Effective antigen-retrieval method for immunohistochemical detection of abnormal isoform of prion proteins in animals OAK

For immunohistochemistry of the prion diseases, several pretreatment methods to enhance the immunoreactivity of human and animal abnormal proteinase-resistant prion protein (PrPSc) on the tissue sections have been employed. The method of 121 degrees C hydrated autoclaving pretreatment or the combination method of 121 degrees C hydrated autoclaving with a certain chemical reagent ( formic acid or proteinase K, etc) are now widely used. We found that an improved hydrated autoclaving method at 135 degrees C, more effectively enhanced PrPSc immunoreactivity for the antibodies recognizing the linear epitope. Effects of autoclaving on the apparent digestibility coefficient of dehulled pea seed meal (Pisum sativum L.) in rainbow trout (Oncorhynchus mykiss W.) Ciencia e Investigación Agraria

The effect of autoclaving on the nutrients' apparent digestibility coefficient (ADC), digestible protein and energy of pea seed meal (P. sativum) fed to Rainbow trout (O. mykiss) was examined. Two samples of the pea meal were autoclaved at 121°C and 1.1 atm for 5 min (5'APM) or 15 min (15'APM), respectively. A third sample, used as control, was not treated (RPM). One reference diet (Basal diet) and 3 experimental diets were elaborated and labelled based on autoclaving time applied to the ingredient (RPM, 5'APM and 15'APM). The four diets were assigned using a completely randomised design, with each treatment having three replicates. 12 tanks were stocked each with 15 trouts with an average weight of 235 ± 10.4 g. Chemical sterilization of culture medium for in vitro multiplication of Cochlospermum regium Ciência Rural

ABSTRACT: Cochlospermum regium roots are used in popular medicine and its extract has diverse phytochemical molecules some with antimicrobial activity, consequently exposing this specie to genetic erosion risks. Thus, the objective of this study was to develop an in vitro multiplication protocol using chemical sterilization of culture medium. Therefore, explants obtained from apical buds of C. regium seedlings were inoculated into with 0.05mg L-1 NAA and 1mg L-1 BAP sterilized by chemical agent sodium hypochlorite (NaOCl) at 0.001%, 0.003% and 0.005% of active chlorine (Cl).
