Effects de le temperature et du regime alimentaire sur certains parametres de l'immunite cellulaire du loup de mer en elevage ArchiMer
Deschaux, P; Peres, G.
The data showed that antibody response and NK cells activity of a teleostean fish Dicentrarchus labrax (Loup de mer) were maximal when animals were bred at a temperature equal to 18 degree C. A proteinoprive diet enhanced T cytotoxic response and diminished NK cell activity. A fast of 15 days had no effect on these two cellular responses.

Fixation d''un anticorps anti-lysozyme dans des cellules de mollusques bivalves ArchiMer
Auffret, Mathieu; Diouris, M; Herry, A.
A polyclonal rabbit anti-human lysozyme (Dako) was applied on histological sections from bivalve mollusc and revealed with a mouse anti-rabbit IgG fluorescein conjugated antibody to localise cellular lysozyme in tissues where its activity had previously detected by enzymatic techniques. Labelling occurred in the cytoplasm of epithelial cells covering the body. This is in agreement with the intracellular distribution of this enzyme. In the Lucinid Lucinella divaricata gills, the cytoplasm of bacteriocytes was also labelled. This suggests that antibacterial activity occurred in these cells. Control sections indicate that fluorescence was a consequence of true primary antibody fixation. However, its specificity should be verified by electrophoretical...

TOXIC SHOCK SYNDROME J. Venom. Anim. Toxins
BERGDOLL,M. S..
Toxic shock syndrome is a staphylococcal disease caused by toxins produced by the staphylococci, toxic shock syndrome toxin-1 and enterotoxin B. The disease results from staphylococci growing in the vagina with the use of tampons during menstruation, primarily in young women. However, any staphylococcal infection can result in toxic shock syndrome if the staphylococci produce the appropriate toxins and the individual has no antibodies to the toxins. The symptoms can be quite severe, with high fever, low blood pressure, diffuse macular erythroderma, orthostatic dizziness, vomiting and or diarrhea at the onset, severe myalgia, peeling of the skin of the palms of the hands and the soles of the feet after seven to ten days, and death in some cases. The disease...

In vitro assay of biological and chemical toxins using antibodies against lethal toxin neutralizing factor J. Venom. Anim. Toxins
LIPPS,B. V..
Lethal Toxin Neutralizing Factor (N-LTNF), MW 63.0 kDa, was isolated from opossum serum. After trypsin digestion, the active domain of N-LTNF was isolated and sequenced. The synthetic peptide consisting of ten amino acids was designated as LT-10. N-LTNF and LT-10 inhibited the lethality of animal, plant and bacteria toxins when tested on mice non-immunologically. The antibodies against N-LTNF and LT-10 only reacted immunologically with toxins and not with non-toxic substances. Anti-LTNF and anti-LT-10 reacted immunologically by ELISA test with toxins that were not detected by mouse test, such as cholera toxin and digoxin. Anti-LTNF and anti-LT-10 failed to react immunologically with non-toxic substances, such as nerve growth factor and collagen. Currently,...

Leptospira spp. antibodies in captive coatis (Nasua nasua Storr, 1780) (Carnivora: Procyonidae) J. Venom. Anim. Toxins incl. Trop. Dis.
Langoni,H; Kawaguchi,MF; Oshika,JC; Da Silva,RC; Teixeira,CR.
Leptospirosis is a worldwide infection that affects many species, including wild animals. The present study aimed to detect Leptospira spp. antibodies in 17 captive coatis (Nasua nasua) by the microscopic agglutination test (MAT). Nine (52.94%) animals tested positive and the following serovars were identified: Copenhageni (22.22%), Shermani (22.22%), Andamana (11.11%), Wolfii (11.11%) and Pyrogenes (11.11%). Two samples presented coagglutination, one (11.11%) for Hebdomadis and Wolfii, and another (11.11%) for Hebdomadis, Hardjo and Wolfii. The current study revealed the presence of the infection even in animals without any clinical signs, reinforcing the possibility that wild animals in captivity may be infected by leptospire serovars, thus enabling...

Detection of antibodies to Toxoplasma gondii in wild animals in Brazil J. Venom. Anim. Toxins incl. Trop. Dis.
Silva,Rodrigo Costa da; Machado,Gustavo Puglia; Cruvinel,Tatiane Morosini de Andrade; Cruvinel,Ciro Alexandre; Langoni,Helio.
Background Toxoplasmosis is a worldwide zoonosis caused by an obligate intracellular protozoan parasite,Toxoplasma gondii, that affects all warm-blooded animals, including wild animals. The increased number of cases of parasitic infections is mainly due to the destruction of environmental conservation areas, which is driving wild animals out of their habitats and towards urban areas. In this study, the occurrence of T. gondii infection was investigated by the modified agglutination test (MAT) in 26 different species of run over and injured wild animals that were treated at a Brazilian university veterinary hospital, from June 2007 to August 2008.Findings Of the studied animals, six (23.1%; CI95% 11.1-42.2%) had T. gondii antibodies, with titers equal to 10...

Diagnosis of infectious bronchitis: an overview of concepts and tools Rev. Bras. Ciênc. Avic.
Villarreal,LYB.
Infectious bronchitis (IB) casues multi-systemic infection in chickens with signs similar caused by other poultry pathogens and thus a high diagnostic accuracy can only be achieved by s series of laboratory assays. This article reviews in a brief way the traditional virus assays such as embryo innoculation, tracheal rings and virus neutralization assays for the direct detection of Avian infectious bronchitis virus (IBV) and methods based on gene molecular biology and some assays for the detection of anti-IBV antibodies, including ELISA. A critical view on each technique is also provived by the author.

Development of an enzyme-linked immunosorbent assay for quantification of Salmonella enteritidis-specific antibodies in egg yolk Rev. Bras. Ciênc. Avic.
Tayeb,IT; Nehme,P; Jaber,L; Barbour,EK.
The present study aims at developing an indirect ELISA to quantify yolk antibodies specific to all surface proteins of the invasive Salmonella Enteritidis (SE), which acquired the 1.8, 14.1, and ~ 50 Kb plasmids. An ELISA checkerboard was used in four different experiments to account for the different parameters included in the preliminary ELISA procedure, and consequently to maximize the difference in Optical Density (OD) values between control positive and negative yolk samples. The first experiment aimed at studying the impact of 5% Bovine Serum Albumin (BSA) dissolved in distilled water as a blocking reagent on a 28 µg/well SE antigen-coated plate, while applying the positive and negative control yolk samples to different concentrations of...

Colostral immunoglobulins absorption in Canchim and Nelore calves R. Bras. Zootec.
Machado Neto,Raul; Packer,Irineu Umberto; Prado,Gladys Villas Boas do; Bessi,Rosana; Pauletti,Patricia.
The efficiency of absorption of colostral immunoglobulins was evaluated in five Canchim and seven Nelore calves. They received colostrum pools with concentration of 70.20 ± 6.14 mg/mL through esofageal feeder at 2, 12, 24 and 36 hours after birth. The immunoglobulins concentrations of the pools were estimated through specific gravity and measured by radial immunodifusion. In the blood collection at birth and during the first 70 days of life, the total protein was assayed by biuret method and the immunoglobulins were assayed by radial immunodifusion. Data were analysed as a randomized split-plot statistical model. The highest concentrations of serum immunoglobulins and total protein were observed at 24 hours of age. No significant differences (P>0.5484)...

HUMORAL RESPONSE TO DIFFERENT VACCINATION SCHEMES AGAINST CLASSICAL SWINE FEVER (CSF) SUCCESSIVELY APPLIED DURING AN OUTBREAK OF THE DISEASE Rev Salud Anim.
Percedo,María Irian; Alfonso,P; Frías,María Teresa; Díaz de Arce,Heydi; Barrera,Maritza; Fonseca,O; Castell,Sara.
Vaccination against classical swine fever (CSF) is an essential measure for its control in endemic areas, and the Chinese Strain has been used with success in already disease free countries. Nevertheless the complexity of the factors interacting on the epidemiology and control of CSF, when the expected success is not reached with the vaccination of premises, doubts always fall on the vaccine used and the vaccination scheme applied In consecuences, many changes in vaccination scheme by the breeders are frecuently observed whitout positive influence in CSF control. To clarify the CSF recurrence in a herd vaccinated by a different scheme as the one recommended in the national control program, serological samples were taken to different animal groups in three...

Quality of colostral passive immunity and pattern of serum protein fluctuation in newborn calves Scientia Agricola
Pauletti,Patricia; Machado Neto,Raul; Packer,Irineu Umberto; D'Arce,Raul Dantas; Bessi,Rosana.
Immunity acquired by newborn animals is known as passive immunity, and for ruminants, antibody acquisition depends on the ingestion and absorption of adequate amounts of immunoglobulins from colostrum. This study relates different initial levels of acquired passive protection and serum total protein (TP) and immunoglobulin G (IgG). Serum immunoglobulin concentration and total protein were evaluated for female Holstein calves in the first sixty days of life. Animals were separated into three groups according to their initial level of passive immunity: group 1- animals with a low level of passive immunity (below 20 mg mL-1); group 2- animals with a medium level (between 20 and 30 mg mL-1), and group 3- animals with a high level (above 30 mg mL-1). Expression of Grapevine leafroll-associated virus 3 coat protein gene in Escherichia coli and production of polyclonal antibodies Trop. Plant Pathol.
Fajardo,Thor V.M.; Barros,Danielle R.; Nickel,Osmar; Kuhn,Gilmar B.; Zerbini,F. Murilo.
Grapevine leafroll-associated virus 3 (GLRaV-3), the main viral species of the grapevine leafroll complex, causes yield and quality reduction in grapes (Vitis spp.). The coat protein gene was RT-PCR-amplified from total RNA extracted from infected grapevine leaves and the amplified fragment was cloned and completely sequenced. The fragment was subsequently subcloned into the pRSET-C expression vector. The recombinant plasmid was used to transform Escherichia coli BL21:DE3 and express the capsid protein. The coat protein, fused to a 6 His-tag, was purified by affinity chromatography using an Ni-NTA resin. The identity of the purified protein was confirmed by SDS-PAGE and Western blot. The in vitro-expressed protein was quantified and used for rabbit...
