsive to the haplodiploidization process, which leads to the production of homozygous lines in one step, increasing breeding efficiency. Androgenetic haploids in small grain cereal crops are obtained from microspores cultured at the mononucleate stage, which can be identified by the size of anthers. In the last three decades, research on soybean microspore embryogenesis was restricted to anther culture, which presents limitations such as the small number of responsive microspores and the high embryogenic potential of sporophytic tissues. Therefore, a sequence of studies was performed to establish appropriate conditions for the isolation and culture of soybean microspores and pollen grains as an alternative to anther culture.
