The genetically transformed roots of red beet have been shown, for the first time, to produce very high levels of peroxidase (POD; EC 1.11.1.7) accounting for 1.21 x 10(6) Units L-1. Of the ten clones established using different strains of Agrobacterium rhizogenes, one was that from the strain LMG-150, three each from A 2/83, A 20/83 and A4. All the clones showed true integration of T-DNA when tested by PCR and Southern hybridization methods. Each clone differed significantly from the others in growth, hormone dependency and POD production where LMG-150 produced highest biomass (140 g FW L-1) as well as POD (ranging from 8000-9000 U g-1 FW and 1.18 x 10(6) U L-1 with a specific activity of 600 U mg-1 protein) on hormone-free medium, both in shake-flask as... |

Auxin and insulin promote Zea mays embryo growth, induce S6 ribosomal protein (S6rp) phosphorylation, and promote specific protein synthesis. The objective of this research was to test a possible cross-talk between insulin and auxin transduction pathways in Z. mays coleoptiles, typical auxin target tissues. Auxin and insulin produced differential quantitative and qualitative stimulation of cytoplasmic and ribosomal protein phosphorylation, and specific patterns of de novo synthesized cytoplasmic proteins. In addition, insulin induced S6rp phosphorylation was strongly inhibited by rapamycin, indicating target of rapamycin (TOR) kinase participation; auxin-induced S6rp phosphorylation was insensitive to this inhibitor. Phosphatidic acid (PA), a second... |

ABSTRACT The salinity has complex effects on the physiological, nutritional and hormonal interactions of plants. This study aimed to evaluate the mitigating action of exogenous application of different types and concentrations of phytohormones on Carica papaya under salt stress. The experiment was conducted in greenhouse, using a randomized block design with treatments formed from a 2 x 3 x 5 factorial scheme, relative to two concentrations of salts in the irrigation water (0.6 and 2.4 dS m-1), three types of phytohormones [Auxin = indole-3-acetic acid; Cytokinin = CPPU (N-(2-chloro-pyridyl)-N-phenylurea) and Gibberellin = gibberellic acid(AG3)], and five concentrations of the phytohormone (0, 12.5, 25.0, 37.5 and 50.0 µM L-1), with three repetitions,... |

Rooting and acclimatization are limiting steps in plant micropropagation, especially in woody plant species. This study aimed to evaluate the IAA and IBA effect on the in vitro rooting and acclimatization of micropropagated shoots of Japanese plum (Prunus salicina Lindl.) cv. América. Shoots from 3 to 4 cm long were inoculated in MS medium with half salt and vitamin concentrations (MS/2) added with IAA and IBA (0, 0.25, 0.5, 0.75 and 1 mg L-1). After a 20-day period in in vitro cultivation, the shoots were evaluated, and then transferred to a greenhouse, and evaluated after 30 days. At the end of the in vitro cultivation period, no significant interactions were observed for number of roots per shoot and rooting percentage, but a significant effect was... |

Abstract: Somatic embryogenesis from explants from hermaphrodite papaya mother plants is an alternative for the production of true-to-type plants without the need for sexing. This study aimed to analyze hormonal and osmotic inducers in different somatic embryogenesis stages in the commercial hermaphrodite hybrid papaya UENF/Caliman 01. Leaf disks from in vitro shoots originated from ex vitro hermaphrodite plants were cultured in induction medium supplemented with different concentrations of 2,4-D (6, 9, 12, 15, and 18 μM) and 4-CPA (19, 22, 25, 28, and 31 μM). After 90 days, the formation of somatic embryos was verified. The 2,4-D induced the formation of light brown calli with low frequency (20%) of somatic embryogenesis. However, 4-CPA (25 μM) induced... |

The aim of the present work was to evaluate the effects of auxins and gibberellins when applied at pre-harvest to the fruit development, and to the ripening and natural fall of the fruit, in 'Pêra' oranges. Trees of Citrus sinensis Osbeck cv. Pêra, 5 years old, were utilized. The treatments applied were: GA3 + 2,4-D 12.5mg L-1 of each; GA3 + 2,4-D 25mg L-1 ; GA3 + 2,4-D 37.5mg L-1; GA3 + NAA 12.5mg L-1; GA3 + NAA 25mg L-1; GA3 + NAA 37.5mg L-1; NAA + 2,4-D 12.5mg L-1; NAA + 2,4-D 25mg L-1; NAA + 2,4-D 37.5mg L-1; and water (control). The treatments were applied 3 times, at intervals of 45 days. The variables evaluated were: rate of natural fall (%), fruit length and diameter (mm), and fresh fruit weight (g). None of the treatments promoved alterations in... |

The effects of various conc of (10-11, 10-9 and 10-7 M) 24-epibrassinolide (24-epiBL) on polyamines and auxins contents was analyzed in 7days old seedlings of Raphanus sativus L. cv. Pusa chetki under copper metal stress. Cu metal (0.2mM) treatment significantly increased endogenous levels of putrescine, spermidine and spermine when compared with control values. Cadaverine content got decreased under metal stress. Endogenous polyamines showed varying levels in seedlings when applying 24-epiBL treatment only. Maximum increase was found in putrescine, cadaverine and spermidine in 10-11, 10-9 and 10-7 M conc of 24-epiBL respectively when applied alone to R. sativus seedlings. However, metal treatment supplemented with 24-epiBL revealed small increase in... |

To define preliminary aspects of walnut (Juglans regia L.) callogenesis in indirect propagation systems, this work analyzed callogenic induction in adult leaves of walnut in broadleaf trees (BTM), Murashige and Skoog (MS) Driver Kuniyuki Walnut (DKW) and Woody Plant (WPM) media. Leaf segments of 1 cm², previously sterilized, were placed in each culture medium, supplemented with 21.4 µM naphtalene acetic acid (NAA) and 8.8 µM 6-benzylaminopurine (BAP). A completely random design was used; the experimental unit was a Petri dish with six callus explants. Each treatment was composed of seven Petri dishes. Callogenesis (%), nodular calli (%) and histology of calli explants were evaluated at 4 weeks by staining 10 µm slices with safranine-fastgreen. There were... |
