Background: Many buildings in Egypt e.g. museums, mosques and churches, do not possess controlled environments for minimizing the risks of damage of wooden artifacts due to the growth of fungi. Fungal damage usually appears as change in wood color, appearance of stains, and sometimes deformation of wooden surfaces. In this study we focused on the effect that some fungi exert on the properties of wooden artifacts and evaluated the effectiveness of different concentrations of chitosan on their protection against damage by mold fungi. An analysis of the spontaneous mutation rate measurement in filamentous fungi Genet. Mol. Biol.

Mutations related to gene methG1 of Aspergillus nidulans were analyzed, in order to study a mathematical model for the determination of the mutation rate per nucleus per generation, in filamentous fungi. A replica plating technique was used to inoculate, in a single operation, 26 colonies of the strain, into Petri dishes containing culture medium, and the nine central colonies were analyzed for size and number of conidia in each colony. Using this technique, several central colonies were analyzed with regard to the appearance of mutation, and the number and type of reversions were determined for each colony. Structural organization of polygalacturonase-encoding genes from Penicillium griseoroseum Genet. Mol. Biol.

The pectinolytic system of Penicillium griseoroseum has been studied as a model to investigate aspects of gene organization in filamentous fungi. Here we show that the endopolygalacturonase-coding genes previously isolated exist as single copies in the fungus genome. DNA blot analysis revealed the presence of corresponding genes in other Penicillium species, although only one or two genes were found in opposition to the endoPG gene family reported for other filamentous fungi. The biology and potential for genetic research of transposable elements in filamentous fungi Genet. Mol. Biol.

Recently many transposable elements have been identified and characterized in filamentous fungi, especially in species of agricultural, biotechnological and medical interest. Similar to the elements found in other eukaryotes, fungal transposons can be classified as class I elements (retrotransposons) that use RNA and reverse transcriptase and class II elements (DNA transposons) that use DNA. The changes (transposition and recombination) caused by transposons can supply wide-ranging genetic variation, especially for species that do not have a sexual phase. The application of transposable elements to gene isolation and population analysis is an important tool for molecular biology and studies of fungal evolution.

The objective of this study was to evaluate the effect of bacterial inoculants at two inoculation rates on chemical and biological characteristics of maize silage. The treatments consisted of two inoculating rates (5 and 6 log cfu g-1of forage) for each strain of lactic acid bacteria (LAB) identified as Lactobacillus buchneri, L. hilgardii, or L. plantarum. Intra and extracellular nuclease production by Aspergillus niger and Aspergillus nidulans Rev. Microbiol.

Intra and extracellular nuclease production by strains of Aspergillus niger and Aspergillus nidulans was estimated using a modified DNAse test agar and cell-free extract assays. Differences in the production of nucleases by A. niger and A. nidulans were observed. These observations suggest that the DNAse test agar can be helpful for a quick screening for some types of nucleases in filamentous fungi. The assays using cell-free extracts can also be useful for initial characterization of other types of nucleases.

Pathogenicity characteristics of filamentous fungi strains isolated from processed oat Rev. Microbiol.

Nineteen strains of filamentous fungi isolated from processed oat were tested for pathogenicity factors, based on three parameters: growth at 37oC, production of phospholipase and urease. Aspergillus niveus, Oidiodendron gryseum and Sporothrix cyanescens were positive for the three parameters. The other species were positive only for one or two of them.

Phosphate-solubilizing fungi isolated from a semiarid area cultivated with melon (Cucumis melo L. cv. gold mine) Acta Botanica

Considering that little is known about the occurrence of phosphate-solubilizing fungi from areas cultivated with melon, the phosphate solubilization ability of filamentous fungi isolated in these areas was evaluated. Three hundred and eighteen filamentous fungal isolates belonging to 23 genera were evaluated, besides Aphyllophorales and Mycelia sterilia. From those, 52 were able to solubilize P: Aphyllophorales (2), Aspergillus (34), Penicillium (10) and Rhizopus (6). These results will contribute to subsidizing further research regarding the capacity of these fungi to solubilize other sources of phosphate applied to the melon crop, as well as indicate the need for a screening program to select those with higher capacity and potential for solubilization.

Production of xylanases by an Aspergillus niger strain in wastes grain Biological Sciences

Many fungi are used in order to extract products from their metabolism through bioprocesses capable of minimizing adverse effects caused by agro-industrial wastes in the environment. The aim of this study was to evaluate the xylanase production by an Aspergillus niger strain, using agro-industrial wastes as substrate. Brewer’s spent grain was the best inducer of xylanase activity. Higher levels of xylanase were obtained when the fungus was grown in liquid Vogel medium, pH 5.0, at 30ºC, during 5 days. The temperature for optimum activity was 50ºC and optimum pH 5.0. The enzyme was stable at 50ºC, with a half-life of 240 min. High pH stability was verified from pH 4.5 to 7.0. The treatment of acute lymphoblastic leukemia is challenging due to side effects, efficacy of the available drugs, and costs. Utilization of L-asparaginase as a therapeutic agent is essential to increase survival of patients. However, costs are elevated and the bacterial forms of the enzyme cause reactions that result in its inhibition by the immune system. Such biodiversity is barely known, making room for a great potential still to be discovered, especially in tropical environments - which are favorable to growth and species variety. Concentration determination of extracellular and intracellular red pigments produced by Monascus sp BABT

In this work red pigments production was evaluated (by spectrophotometry) in semi-synthetic medium by rotary shaker cultures, utilizing seven strains of Monascus sp. The strains isolated at LEB/DEQ/EPUSP had a absorbance average of extracellular red pigments higher than the others strains (13.0 and 9.6 U, respectively) and the specific production of extracellular red pigments was from 1.7 until 3.5 times higher than the specific production of intracellular ones.

Selection of cellulolytic fungi isolated from diverse substrates BABT

The aim of the present work was to select filamentous fungi isolated from diverse substrates to obtain the strains with potential to produce the hydrolytic enzymes. From a total of 215 strains, seven strains from the soils, six from the plants and one from sugarcane bagasse were selected and identified as belonging to the Trichoderma, Penicillium and Aspergillus genera. The best hydrolytic activities obtained by semi-solid fermentation using these strains were approximately: 35; 1; 160; 170 and 120 U/gdm (CMCase, FPase, β-glucosidase, xylanase and polygalacturonase, respectively), demonstrating their potential to synthesize the enzymes compared with the results reported in the literature.

Filamentous fungi isolated from Brazilian semiarid tolerant to metallurgical industry wastes: an ex situ evaluation BABT

The purpose of this study was to assess the impact of metallurgical industry wastes on the semiarid soil microbiota using physico-chemical and microbiological parameters, highlighting the filamentous fungi assembly. Soil samples were collected in an area of industrial waste deposit contaminated with lead and mixed with natural soil (control soil) in seven different concentrations (0, 7.5, 15, 30, 45, 60 and 100%). Yeasts and filamentous fungi in bottled mineral water and tap water from municipal supplies BABT

The main objective of this study was to analyse the occurrence of yeasts and filamentous fungi in drinking water as well as to investigate their correlation with the indicator bacteria of faecal pollution. Yeasts were detected in 36.6% and 11.6% of the bottled mineral on water dispensers and tap water samples from municipal system, respectively. Twenty-one (35.0%) of bottled mineral water and two (3.3%) of tap water samples were positive for filamentous fungi.
