Haplodiploid androgenetic breeding in oat: genotypic variation in anther size and microspore development stage Scientia Agricola

Oat (Avena spp.) is poorly responsive to the haplodiploidization process, which leads to the production of homozygous lines in one step, increasing breeding efficiency. Androgenetic haploids in small grain cereal crops are obtained from microspores cultured at the mononucleate stage, which can be identified by the size of anthers. In order to identify the appropriate anther size for in vitro culture, microspore cytological analyses were made in Avena sativa cultivars UPF 7, UPF 18, UFRGS 14, Stout and Avena sterilis CAV 3361, cultivated in growth chamber under controlled light and temperature conditions. Production of haploids is an important tool for the rapid generation of homozygous breeding lines. Experiments have been conducted with the aim of developing a method for haploids peanut plant production by us -ing anther culture. Peanut genotype Tainan 9 was tested with different media compositions. The suitable medium for anther culture of peanut in this study was N6 The callus was white to creamish at the time of induction with loose to compact form. Callus proliferation was well developed in MS medium supplemented with 4 mg/l IAA and 2 mg/l Kinetin. In some cases, green spots were observed in the callus.

Anther culture (AC) technology is being used to conduct rice blast resistant true breeding line at Phrae Rice Research Center in 2012. Five crosses from irrigated rice research and development program were cultured on N6 formula media to induce callus formation in the dark room, 20-250C and light intensity about 2000 Lux for 1-2 months and followed by MS media for 1-2 months. Then, the plantlet was transplanted to nursery and 96 progenies of five crosses were planted to observational trial in the field during wet season, 2014. Marker-assisted selection for the rice blast resistance gene Pi-ar in a backcross population. Repositório Alice

A doubled-haploid (DH) population, obtained by anther culture of F 1 plants from a cross between a highly susceptible rice cultivar Lijiangxintuanheigu and the resistant somaclone (SC09), of the cultivar Araguaia, was used to identify RAPD markers linked to the blast resistance gene Pi-ar. The 86 DH plants, inoculated with the race IB-9 of Magnaporthe oryzae, segregated in 1:1 ratio of resistant and susceptible plants. Of the 67 primers used 31 produced DNA profiles that differentiated resistant and susceptible bulks as well as the parental cultivars. Association between floral bud size and developmental stage in soybean microspores BABT

This study was carried out to establish the association between floral bud size and the corresponding microspore developmental stages for Brazilian soybean cultivars. Microspore developmental stage distributions were examined in young buds from cv Década, IAS5 and RS7. The data indicated that for a given bud-size group, the microspores of different cultivars were at different developmental stages, with cv RS7 and Década distributed at the youngest and cv IAS5 at the most advanced stages. Microspore stages distribution were also compared among the ten anthers of the same bud of the above cultivars. Effect of temperature shock on soybean microspore embryogenesis BABT

In this study the effect of cold and heat shock on androgenetic induction in soybean anther culture was tested. Anthers of soybean were subjected to 4 and 33ºC, while the anthers used for control were maintained at 25ºC. Cytological analysis done during the 30 days of culture showed that frequencies of symmetrical binucleated and multicellular pollen grain did not differ among temperature treatments. Multicellular pollen grain could be formed by symmetrical division, as well as by assymetrical division. In relation to the embryo induction, the results of the treatments did not differ too. These results suggested that these treatments did not induce a sporophytic pathway in soybean.

Morphological markers for microspore developmental stage in maize BABT

The use of maize in anther culture has been limited because only few genotypes presented a high androgenetic potential. Obtaining the proper stage of microspore development at culture initiation is one of the most crucial factors for success in the androgenesis. For Brazilian maize genotypes there are no studies reporting a correlation between cytological features and morphological parameters. In this study, morphological parameters were recorded and associated with cytological specific stages of the the microsporogenesis in two Brazilian maize genotypes that were sowed in different places (field and growing chamber). Evaluation of gelling agents on anther culture: response of two soybean cultivars BABT

Anthers of two soybean cultivars were cultured in B5 long basal culture media gelled with agarose or PhytagelTM. Cytological examinations of the anthers were carried out during the first 45 days of culture to assay the viability and developmental stage of microspores. Frequency of callus formation was recorded at 45 days of culture. The analysis of variance of the microspore viability assay showed significant Cultivar X Gelling Agent X Day of Culture interactions. Ontogenetic events in androgenesis of Brazilian barley genotypes BJB

This paper describes a simple procedure for obtention of barley (Hordeum vulgare L.) doubled haploid plants from Brazilian hybrid genotypes. Anatomical and histological examinations showed the reversion of barley pollen to an sporophytic mode of development. A sequence of mitosis led to the formation of multicellular pollen grains. Regeneration of plants occurred either by direct embryogenesis or callus formation followed by differentiation through direct embryogenesis or organogenesis. Plants were formed in the same medium used for induction dispensing an additional regeneration step. This procedure makes doubled haploid production simpler and faster.
