Production of antibody fragment (Fab) throughout Escherichia coli fed-batch fermentation process: Changes in titre, location and form of product Electron. J. Biotechnol.
Jalalirad,Reza.
Background: Recombinant proteins, including antibodies and antibody fragments, often contain disulfide bond bridges that are necessary for their folding, stability and function. Production of disulfide-bond-containing proteins in the periplasm of Escherichia coli has been very useful, due to unique characteristics of the periplasm, for obtaining fully active and correctly folded products and for alleviating downstream processing. Results: In this study, fed-batch cultivation of Escherichia coli (E. coli) for production of Fab D1.3, which is an anti-hen egg white lysozyme (HEWL) antibody fragment was carried out at 37ºC, and the bacterial cells were induced by adding 0.1 mM IPTG to the culture medium. Fermentor was sampled over the course of fermentation;...

Eletrolite leakage as a technique to diagnose euphorbia heterophylla biotypes resistant to ppo-inhibitors herbicides Planta Daninha
Trezzi,M.M; Vidal,R.A; Kruse,N.D; Gustman,M.S; Xavier,E; Rosin,D; Dedordi,G.F.
The action of herbicides that affect the integrity of cell membranes and cause leakage, like PPO-inhibitors, can be detected by measuring the electric conductivity (EC) of a solution in which the plant tissue target is incubated in the presence of herbicide. The objectives of this work were to confirm PPO resistance in a new Euphorbia heterophylla (EPHHL) biotype, and to compare the electrolyte leakage from R and S to PPO-inhibitors biotypes, using two different methods of incubation in a solution containing herbicides. One experiment was carried in greenhouse and three in laboratory, with a completely randomized design. In the greenhouse experiment, four biotypes of EPHHL were sprayed with seven rates of fomesafen to confirm resistance in suspected...

Concentration determination of extracellular and intracellular red pigments produced by Monascus sp BABT
Hamano,Patricia Sanae; Orozco,Sandra Fernanda Bilbao; Kilikian,Beatriz Vahan.
In this work red pigments production was evaluated (by spectrophotometry) in semi-synthetic medium by rotary shaker cultures, utilizing seven strains of Monascus sp. The strains isolated at LEB/DEQ/EPUSP had a absorbance average of extracellular red pigments higher than the others strains (13.0 and 9.6 U, respectively) and the specific production of extracellular red pigments was from 1.7 until 3.5 times higher than the specific production of intracellular ones.

Enhancement of pigment extraction from B. braunii pretreated using CO2 rapid depressurization BJM
Uquiche,Edgar; Antilaf,Ivette; Millao,Sonia.
Abstract Extraction of compounds from microalgae requires cell disruption as a pretreatment to increase extraction yield. Botryococcus braunii is a microalga with a significant content of carotenoids and other antioxidant compounds, such as chlorophylls. Cell disruption of B. braunii using CO2 rapid depressurization was studied as a pretreatment for the extraction of carotenoid and chlorophyll pigments. We studied the effect of temperature (21–49 °C) and pressure (6–13 MPa) during static compression on pigment recovery with supercritical CO2 at 40 °C, 30 MPa and solvent flow of 4.7 L NPT/min. Within the experimental region, the extraction yield of carotenoids and chlorophylls increased by 2.4- and 2.2-fold respectively. Static compression conditions of...
