This study was designed to micropropagate E. benthamii x E. dunnii, by testing chlorine concentrations for explant asepsis, the optimal concentrations of benzylaminopurine (BAP) and naphthaleneacetic acid (NAA) for bud proliferation, and the ratio between BAP and gibberellic acid (GA3) in two nutrient media for shoot elongation. Nodal segments from H12, H19 and H20 clones were disinfected with 0.5, 1.0, 1.5 and 2.0% (v v-1) of chlorine. Coherent soliton propagation through doped optical fibers: cloning, breakup, and soliton interactions Anais da ABC (AABC)

The simultaneous propagation of two optical pulses through a doped nonlinear dispersive medium modelled by a resonant three-level system was investigated numerically, within the framework of a pair of coupled extended nonlinear Schrödinger equations. These included the contribution of the dopant resonances whose dynamics is governed by Bloch equations. In this work, we review the interesting possibilities on the manipulation of fields such as cloning, breakup and soliton interactions, that the combination of coherent population trapping with nonlinear dispersive media offers.

cDNA cloning, Phylogenic Analysis and Gene Expression Pattern of Phenylalanine ammonia-lyase in Sugarcane (Saccharum officinarum L.) BABT

The aim of the present study was to clone and characterize a full length cDNA of sugarcane (Saccharum officinarum) phenylalanine ammonia-lyase (SoPAL). Differential tissue expression pattern of the SoPAL transcript and its enzyme activity was also analyzed during the tillering stage of growth. The full-length of SoPAL cDNA was 2118 bp long and contained a protein with 706 amino acids, determined by encoding technique. The amino acid sequence and phylogenic analysis of the cloned SoPAL showed high similarity to PAL from other monocotyledonous such as sorghum (96%), maize (93%) and Bamboos (87.12%). Molecular Cloning and Sequencing of AlkalophilicCellulosimicrobium cellulans CKMX1 Xylanase Gene Isolated from Mushroom Compost and Characterization of the Gene Product BABT

ABSTRACT A xylanolytic bacterium was isolated from mushroom compost by using enrichment technique. Results from the metabolic fingerprinting, whole-cell fatty acids methyl ester analysis and 16S rDNA sequencing suggested the bacterium to be Cellulosimicrobium cellulans CKMX1. Due to the xylanolytic activity of this bacterium, isolation and characterization of the xylanase gene were attempted. A distinct fragment of about 1671 bp was successfully amplified using PCR and cloned into Escherichia coli DH5α. A BLAST search confirmed that the DNA sequence from the amplified fragment was endo-1, 4-beta-xylanase, which was a member of glycoside hydrolase family 11. Three recombinant antigens of Treponema pallidum Nichols strain were fused with GST, cloned and expressed in Escherichia coli, resulting in high levels of GST-rTp47 and GST-rTp17 expression, and supplementation with arginine tRNA for the AGR codon was needed to obtain GST-rTp15 overexpression. Purified fusion protein yields were 1.9, 1.7 and 5.3 mg/l of cell culture for GST-rTp47, GST-rTp17 and GST-rTp15, respectively. Our objective was to clone, express and characterize adult Dermatophagoides farinae group 1 (Der f 1) allergens to further produce recombinant allergens for future clinical applications in order to eliminate side reactions from crude extracts of mites. Based on GenBank data, we designed primers and amplified the cDNA fragment coding for Der f 1 by nested-PCR. After purification and recovery, the cDNA fragment was cloned into the pMD19-T vector. The present study examined the in vitro and in vivo development of bovine nuclear-transferred embryos. A bovine fetal fibroblast culture was established and used as nucleus donor. Slaughterhouse oocytes were matured in vitro for 18 h before enucleation. Enucleated oocytes were fused with fetal fibroblasts with an electric stimulus and treated with cytochalasin D and cycloheximide for 1 h followed by cycloheximide alone for 4 h. Reconstructed embryos were cultured for 7-9 days and those which developed to blastocysts were transferred to recipient cows. Entomopathogenic fungus Verticillium lecanii is a promising whitefly and aphid control agent. Chitinases secreted by this insect pathogen have considerable importance in the biological control of some insect pests. An endochitinase gene Vlchit1 from the fungus was cloned and overexpressed in Escherichia coli. The Vlchit1 gene not only contains an open reading frame (ORF) which encodes a protein of 423 amino acids (aa), but also is interrupted by three short introns. A homology modelling of Vlchit1 protein showed that the chitinase Vlchit1 has a (α/β)8 TIM barrel structure. A total of 107 rhizobacterial isolates, obtained from the rhizosphere of eucalypt clones were tested as rooting inducers of cuttings and mini-cuttings planted in substrate composed of carbonized rice husk and vermiculite (1:1). Cuttings and mini-cuttings were planted in conical plastic tubes containing treated and untreated (control) substrate and kept under intermittent mist irrigation at 26-28ºC. After 35 days, rooting percentage and dry root matter of cuttings were evaluated. Ten isolates capable of providing gains of up to 110% in root formation and up to 250% in root biomass over non-inoculated control cuttings were selected. In order to find out the status of magnetotactic bacteria at three different sediment in Czech Republic, samples collected from both standing and running freshwaters were subjected to molecular diversity analysis by using 16S rRNA gene approach. En 1993 se reportó por primera vez en México a Candidatus Liberibacter solanacearum, como el agente causal de la pigmentación morada en las hojas, reducción de tamaño de la planta y necrosis interna de los tubérculos de la papa; posteriormente se detectó este bacterium en los cultivos de tomate, chile y berenjena. Sin embargo, la distribución de las plantas sintomáticas en el campo no tenía un patrón definido hasta que en 2009 se observó la totalidad de plantas atípicas de chile jalapeño cv Ixtapa en dos campos de los estados de Nayarit y Jalisco. Chickpea, a lesser-studied grain legume, is being investigated due to its taxonomic proximity with the model legume genome Medicago truncatula and its ability to endure and grow in relatively low soil water contents making it a model legume crop for the study of agronomic response to drought stress. Aquaculture still faces serious economic impacts due to the loss of animals to disease. A conservative estimate of 5% losses due to disease means that the finfish aquaculture industry loses over $1 billion annually on a global scale. One proven way to prevent costly disease outbreaks is to vaccinate fish against common or known pathogens. Current vaccination schemes still result in losses, however, and this may be due in part to vaccine design. Vaccines are currently designed using state of the art knowledge of immune responses, which is based primarily on mammalian studies.
