Evaluación de las propiedades antioxidantes y antimicrobianas de dos especies del hongo medicinal ganoderma nativo de México y su contribución al desarrollo regional. Colegio de Postgraduados
Quiriz Cerezo, Fabiola Argelia.

Kim,Jong-Wan; Yoe,Jeehyun; Lee,Gil Ho; Yoe,Sung Moon.
The chicken-type lysozyme of the insect Spodoptera litura (SLLyz) is a polypeptide of 121 amino acids containing four disulfide bridges and 17 rare codons and participates in innate defense as an anti-bacterial enzyme. The recombinant S. litura lysozyme (rSLLyz) expressed as a C-terminal fusion protein with glutathione S-transferase (GST) in Rosetta(DE3) Singles. The protein was produced as an inclusion body which was solubilized in 8 M urea, renatured by on-column refolding, and purified by reversed-phase chromatography to 95% purity. The purified rSLLyz demonstrated antibacterial activity against B. megaterium confirmed by inhibition zone assay. The overexpression and refolding strategy described in this study will provide a reliable technique for...

Novel antimicrobial secondary metabolites from a Penicillium sp. isolated from Brazilian cerrado soil Electron. J. Biotechnol.
Petit,Philippe; Lucas,Esther M. F.; Abreu,Lucas M; Pfenning,Ludwig H; Takahashi,Jacqueline A.
The morphological features of a Penicillium, isolated from Brazilian cerrado soil, were characterized and showed to be distinctly different from all well-defined Penicillium species. Chemical and biological investigation on the ethyl acetate extract of this Penicillium isolate resulted in the isolation of three new naphthalenoids: a major metabolite, methyl 6-acetyl-4-methoxy-5,7,8-trihydroxynaphthalene-2-carboxylate and two minor ones, methyl 6-acetyl-4-methoxy-7,8-dihydroxynaphthalene-2-carboxylate and methyl 6-acetyl-4-methoxy-5,8-dihydroxynaphthalene-2-carboxylate. Their structures were determined based on their mono and bidimensional nuclear magnetic resonance data. Acetyl, allyl and methoxyl derivatives of the major metabolite were prepared in order...

Évaluation d’une suite de bio-essais pour la détection et l’étude de composés lipophiles de micro-organismes marins issus de mollusques bivalves et de leur environnement ArchiMer
Geiger, Marie.
The context of this study was the assessment of the safety of edible bivalve molluscs. Bivalve can indeed accumulate high concentrations of phycotoxins into their hepatopancreas, potentially leading to human intoxication outbreaks after consumption of such contaminated bivalves. In order to ensure the protection of shellfish consumers, regulated toxins are monitored using physico-chemical techniques. In parallel, emerging toxins, that can be putatively produced by microalgae or marine fungi, are monitored using the intraperitoneal mouse bioassay. However, this bioassay suffers ethical and methodological drawbacks. Thus, the objective of this work was to assess the ability of a bioassay suite to detect lipophilic toxins. This suite was composed of three...

Complex Relationships between the Blue Pigment Marennine and Marine Bacteria of the Genus Vibrio ArchiMer
Falaise, Charlotte; James, Adele; Travers, Marie-agnes; Zanella, Marie; Badawi, Myriam; Mouget, Jean-luc.
Marennine, the water-soluble blue pigment produced by the marine diatom Haslea ostrearia, is known to display antibacterial activities. Previous studies have demonstrated a prophylactic effect of marennine on bivalve larvae challenged with a pathogenic Vibrio splendidus, suggesting that the blue Haslea is a good candidate for applications in aquaculture as a source of a natural antimicrobial agent. Indeed, the genus Vibrio is ubiquitous in aquaculture ecosystems, and regular events of pathogenic invasion cause some of the biggest losses worldwide. To better characterize the effects of marennine on Vibrios, a panel of 30 Vibrio strains belonging to 10 different species was tested, including bivalve pathogenic species (e.g., Vibrio crassostreae and Vibrio...

Review and perspectives of physiological mechanisms underlying genetically-based resistance of the Pacific oyster Crassostrea gigas to summer mortality ArchiMer
Samain, Jean-francois.
French oyster farming has been subject to severe mortalities during the summer months. Results from the research program "Morest", which ran from 2000 to 2006 and examined the possible causes of these mortalities, led to the construction of a model to explain the interaction between environmental factors, oyster physiology and different opportunistic pathogens underlying oyster summer mortality. Temperature, food, reproduction and stress were the main factors required for oyster mortality. Genetically-based resistance ("R" oysters) or susceptibility ("S" oysters) to summer mortality was revealed by divergent selection. Building on these results, a literature search was made in 2007 on the molecular origin of genetic resistance to such a complex mortality...

The wide polarity range and highly polar compounds of two selected red seaweed, Grateloupia turuturu and Palmaria palmata were extracted using two different types of solvent, dichloromethane/methanol and methanol/water. Monthly in vitro antibacterial activities were studied using the microplate method against the marine bacteria Vibrio harveyi strain ORM4, known to infect abalone. Inhibition, slowdown and delay of Vibrio harveyi growth were investigated. Polar compounds of seaweed showed an activity against the abalone pathogen. The best activity was recorded from P. palmata collected in spring, with an inhibition of 7.9% of the bacterial growth. Preliminary 1H NMR profiles identified the differences between the extracts.

NMR structure of mussel mytilin, and antiviral-antibacterial activities of derived synthetic peptides ArchiMer
Roch, Philippe; Yang, Y; Toubiana, Mylene; Aumelas, A.
Mytilin is a 34-residue antibacterial peptide from the mussel Mytilus galloprovincialis, which in addition possesses in vitro antiviral activity. The three-dimensional solution structure of the synthetic mytilin was established by using 1H NMR and consists of the common cysteine-stabilized alpha beta motif close to the one observed in the mussel defensin MGD-1. Mytilin is characterized by 8 cysteines engaged in four disulfide bonds (2-27, 6-29, 10-31, and 15-34) only involving the beta-strand II. Hydrophilic and hydrophobic areas of mytilin account for 63% and 37%, respectively, a ratio very close to that of MGD-1 (64% and 36%). One linear and three cyclic fragments were designed from the interstrand loop sequence known to retain the biological activities...

First evidence of a potential antibacterial activity involving a laccase-type enzyme of the phenoloxidase system in Pacific oyster Crassostrea gigas haemocytes ArchiMer
Luna-acosta, Andrea; Saulnier, Denis; Pommier, Mylene; Haffner, Philippe; De Decker, Sophie; Renault, Tristan; Thomas-guyon, Helene.
Phenoloxidases (POs) are a group of copper proteins including tyrosinase, catecholase and laccase. In several insects and crustaceans, antibacterial substances are produced through the PO cascade, participating in the direct killing of invading microorganisms. However, although POs are widely recognised as an integral part of the invertebrate immune defence system, experimental evidence is lacking that these properties are conserved in molluscs, and more particularly in the Pacific oyster Crassostrea gigas. In the present study, Vibrio splendidus LGP32 and Vibrio aestuarianus 02/041 growths were affected, after being treated with C. gigas haemocyte lysate supernatant (HLS), and either a common substrate of POs, l-3,4-dihydroxyphenylalanine (l-DOPA), to...

Purification and biological effects of a C-type lectin isolated from Bothrops moojeni J. Venom. Anim. Toxins incl. Trop. Dis.
Barbosa,PSF; Martins,AMC; Toyama,MH; Joazeiro,PP; Beriam,LOS; Fonteles,MC; Monteiro,HSA.
Snake venom proteins from the C-type lectin family have very distinct biological activities despite their highly conserved primary structure, which is homologous to the carbohydrate recognition region of true C-type lectins. We purified a lectin-like protein (BmLec) from Bothrops moojeni venom and investigated its effect on platelet aggregation, insulin secretion, antibacterial activity, and isolated kidney cells. The BmLec was purified using two chromatographic steps: affinity chromatography and reverse phase high performance liquid chromatography (HPLC). BmLec showed a dose-dependent platelet aggregation and significantly decreased the bacterial growth rate in approximately 15%. During scanning electron microscopy, the profile of Xanthomonas axonopodis...

Nowadays, it is widely known that resistant pathogenic strains are becoming one of the greatest health problems related to human sciences. It is also known that fatty acids can present antimicrobial activity. The corn and soy oils are rich in fatty acids. In this regard, this work aimed to evaluate antibacterial and modulatory activity of these two industrial fixed oils. Both tests were performed using the microdilution method to Minimum inhibitory concentration under 1024 mg/ml. The aminoglycosides activity was enhanced by combining corn oil using E. coli 27. One of the mechanisms that might explain the synergism toward the corn oil, in part, might be due to the hidrophobic nature of the saturated and unsaturated lipids present in the sample. The results...
