Lack of significant long-term effect of dietary carbohydrates on hepatic glucose-6-phosphatase expression in rainbow trout (Oncorhynchus mykiss) ArchiMer
Panserat, Stéphane; Medale, Francoise; Breque, Jeanine; Plagnes Juan, E; Kaushik, Sadasivam.
Hepatic glucose-6-phosphatase (G6Pase) plays an important role in glucose metabolism because it catalyzes the release of glucose to the circulatory system in the processes of glycogenolysis and gluconeogenesis. The present study was initiated to analyze the regulation of hepatic G6Pase expression by dietary carbohydrates in rainbow trout. The first step in our study was the identification of a partial G6Pase cDNA in rainbow trout that was highly homologous to that of mammals. Hepatic G6Pase activities and mRNA levels were measured in trout fed one of the experimental diets, with or without carbohydrates. We found no significant effect of intake of dietary carbohydrates on G6Pase Expression (mRNA and activity) 6 hours and 24 hours after feeding. These...

Molecular cloning, tissue distribution and sequence analysis of complete glucokinase cDNAs from gilthead seabream (Sparus aurata), rainbow trout (Oncorhynchus mykiss) and common carp (Cyprinus carpio) ArchiMer
Panserat, Stéphane; Blin, C; Medale, Francoise; Plagnes Juan, E; Breque, Jeanine; Krishnamoorthy, J; Kaushik, Sadasivam.
The enzyme glucokinase (GK) (EC 2.7.1.1) plays an important role in the control of glucose homeostasis. Qualitative and/or quantitative variations in GK enzyme have been postulated by previous studies to explain why dietary carbohydrate utilisation is lower in gilthead seabream (Sparus aurata) and rainbow trout (Oncorhynchus mykiss) than in common carp (Cyprinus carpio). In this study, we report the isolation and characterisation of a full-length cDNA coding for GK in these teleosts. Amino acid sequences derived from these cDNA clones are highly similar to other vertebrate GKs. These findings, including a detailed phylogenetic analysis, reveal that GK gene highly homologous to mammalian GK exists in these fish species with similar tissue specific...

Early feeding of carnivorous rainbow trout (Oncorhynchus mykiss) with a hyperglucidic diet during a short period: effect on dietary glucose utilization in juveniles ArchiMer
Geurden, Inge; Aramendi, M; Zambonino, Jose-luis; Panserat, S.
Based on the concept of nutritional programming in higher vertebrates, we tested whether an acute hyperglucidic stimulus during early life could induce a long-lasting effect on carbohydrate utilization in carnivorous rainbow trout. The trout were fed a hyperglucidic diet (60% dextrin) at two early stages of development: either at first feeding Q days, stimulus 1) or after yolk absorption (5 days, stimulus 2). Before and after the hyperglucidic stimulus, they received a commercial diet until juvenile stage (> 10 g). Fish that did not experience the hyperglucidic stimuli served as controls. The short- and long-term effects of the stimuli were evaluated by measuring the expression of five key genes involved in carbohydrate utilization: a-amylase, maltase...

Effect of dietary inulin and oligosaccharides as prebiotics for weaning turbot, Psetta maxima (Linnaeus, C. 1758) ArchiMer
Mahious, A.s.; Gatesoupe, Joel; Hervy, Magali; Metailler, Robert; Ollevier, F.
Preliminary experiments were undertaken to investigate the effect of dietary inulin (Raftiline ST), oligofructose (Raftilose P95) and lactosucrose on the growth and intestinal bacteria of the marine carnivorous turbot, Psetta maxima. Turbot larvae were weaned on compound diets containing 2% Rafiline ST, 2% Raftilose P95 or 2% lactosucrose; 2% cellulose was the carbon source in the control group. The final mean weight of the group weaned with Raftilose P95 was significantly higher than those observed with the other diets. The bacterial load was highly variable in weaning turbot, especially with respect to the putative Vibrio spp. growing on TCBS agar which, in general, seemed to be dominant. Of the total load of bacterial isolates from turbot weaned on...

Morphology and Histology of the Pirarucu (Arapaima gigas) Digestive Tract International Journal of Morphology
Rodrigues,Ana Paula Oeda; Cargnin-Ferreira,Eduardo.
The present study describes the morphology and histology of the digestive tract of juvenile pirarucu, which is a Neotropical fish species with great potential for aquaculture in the Amazon region. Three size classes of pirarucu juveniles from commercial fish farms were sampled for histological examination of the oesophagus, stomach, pyloric caeca, intestine and liver. The morphological and histological features of the pirarucu digestive tract are similar to other carnivorous teleosts and enable the species to ingest, store and digest large food items. Acidic and neutral mucins are secreted by the mucosa of the oesophagus, stomach, pyloric caeca and intestine to protect the mucosal epithelium and to lubricate and facilitate food passage along the digestive...

Digestibility and pricing of Chlorella sorokiniana meal for use in tilapia feeds Scientia Agricola
Barone,Rafael Simões Coelho; Sonoda,Daniel Yokoyama; Lorenz,Evandro Kleber; Cyrino,José Eurico Possebon.
ABSTRACT: Several microalgae contain in excess of 50 % crude protein with amino acid profile comparable to that of fish meal. In addition, high polyunsaturated fatty acid contents encourage their use in animal feeding and nutrition, particularly in the formulation and processing of aquafeeds. This study aims at estimating the feasibility of Chlorella meal as feed ingredient for the feeding and nutrition of farmed tilapia based upon digestibility data. Juvenile tilapia were stocked in conical-bottomed tanks (200 L) with superficial, continuous water flow, and fed to apparent satiation in three daily meals with a reference diet and a test diet containing 30 % lyophilized Chlorella sorokiniana added of an inert marker. Feces were collected overnight by...
