Patogenicidad de Metarhizium anisopliae (Metch.) Sorokin y Beauveria bassiana (Bals.) Vuill sobre Periplaneta americana (L.) y su sinergismo con el ácido bórico Colegio de Postgraduados

Los hongos entomopatógenos Beauveria bassiana (Balsamo) Vuillemin 1912 y Metarhizium anisopliae (Metschnikoff) Sorokin 1976 fueron aplicados sobre ninfas y adultos de la cucaracha Periplaneta americana (Linnaeus, 1758) en condiciones de laboratorio (1.1x106 conidia, temperatura 24 ±2°C y humedad relativa, HR, 50±10%). Los aislamientos probados mostraron bajo potencial patogénico y porcentajes de mortalidad significativamente diferentes entre aislamientos. Además se observó menor mortalidad en adultos que en ninfas, en las que se presentaron cambios de conducta a la aplicación de la suspensión conidial. Genetic diversity of Hirsutella thompsonii isolates from Thailand based on AFLP analysis and partial beta-tubulin gene sequences Genet. Mol. Biol.

Amplified fragment length polymorphism (AFLP) was used to investigate polymorphism among 43 Hirsutella thompsonii isolates (33 from Thailand) obtained from various mite species. The outgroups were an unidentified Hirsutella isolate along with Hirsutella nodulosa and Hirsutella kirchneri. Phylogenetic analyses of the AFLP data showed significant variation among isolates and the existence of three H. thompsonii clades. Selection of culture media and in vitro assessment of temperature-dependent development of Nomuraea rileyi Neotropical Entomology

In vitro development - radial growth and sporulation - of Nomuraea rileyi isolates from Anticarsia gemmatalis Hübner and Plusiinae larvae was studied on four culture media: Sabouraud, maltose, agar and yeast - SMAY; SMAY plus rice extract - SMAYR; complete medium for N. rileyi - CMNr and maltose agar yeast with potato extract - MAYP. Their development on a selected medium (MAYP) was analysed at five temperatures. Two mathematical models were fitted to in vitro and in vivo temperature dependent radial growth rates and thermal requirements were estimated. A bioassay method that uses melon leaves as substrate for eggs and nymphs of the biotype B of Bemisia tabaci (Genn.) was established. Root formation of petioles immersed in tap water guaranteed the turgidity and normal coloration of leaves for a period of 20 to 25 days, enabling the execution of tests with entomopathogenic fungi. The in vitro compatibility of the entomopathogenic fungus Beauveria bassiana (Bals.) Vuil. with 12 acaricide formulations was evaluated. The active ingredients Clorfenapyr, Fenpyroximate, Amitraz, Acrinathrin, Hexythiazox, Abamectin, Pyridine, Dimethoate, Pyridaphethion, Fenbutatin Oxide, Azocyclotin and Cyhexatin were tested in three different concentrations (mean concentration - MC, half MC and twice the MC). Action of pesticides to Metarhizium anisopliae in soil Neotropical Entomology

The present study aimed to analyze the action of some acaricides, fungicides, insecticides and herbicides containing different active ingredients on Metarhizium anisopliae Metsch. (Sorokin) inoculated into autoclaved soil. The action of the pesticides was evaluated based on the fungal respiratory activity. The first assessment was done at 48h after inoculation. The pesticides were then added and respiratory activity was determined nine times every 48h and an additional five times every 4 days. The growth and autolysis of two strains of the entomopathogenic deuteromycete fungus Metarhizium anisopliae var. anisopliae were evaluated in medium containing casein or glucose as carbon source. Parameters such as economic coefficient and degree of autolysis were determined for each strain. Protease production was determined throughout the growth and autolysis phases of the cultures on medium under conditions of protease induction (in the presence of casein as sole source of carbon and nitrogen). Differentiation of the entomopathogenic fungus Metarhizium flavoviride (Hyphomycetes) Rev. Microbiol.

The differentiation of a Brazilian isolate of Metarhizium flavoviride (CG 423), a promising candidate for the biocontrol of grasshoppers, was investigated. Conidia were spread onto solid medium (1% yeast extract, 2.8% agar, 96.2% distilled water), incubated at 28°C and observed during 26 h. Germination initiated as conidia size increased from 5.3 (±0.6) x 3.1 (±0.3) µm (0 h incubation) to 8.1 (±0.2) x 6.1 (±0.2) µm (8 h incubation). Selection of Beauveria bassiana and Metarhizium anisopliae isolates for the control of Blissus antillus (Hemiptera: Lygaeidae) Scientia Agricola

Chinch bugs of the genus Blissus (Hemiptera: Lygaeidae) are serious pests of maize, wheat, sorghum and various grasses. This study is the first to screen isolates of Beauveria bassiana and Metarhizium anisopliae against 4th instar and adult Blissus antillus aiming to develop a biological control program for this important pasture pest. Ten fungal isolates were initially screened and three isolates were chosen for further investigation. To determine virulence, insects were inoculated by immersion in concentrations of 5 ´ 10(8) conidia mL-1. Mortality was evaluated for 10 days. Molecular characterization of Paecilomyces fumosoroseus (Deuteromycotina: Hyphomycetes) isolates Scientia Agricola

ITS and RAPD analyses were used to investigate molecular variations within samples of Paecilomyces isolates and to resolve five morphologically atypical isolates resembling P. fumosorosus, obtained from whitefly in Northern Paraná State. The ITS4-ITS5 amplicon was 700 base pairs (bp) long in all isolates. The five isolates of Paecilomyces not assigned to species produced restriction profiles identical to all the reference strains of P. fumosoroseus. The extent of fingerprint variability observed by RAPD was sufficient to discriminate all the isolates. This study investigated whether populations of naturally-occurring soil bacteria, fungi and actinomycetes influence the effect of fungicides on the survival and growth of Beauveria bassiana. The toxicity of methyl thiophanate, pyraclostrobin, mancozeb and copper oxychloride at the recommended doses was analyzed in culture medium and in soil inoculated with fungus at various time points after addition of fungicides. All fungicides completely inhibited the growth and sporulation of B. bassiana in the culture medium. Foliage consumption by nymphs and adults of the grasshopper Rhammatocerus schistocercoides Rehn infected with the fungus Metarhizium flavoviride Gams & Rozsypal was evaluated. Sixth and eighth instar nymphs and female adults consumed on average (±SEM) 6.9 ± 0.36 cm² (60.7 ± 2.92 mg of dry matter), 11.0 ± 1.35 cm² (74.9 ± 9.24 mg d.m.), and 16.0 ± 0.50 cm² (126.5 ± 6.35 mg d.m.) of sugarcane leaves per day, equivalent to 85.4, 35.9 and 41.9% of the insect body weights, respectively. The genetic variability of 27 individuals from two distinct populations (Northeast and Central Brazil) of the grasshopper Schistocerca pallens (Thunberg) and eight isolates of its natural pathogen, the fungus Metarhizium flavoviride Gams & Rozsypal, was investigated using RAPD analysis. For the grasshoppers, ten different 10-mer oligonucleotide primers of arbitrary sequence were selected for analysis, resulting in 79 scorable binary characters.
