Diversity of mycobiota in colonies of different species of leaf-cutting ants and sampling sites across Argentina Ecología austral
Marfetan,Jorge A; Folgarait,Patricia J.
Attine ants are a monophyletic group comprising more than 230 species, distributed exclusively in the New World. All higher Attini ants depend on the cultivation of fungus gardens for food, and those gardens are continuously exposed to alien microorganisms. The present work describes, for the first time, the composition, relative frequency and the richness of the fungal species comprising the mycobiota from the garden of the most abundant Acromyrmex species from different regions of Argentina. We also compared, using a multivariate approach, the mycobiota composition with the purpose of testing two hypotheses: 1) whether mycobiota is defined by the ant species, or 2) whether mycobiota is determined by the geographical region. We found that each fungal...

Redefining Microascus, Scopulariopsis and allied genera Naturalis
Sandoval-Denis, M.; Gené, J.; Sutton, D.A.; Cano-Lira , J.F.; Hoog, G.S. de; Decock, C.A.; Wiederhold, N.P.; Guarro, J..
The genera Microascus and Scopulariopsis comprise species commonly isolated from soil, decaying plant material and indoor environments. A few species are also recognised as opportunistic pathogens of insects and animals, including humans. In the past, the taxonomy of these fungi has been based on morphology only. With the aim to clarify the taxonomy and phylogeny of these fungi, we studied a large set of clinical and environmental isolates, including the available ex-type strains of numerous species, by means of morphological, physiological and molecular analyses. Species delineation was assessed under the Genealogical Phylogenetic Species Recognition (GCPSR) criterion using DNA sequence data of four loci (ITS region, and fragments of rDNA LSU, translation...

Roselliniella revealed as an overlooked genus of Hypocreales, with the description of a second species on parmelioid lichens Naturalis
Hawksworth, D.L.; Millanes, A.M.; Wedin, M..
Based on newly obtained 28S rDNA sequences from Roselliniella atlantica and R. euparmeliicola sp. nov., the genus Roselliniella has to be placed in Hypocreales and not in Sordariales; however, the family placement could not be resolved from the sequences obtained. The mature ascospores are single-celled and brown, but young ascospores are hyaline and sometimes have a median septum. The new species occurs on a Parmelia s.str. species in China, and differs in 24 nucleotide substitution positions in the nu-LSU rDNA region and ascospore size from R. atlantica. In this case, small variations in ascospore sizes and shape prove to be phylogenetically and taxonomically informative. The two species occur in the same clade with 95 % jack-knife support. Roselliniella...

The Colletotrichum gigasporum species complex Naturalis
Liu, F.; Cai, L.; Crous, P.W.; Damm, U..
In a preliminary analysis, 21 Colletotrichum strains with large conidia preserved in the CBS culture collection clustered with a recently described species, C. gigasporum, forming a clade distinct from other currently known Colletotrichum species complexes. Multi-locus phylogenetic analyses (ITS, ACT, TUB2, CHS-1, GAPDH) as well as each of the single-locus analyses resolved seven distinct species, one of them being C. gigasporum. Colletotrichum gigasporum and its close allies thus constitute a previously unknown species complex with shared morphological features. Five of the seven species accepted in the C. gigasporum species complex are described here as novel species, namely C. arxii, C. magnisporum, C. pseudomajus, C. radicis and C. vietnamense. A...

Asperisporium and Pantospora (Mycosphaerellaceae): epitypifications and phylogenetic placement Naturalis
Minnis, A.M.; Kennedy, A.H.; Grenier, D.B.; Rehner, S.A.; Bischoff, J.F..
The species-rich family Mycosphaerellaceae contains considerable morphological diversity and includes numerous anamorphic genera, many of which are economically important plant pathogens. Recent revisions and phylogenetic research have resulted in taxonomic instability. Ameliorating this problem requires phylogenetic placement of type species of key genera. We present an examination of the type species of the anamorphic Asperisporium and Pantospora. Cultures isolated from recent port interceptions were studied and described, and morphological studies were made of historical and new herbarium specimens. DNA sequence data from the ITS region and n LSU were generated from these type species, analysed phylogenetically, placed into an evolutionary context...

Trichoderma (Hypocrea) species with green ascospores from China Naturalis
Zhu, Z.X.; Zhuang, W.Y..
Stromata of Trichoderma species having green ascospores were collected in various regions of China. Based on morphology of the sexual and asexual morph, culture characteristics, and sequence analyses of rpb2 and tef1 genes, 17 species with green ascospores were identified. Among them, Trichoderma rosulatum, T. rufobrunneum and T. stipitatum are described as new species, and seven other species are reported for the first time from China. Trichoderma rosulatum produces small bright yellow or pale greenish stromata with dense dark green ostioles and gliocladium-like conidiophores, shows a close relationship to T. thelephoricola, and belongs to the Chlorospora clade. Trichoderma rufobrunneum, which typically forms reddish brown stromata, is recognised as a...

Juglanconis gen. nov. on Juglandaceae, and the new family Juglanconidaceae (Diaporthales) Naturalis
Voglmayr, H.; Castlebury, L.A.; Jaklitsch, W.M..
Molecular phylogenetic analyses of ITS-LSU rDNA sequence data demonstrate that Melanconis species occurring on Juglandaceae are phylogenetically distinct from Melanconis s.str., and therefore the new genus Juglanconis is described. Morphologically, the genus Juglanconis differs from Melanconis by light to dark brown conidia with irregular verrucae on the inner surface of the conidial wall, while in Melanconis s.str. they are smooth. Juglanconis forms a separate clade not affiliated with a described family of Diaporthales, and the family Juglanconidaceae is introduced to accommodate it. Data of macro- and microscopic morphology and phylogenetic multilocus analyses of partial nuSSU-ITS-LSU rDNA, cal, his, ms204, rpb1, rpb2, tef1 and tub2 sequences revealed...

Two new classes of Ascomycota: Xylobotryomycetes and Candelariomycetes Naturalis
Voglmayr, H.; Fournier, J.; Jaklitsch, W.M..
Phylogenetic analyses of a combined DNA data matrix containing nuclear small and large subunits (nSSU, nLSU) and mitochondrial small subunit (mtSSU) ribosomal RNA and the largest and second largest subunits of the RNA polymerase II (rpb1, rpb2) of representative Pezizomycotina revealed that the enigmatic genera Xylobotryum and Cirrosporium form an isolated, highly supported phylogenetic lineage within Leotiomyceta. Acknowledging their morphological and phylogenetic distinctness, we describe the new class Xylobotryomycetes, containing the new order Xylobotryales with the two new families Xylobotryaceae and Cirrosporiaceae. The two currently accepted species of Xylobotryum, X. andinum and X. portentosum, are described and illustrated by light and scanning...

Resolution of morphology-based taxonomic delusions: Acrocordiella, Basiseptospora, Blogiascospora, Clypeosphaeria, Hymenopleella, Lepteutypa, Pseudapiospora, Requienella, Seiridium and Strickeria Naturalis
Jaklitsch, W.M.; Gardiennet, A.; Voglmayr, H..
Fresh material, type studies and molecular phylogeny were used to clarify phylogenetic relationships of the nine genera Acrocordiella, Blogiascospora, Clypeosphaeria, Hymenopleella, Lepteutypa, Pseudapiospora, Requienella, Seiridium and Strickeria. At first sight, some of these genera do not seem to have much in common, but all were found to belong to the Xylariales, based on their generic types. Thus, the most peculiar finding is the phylogenetic affinity of the genera Acrocordiella, Requienella and Strickeria, which had been classified in the Dothideomycetes or Eurotiomycetes, to the Xylariales. Acrocordiella and Requienella are closely related but distinct genera of the Requienellaceae. Although their ascospores are similar to those of Lepteutypa,...

Phylogeny and taxonomic revision of the Planistromellaceae including its coelomycetous anamorphs: contributions towards a monograph of the genus Kellermania Naturalis
Minnis, A.M.; Kennedy, A.H.; Grenier, D.B.; Palm, M.E.; Rossman, A.Y..
The core species of the family Planistromellaceae are included in the teleomorphic genera Planistroma and Planistromella and the connected anamorphic, coelomycetous genera Alpakesa, Kellermania, and Piptarthron. These genera have been defined primarily on the basis of ascospore septation or number of conidial appendages. Due to a lack of DNA sequence data, phylogenetic placement of these genera within the Dothideomycetes, evaluation of monophyly, and questions about generic boundaries could not be adequately addressed in the past. Isolates of nearly all of the known species in these genera were studied genetically and morphologically. DNA sequence data were generated for the nSSU, ITS, nLSU, and RPB1 markers and analysed phylogenetically. These results...

Phylogenetic and taxonomic revision of Lopadostoma Naturalis
Jaklitsch, W.M.; Fournier, J.; Rogers, J.D.; Voglmayr, H..
The genus Lopadostoma (Xylariaceae, Xylariales) is revised. Most species formerly assigned to Lopadostoma do not belong to the genus. Twelve species are herein recognised, of which two are only known from morphology. Ten species, of which six (L. americanum, L. fagi, L. insulare, L. lechatii, L. meridionale and L. quercicola) are newly described, are characterised by both morphology and DNA phylogeny using LSU, ITS and rpb2 sequences. Morphologically, ecologically and phylogenetically Lopadostoma is a well-defined genus comprising exclusively species with pustular pseudostroma development in bark of angiospermous trees. Phaeosperma ailanthi, Phaeosperma dryophilum and Sphaeria linosperma are combined in Lopadostoma. Lopadostoma gastrinum is neotypified and...

New primers for promising single-copy genes in fungal phylogenetics and systematics Naturalis
Schmitt, I.; Crespo, A.; Divakar, P.K.; Fankhauser, J.D.; Herman-Sackett, E.; Kalb, K.; Nelsen, M.P.; Nelson, N.A.; Rivas-Plata, E.; Shimp, A.D.; Widhelm, T.; Lumbsch, H.T..
Developing powerful phylogenetic markers is a key concern in fungal phylogenetics. Here we report degenerate primers that amplify the single-copy genes Mcm7 (MS456) and Tsr1 (MS277) across a wide range of Pezizomycotina (Ascomycota). Phylogenetic analyses of 59 taxa belonging to the Eurotiomycetes, Lecanoromycetes, Leotiomycetes, Lichinomycetes and Sordariomycetes, indicate the utility of these loci for fungal phylogenetics at taxonomic levels ranging from genus to class. We also tested the new primers in silico using sequences of Saccharomycotina, Taphrinomycotina and Basidiomycota to predict their potential of amplifying widely across the Fungi. The analyses suggest that the new primers will need no, or only minor sequence modifications to amplify...

Families and genera of diaporthalean fungi associated with canker and dieback of tree hosts Naturalis
Fan, X.L.; Bezerra, J.D.P.; Tian, C.M.; Crous, P.W..
In this study we accept 25 families in Diaporthales based on phylogenetic analyses using partial ITS, LSU, rpb2 and tef1-α gene sequences. Four different families associated with canker and dieback of tree hosts are morphologically treated and phylogenetically compared. These include three new families (Diaporthostomataceae, Pseudomelanconidaceae, Synnemasporellaceae), and one new genus, Dendrostoma (Erythrogloeaceae). Dendrostoma is newly described from Malus spectabilis, Osmanthus fragrans and Quercus acutissima having fusoid to cylindrical, bicellular ascospores, with three new species namely D. mali, D. osmanthi and D. quercinum. Diaporthostomataceae is characterised by conical and discrete perithecia with bicellular, fusoid ascospores on branches of...
