Zinc detoxification by a cyanobacterium from a metal contaminated bay in Brazil BABT
Andrade,Luciana; Keim,Carolina N.; Farina,Marcos; Pfeiffer,Wolfgang C..
We describe here the trapping of zinc in polyphosphate granules of the cyanobacterium Synechocystis aquatilis NPBS-3. Cells were cultured in 25 µM of zinc chloride and prepared for electron microscopy and energy-dispersive X-ray analysis. Some ultrastructural features were changed by zinc exposure, the increase of glycogen granules number being the main change. The polyphosphate granules contained phosphorus, sulphur, calcium, iron and zinc. The trapping of zinc in polyphosphate granules seemed to be an effective way of detoxifying the metal and surviving in the bay. As a non-specific mechanism, these polyphosphate granules could also be effective in trapping other metals in excess.

Degradation and detoxification of three textile Azo dyes by mixed fungal cultures from semi-arid region of Brazilian Northeast BABT
Nascimento,Carlos; Magalhães,Danielly de Paiva; Brandão,Martha; Santos,André Batouli; Chame,Marcia; Baptista,Darcílio; Nishikawa,Marília; Silva,Manuela da.
The aim of this work was to study the degradation and detoxification of three textile azo dyes (Reactive Red 198, Reactive Red 141 and Reactive Blue 214) by mixed fungal cultures from semi-arid region of Brazilian Northeast. Sediment samples of twenty water reservoirs in the surroundings of Serra da Capivara National Park, area of environmental preservation in the caatinga in the State of Piauí, with semi-arid climate, were evaluated in order to select the consortia of fungi capable to degrade and detoxify these dyes. The mixed fungal culture from Caldeirão Escuridão (CE) reservoir was the most efficient in the degradation and detoxification of the dyes tested.

Decolorization of different textile dyes by Penicillium simplicissimum and toxicity evaluation after fungal treatment BJM
Bergsten-Torralba,L.R.; Nishikawa,M.M.; Baptista,D.F.; Magalhães,D.P.; Silva,M. da.
The objective of this study was to investigate the capacity of decolorization and detoxification of the textile dyes Reactive Red 198 (RR198), Reactive Blue 214 (RB214), Reactive Blue 21 (RB21) and the mixture of the three dyes (MXD) by Penicillium simplicissimum INCQS 40211. The dye RB21, a phthalocyanine, was totally decolorized in 2 days, and the others, the monoazo RR198, the diazo RB214 and MXD were decolorized after 7 days by P. simplicissimum. Initially the dye decolorization involved dye adsorption by the biomass followed by degradation. The acute toxicity after fungal treatment was monitored with the microcrustacean Daphnia pulex and measured through Effective Concentration 50% (EC50). P. simplicissimum reduced efficiently the toxicity of RB21...

Effect of caffeine and tannins on cultivation and fructification of Pleurotus on coffee husks BJM
Fan,Leifa; Soccol,Andréa Thomaz; Pandey,Ashok; Vandenberghe,Luciana Porto de Souza; Soccol,Carlos Ricardo.
The objective of this work was to study the effect of caffeine and tannins on Pleurotus sp. cultivation and to evaluate the feasibility of using coffee husks as substrate for mushroom cultivation. Eight strains of P. ostreatus and two strains of P. sajor-caju were screened on a medium prepared from agar extract of coffee husk. Based on best mycelial growth and biomass production, the strain P. ostreatus LPB 09 was selected for detailed studies. With the increase of caffeine concentration, the mycelial growth and the biomass production decreased, and no growth was observed when concentration of caffeine was 2500 mg/L. Furthermore, Pleurotus did not degrade the caffeine, but absorbed it. Tannin under 100 mg/L in the medium stimulated the growth of mycelia,...

Copper in plants Braz. J. Plant Physiol.
Yruela,Inmaculada.
Copper is an essential metal for normal plant growth and development, although it is also potentially toxic. Copper participates in numerous physiological processes and is an essential cofactor for many metalloproteins, however, problems arise when excess copper is present in cells. Excess copper inhibits plant growth and impairs important cellular processes (i.e., photosynthetic electron transport). Since copper is both an essential cofactor and a toxic element, involving a complex network of metal trafficking pathways, different strategies have evolved in plants to appropriately regulate its homeostasis as a function of the environmental copper level. Such strategies must prevent accumulation of the metal in the freely reactive form (metal detoxification...

Lead toxicity in plants Braz. J. Plant Physiol.
Sharma,Pallavi; Dubey,Rama Shanker.
Contamination of soils by heavy metals is of widespread occurrence as a result of human, agricultural and industrial activities. Among heavy metals, lead is a potential pollutant that readily accumulates in soils and sediments. Although lead is not an essential element for plants, it gets easily absorbed and accumulated in different plant parts. Uptake of Pb in plants is regulated by pH, particle size and cation exchange capacity of the soils as well as by root exudation and other physico-chemical parameters. Excess Pb causes a number of toxicity symptoms in plants e.g. stunted growth, chlorosis and blackening of root system. Pb inhibits photosynthesis, upsets mineral nutrition and water balance, changes hormonal status and affects membrane structure and...

De novo transcriptome assembly of the lobster cockroach Nauphoeta cinerea (Blaberidae) Genet. Mol. Biol.
Segatto,Ana Lúcia Anversa; Diesel,José Francisco; Loreto,Elgion Lucio Silva; da Rocha,João Batista Teixeira.
Abstract The use of Drosophila as a scientific model is well established, but the use of cockroaches as experimental organisms has been increasing, mainly in toxicology research. Nauphoeta cinerea is one of the species that has been studied, and among its advantages is its easy laboratory maintenance. However, a limited amount of genetic data about N. cinerea is available, impeding gene identification and expression analyses, genetic manipulation, and a deeper understanding of its functional biology. Here we describe the N. cinerea fat body and head transcriptome, in order to provide a database of genetic sequences to better understand the metabolic role of these tissues, and describe detoxification and stress response genes. After removing low-quality...

Detoxification of Pacific oyster Crassostrea gigas fed on diets of Skeletonema costatum with and without silt, following PSP contamination by Alexandrium minutum ArchiMer
Gueguen, Marielle; Bardouil, Michele; Baron, Regis; Lassus, Patrick; Truquet, Philippe; Massardier, Julie; Amzil, Zouher.
Contamination of shellfish by paralytic shellfish poisoning ( PSP) toxins poses an economic threat to shellfish farmers. As contaminated shellfish cannot be harvested for long periods of time, it would be very useful to develop processes to optimise and shorten their detoxification. In this study, Pacific oysters Crassostrea gigas were first experimentally contaminated over a period of 13 days with a continuous flow of toxic Alexandrium minutum cultures at concentrations ranging from 150 to 200 cell ml(-1) ( toxin content after 13 days of contamination 438 mu g STX equiv. 100 g(-1) wet weight). Then, two different detoxification treatments were tested and showed detoxification rates greater than those observed in coastal environments. The first treatment...

Variation of heavy metal concentrations (Ag, Cd, Co, Cu, Fe, Pb, V, and Zn) during the life cycle of the common cuttlefish Sepia officinalis ArchiMer
Miramand, Pierre; Bustamante, Paco; Bentley, Daniel; Koueta, Noussithe.
The developmental changes in the concentration of 8 essential and non-essential heavy metals (Ag, Cd, Cu, Co, Fe, Pb, V, Zn) in the tissues (digestive gland, cuttlebone and whole animal) of the common cuttlefish Sepia officinalis collected in the bay of the river Seine were monitored from the end of the embryogenesis until the adult reproductive stage. Compared to embryos, juveniles after hatching displayed much higher concentrations of Ag, Cu, Fe and Zn, suggesting an efficient incorporation from seawater. Conversely, the amounts of Cd, Pb and V in hatchlings remained constant suggesting that these metals are barely bioavailable for juveniles. Once the juveniles start to feed, the digestive gland appears to play a major role in the storage of all metals....

Subcellular distribution of Ag, Cd, Co, Cu, Fe, Mn, Ph, and Zn in the digestive gland of the common cuttlefish Sepia officinalis ArchiMer
Bustamante, Paco; Bertrand, M; Boucaud Camou, E; Miramand, Pierre.
The subcellular fractionation of the digestive gland cells of the common cuttlefish Sepia officinalis was performed to investigate the distribution of metals between organelles and cytosol and the different cytosolic fractions separated by gel-filtration chromatography. Total metal concentrations vary over 3 orders of magnitude, ranging from dwt for Pb to dwt for Zn. With the exception of Cd, Co, and Cu, metals were mostly bound to the organelles. Whereas no specific organelle compartment was found for Mn, Ph, and Zn, Fe was mainly associated with nucleus, brown body, and "boule" fraction (i.e., 52%) and 44% of the total Ag was contained in the lysosomal and mitochondria enriched fraction. The link of metals with hydrosoluble proteins in the cytosolic...

Modélisation de la détoxication de mollusques bivalves contenant des phycotoxines paralysantes ou diarrhéiques ArchiMer
Gueguen, Marielle.
The purpose of this study is to implement models to describe detoxification kinetics of Pacific oysters Crassostrea gigas and Blue mussels Mytilus edulis containing paralytic and diarrheic phycotoxins respectively. First, experimental trials made it possible to complement data from the literature regarding the understanding of processes involved in mollusc detoxification. In particular, they pointed out a difference in the mollusc capacity to degrade cellwalls of phycotoxin-producing dinoflagellates, according to their en cystement capacities. It was observed that paralytic phycotoxins reached the various oyster tissues whereas diarrheic toxins were mainly sequestred in the digestive gland of mussels. The latter seem to be retained by lysosomes localized...

Improving detoxification Efficiency of PSP-contaminated oysters (Crassostrea gigas Thunberg) ArchiMer
Lassus, Patrick; Bardouil, Michele; Baron, Regis; Berard, Jean-baptiste; Masselin, Pierre; Truquet, Philippe; Pitrat, Jp.
Shellfish production, and production of marine bivalve molluscs in particular, is the main form of marine aquaculture in the European Union, with a production estimated at 1,200,000 tons in 1999 (Food and Agricultural Organization). Moreover, aquaculture production is rapidly expanding throughout Europe and is becoming a major economic resource in developing areas such as Northern Greece, Ireland, and northwestern Spain. These shellfish, mainly mussels, oysters, and clams, are produced in a natural environment. Due to variations in their environment, these otherwise edible species may however become unsafe for human consumption. Potential problems are amplified due to the nature of shellfish themselves: as they are filter feeders, they consume large...

Genotoxicity and activation of cellular defenses in transplanted zebra mussels Dreissena polymorpha along the Seine river ArchiMer
Chatel, Annelle; Faucet-marquis, Virginie; Gourlay-france, Catherine; Pfohl-leszkowicz, Annie; Hubert, Francoise.
The aim of the present study was to confirm the relevance of studying DNA adduct formation in a field study. In that context, freshwater mussels Dreissena polymorpha, collected in a reference station, were transplanted in different sites with a pollution gradient. After one and two months, mussels were collected and DNA adduct formation was analyzed using the 32P post labelling technique on both gills and digestive glands. In addition, the expression of genes involved in the detoxification system (catalase (CAT), superoxide dismutase (SOD), glutathione S-transferase (GST), HSP70, aryl hydrocarbon receptor (AHR), P glycoprotein (PgP), metallothionein (MT)) was assessed by RT-PCR. DNA adducts were observed at amount comparable to data from literature....

Detoxification enhancement in the gymnodimine-contaminated grooved carpet shell, Ruditapes decussatus (Linne) ArchiMer
Medhioub, Walid; Gueguen, Marielle; Lassus, Patrick; Bardouil, Michele; Truquet, Philippe; Sibat, Manoella; Nejib, Medhioub; Philippe, Soudant; Mejdeddine, Kraiem; Amzil, Zouher.
In the Gulf of Gabes (Tunisia, Eastern Mediterranean sea), the grooved carpet shell Ruditapes decussatus has been seen to contain persistent levels of gymnodimine (GYM) for several years. The present experimental work represents the first attempt to assess detoxification kinetics of fast-acting toxins (FAT) in marine molluscs fed specific diets of non-toxic algae (Isochrysis galbana). To find an optimal detoxification method, two experiments were performed in which clams were first fed the toxic dinoflagellate Karenia selliformis to artificially contaminate them with GYM, thus simulating the effect of natural toxic episodes. As a second step, the same clams were fed a non-toxic algae, I. galbana. to speed up the detoxification process. Changes in toxin...

Technological adaptation to harmful algal blooms : socioeconomic consequences for the shellfish farming sector in Bourgneuf Bay (France) ArchiMer
Perez Agundez, José A.; Raux, Pascal; Girard, Sophie; Mongruel, Remi.
The economic impacts of harmful algal blooms (HABs) on the shellfish farming sector depend on their frequency, duration and intensity. Safeguarding storage and accelerated detoxification are technical solutions that could mitigate the effects of these events. This article first analyzes the economic feasibility of the technological changes that can be adopted by the shellfish farming sector in France. It then examines their associated social impacts. Finally, an application is carried out on the Bourgneuf Bay production area. The economic analysis addresses three issues related to HABs: (1) the economic performance of the shellfish farms in Bourgneuf Bay, (2) the costs of these new technologies, and (3) the economic viability of such an investment. Results...

Bioaccumulation of trace elements in pelagic fish from the Western Indian Ocean ArchiMer
Kojadinovic, Jessica; Potier, Michel; Le Corre, Matthieu; Cosson, Richard P.; Bustamante, Paco.
Trace elements were analyzed in fish of commercial interest to determine their importance in marine systems of the Western Indian Ocean and their bioaccumulation patterns. The results are equivalent or lower than levels reported in ichthyofauna worldwide. Certain values of muscular Cd, Hg, Pb and Zn were, however, above thresholds for human consumption. Levels varied among tissues, species and fish length, but were seldom influenced by the nutritional condition of the fish, its gender and its reproductive status. Correlations between hepatic Hg and Se levels in Swordfish (r(2) = 0.747) and Yellowfin Tunas (r(2) = 0.226), and among metallothionein linking metals imply the existence of detoxification processes in these species. Level differences between fish...

The photodynamic action of UV sensitized methylene blue on the venom of Thailand cobra Naja siamensis J. Venom. Anim. Toxins
GAWADE,S. P..
The photodynamic action of UV sensitized methylene blue (MB) was studied on the venom of Thailand cobra Naja siamensis. The reaction mixture comprising phosphate buffer (B), methylene blue (S), and venom protein (P) was exposed to UVR (ultra violet radiation) for 2 hours at 37°C. Stable excited species (S’) were generated by photodynamic action. These in B environment produced reduced substates SH2 and photooxidised products PO in the presence of atmospheric oxygen. Increased absorbance (OD) of the reaction mixture after UVR with a corresponding loss of protein was suggested to be due to the formation of photooxidised venom products. The detoxification profile of venom protein using visible radiation in the presence of MB and oxygen appeared similar to the...
