Abstract To explore the effect of fruit and vegetable (FV) juice on biomarkers of oxidative damage and antioxidant gene expression in rats, 36 adult male Wistar rats were randomly divided into control, low FV juice dosage or high FV juice dosage treatment groups. The rats were given freshly extracted FV juice or the same volume of saline water daily for five weeks. After intervention, serum and tissues specimens were collected for biomarker and gene expression measurement. FV juice intervention increased total antioxidant capacity, glutathione, vitamin C, β-carotene, total polyphenols, flavonoids levels andglutathione peroxidaseenzyme activity in rat serum or tissues (p < 0.05). FV juice intervention caused reduction of malondialdehyde levels in rat... |

Abstract Machado-Joseph disease (SCA3/MJD) is the most common spinocerebellar ataxia worldwide, and particularly so in Southern Brazil. Due to an expanded polyglutamine at ataxin-3, SCA3/MJD presents a relentless course with no current disease modifying treatment. Clinical scales used to measure SCA3/MJD progression present moderate effect sizes, a major drawback for their use as main outcomes in clinical trials, given the rarity and slow progression of the disease. This limitation might be overcome by finding good surrogate markers. We present here a review of studies on peripheral and neurophysiological markers in SCA3/MJD that can be candidates for state biomarkers. Data on markers already studied were summarized, giving emphasis on validation against... |

Abstract The clinical utility of serum ferritin as a biomarker of disease severity and prognosis in Gaucher disease (GD) is still debated. Here, we aimed to evaluate ferritin and its relation to clinicolaboratory parameters of GD patients seen at the Reference Center for Gaucher Disease of Rio Grande do Sul, Brazil, so as to gather evidence on the utility of ferritin as a biomarker of this condition. A retrospective chart review was performed collecting pre-and posttreatment data from GD patients. Eighteen patients with ferritin levels available before and after treatment were included in the study. Nine of these participants were males, and seventeen had type I GD. All patients were given either enzyme replacement (n = 16) or substrate reduction therapy... |

The growing need to evaluate the quality of aquatic ecosystems led to the development of numerous monitoring tools. Among them, the development of biomarker-based procedures, that combine precocity and relevance, is recommended. However, multi-biomarker approaches are often hard to interpret, and produce results that are not easy to integrate in the environmental policies framework. Integrative index have been developed, and one of the most used is the integrated biomarker response (IBR). However, an analysis of available literature demonstrated that the IBR suffers from a frequent misuse and a bias in its calculation. Then, we propose here a new calculation method based on both a more simple formula and a permutation procedure. Together, these... |

A 3-year survey was made of several biological markers in mussels (Mytilus edulis) exposed in situ to the oil that came ashore after the wreck of the "Erika" tanker on the Brittany (France) coast in December 1999. The mussel response was assessed using a set of 7 biomarkers, most of them related to the metabolism of organic contaminants. After a series of validation tests, data was evaluated for only 5 biomarkers: acety1cholinesterase (AChE), glutathione S-transferase (GST), catalase (CAT), malondialdehyde (MDA) and deoxyribonucleic acid (DNA) adducts. No significant reductions in GST or CAT levels were observed, levels of DNA adducts and MDA were high during the 6 months immediately following the accident and levels of AChE were significantly lower during... |

The aim of this study was to evaluate the seasonal variations of six biomarkers on Mytilus galloprovincialis, using comparison with physiological index and chemical contamination levels on two sites of Mediterranean sea. The seasonal variations of acetylcholinesterase (AChE), DNA adducts, benzo(a)pyrene hydroxylase (BPH), heat-shock proteins (HSP), metallothioneins (MT) and multixeniobiotic proteins (MXR) were estimated over a two-year period, on mussels which were sampled in Carteau (original site) and La Fourcade (transplanted organisms). Carteau, an industrial site, presents higher contamination levels (HAP, PCB, heavy metals) than La Fourcade, an urban area. The PAH profiles underline a strong spatio-temporal variability of levels and sources of... |

A suite of biomarkers was measured in caged mussels at areas impacted by different anthropogenic activities along the Greek coastline to assess biological effects of environmental pollution. Mussels were caged at coastal sites in the vicinity of major cities, in areas influenced by major industries, agricultural practices and in islands away from known sources of pollution. Biomarkers indicative of neurotoxicity (acetylcholinesterase, AchE), oxidative stress (catalase, CAT), phase 11 biotransformation of xenobiotics (glutathione S-transferase, GST), metal exposure (metallothioneins, MTs) and protein synthesis (RNA:DNA ratio) were measured to assess effects of various types of pollutants. AchE activity proved to be the most responsive biomarker with... |

The edifice walls of the Eiffel Tower hydrothermal vent site (Mid-Atlantic Ridge, Lucky Strike vent field) are populated with dense communities of dual symbioses harboring vent mussel Bathymodiolus azoricus, some of which are covered by white filamentous mats belonging to sulfur-oxidizing bacteria. Mussels were collected in both the presence and absence of the filamentous bacteria. A sample of the filamentous bacteria was collected and water measurements of temperature, CH4 and H2S were recorded at the collection area. The whole soft tissues were analyzed for total lipid, carbohydrate and total protein. Metallothioneins and metals (Cu, Fe and Zn) levels were determined in the major organs. The results showed no significant physiological and toxicological... |

In the aim to develop biomarkers of genotoxicity for the biomonitoring of the marine environment, the formation of DNA adducts and oxidized DNA bases was studied for the sentinel organism, Mytilus sp, following exposure to B[a]P via the trophic supply. DNA adducts and 8-oxo-7,8-dihydro-2'-désoxyguanosine (8-oxodGuo) were respectively measured by 32P post-labeling technique and high performance liquid chromatography coupled to electrochemical detection. The developped model of exposure was validated for the realisation of toxicological studies in Mytilus sp. The bioavailability of B[a]P for the exposed organisms and its biotransformation resulted in variations in most of the measured enzymatic biomarkers of exposure. Such an exposure allowed the expression... |

Trophic patterns for Greenland Halibut are reported for the first time in the Canadian Beaufort Sea and Amundsen Gulf (n = 269). Samples were collected from 2012 to 2014 on the upper (300–500 m) and lower continental slope (750–1500 m) and were analyzed for stomach contents, stable isotopes ratios and fatty acids (FA). Stomach contents indicated that Arctic Cod, Boreogadus saida, was the main prey ingested on the upper slope (50–94% of total biomass) whereas Gelatinous Snailfish (Liparis fabricii) and Zoarcids (Lycodes spp.) dominated diets on the lower slope (17–62% of total biomass). Stable isotope mixing models and FA analyses also identified benthopelagic fishes (i.e., Liparis spp., B. saida) as key prey and highlighted large dietary overlap among... |

Determining the lipid content and fatty acid (FA) composition of aquatic organisms has been of major interest in trophic ecology, aquaculture, and nutrition for over half a century. Although protocols for lipid analysis are well-described, their application to aquatic sciences often requires modifications to adapt to field conditions and to sample type. Here, we present the current state of knowledge of methods dedicated to both marine and freshwater lipid analyses, from sampling to data treatment. We review: (i) sample preservation, storage and transport protocols, and their effects on lipids, (ii) lipid extraction, separation of polar and neutral lipids, derivatization, and detection methods, and (iii) available tools for the statistical analysis of FA... |

The Kerguelen Islands include various species of freshwater fish such as brown trout (Salmo trutta). These trout are among the most isolated from direct anthropogenic impact worldwide. This study was designed to analyse cadmium (Cd) and copper (Cu) concentrations in the liver of Kerguelen brown trout, and to assess the possible impacts of these metals on hepatic histopathology and oxidative stress parameters (superoxide dismutase and catalase activity and glutathione levels). Trout were caught in the Chateau River, the Studer Lakes and the Ferme Pond, close to the scientific station of the Kerguelen Islands, corresponding to three morphotypes (river, lake and station). Kerguelen trouts' hepatic concentrations of Cd and Cur were similar to those reported in... |

Biological responses and contaminant levels in biological tissues were investigated in fish specimens of grass goby (Zosterisessor ophiocephalus) collected in spring 2000 in a moderately polluted brackish ecosystem, the Orbetello lagoon (southern coast of Tuscany, Italy). The complexity of harmful effects that contaminants may cause in fish over time was assessed. General condition indices (somatic liver index, and gonadal somatic index), cytochrome P450 enzymes, acetylcholinesterase activity and ovarian morphology were integrated with the content of organochlorines (OCs-hexachlorobenzene, DDTs and polychlorinated biphenyls) in liver and ovaries and p-nonylphenol (NP) and lower ethoxylate (NPE(1-2)) content in muscle and ovaries as well. Samples collected... |

Two deep-sea gastropods, Ifremeria nautilei and Alviniconcha hessleri, collected on a hydrothermal site of the North Fiji Basin (Southwestern Pacific) were analysed for polar and neutral lipids using gas-liquid chromatography/mass spectrometry. A high level of monounsaturated fatty acids (MUFAs) and a low level of omega 3 series polyunsaturated fatty acids (PUFAs) indicated that nutrition of both gastropods was related to a food web based mainly on bacterial supply. From differences in MUFA distribution between the 2 gastropods, it appeared that most of the energy requirements of A. hessleri were supplied by sulfur-oxidizing endobacteria whereas I. nautilei probably had a mixotrophic diet based on endogenous as well as exogenous bacteria. Given the... |
