Seasonal changes in mRNA encoding for cell stress markers in the oyster Crassostrea gigas exposed to radioactive discharges in their natural environment ArchiMer
Farcy, Emilie; Voiseux, Claire; Lebel, Jean-marc; Fievet, Bruno.
The North Cotentin area (Normandy, France) hosts several nuclear facilities among which the AREVA reprocessing plant of La Hague is responsible for controlled discharges of liquid radioactive wastes into the marine environment. The resulting increase in radioactivity is very small compared to natural radioactivity. However, concerns about environment protection prompted the scientific community to focus on the effects of the chronic exposure to low concentrations of radionuclides in non-human biota. This study contributes to the evaluation of the possible impact of radioactive discharges on the oyster Crassostrea gigas in the field. Real-time polymerase chain reaction was used to quantify the expression levels of genes involved in cell stress in the...

Molecular identification of differentially regulated genes in the hydrothermal-vent species Bathymodiolus thermophilus and Paralvinella pandorae in response to temperature ArchiMer
Boutet, Isabelle; Jollivet, Didier; Shillito, Bruce; Moraga, Dario; Tanguy, Arnaud.
Background: Hydrothermal vents and cold seeps represent oases of life in the deep-sea environment, but are also characterized by challenging physical and chemical conditions. The effect of temperature fluctuations on vent organisms in their habitat has not been well explored, in particular at a molecular level, most gene expression studies being conducted on coastal marine species. In order to better understand the response of hydrothermal organisms to different temperature regimes, differentially expressed genes (obtained by a subtractive suppression hybridization approach) were identified in the mussel Bathymodiolus thermophilus and the annelid Paralvinella pandorae irlandei to characterize the physiological processes involved when animals are subjected...

Brain and hepatic Hsp70 protein levels in heat-acclimated broiler chickens during heat stress Rev. Bras. Ciênc. Avic.
Guerreiro,EN; Giachetto,PF; Givisiez,PEN; Ferro,JA; Ferro,MIT; Gabriel,JE; Furlan,RL; Macari,M.
In the present study we have investigated the effects of heat acclimation on brain and hepatic Hsp70 protein levels and body temperature of broiler chickens in response to gradual heat stress. Two groups of broilers were raised up to 47 days of age under distinct temperature conditions: thermoneutral (TN, according to bird age) or hot environmental (HS, 31-33°C). At 46 days of age, the birds reared at high ambient temperature were transferred to thermoneutrality conditions. After 18 h, these birds and the birds reared at thermoneutral temperature were submitted to gradual heat stress in a climatic chamber so that environment temperature was increased from 28 to 40ºC at a rate of 2ºC/h. Colonic temperature was measured using a thermometer sensor probe at...

Gabriel,JE; Ferro,MIT; Ferro,JA; Silva,MM; Givisiez,PEN; Macari,M.
This experiment was carried out to study the effect of dietary energy on the colonic temperature and hepatic Hsp70 content in broiler chicken at room temperature and after heat stress conditions. Broiler chickens were reared up to 51 days of life, and fed diets containing high (HE -13,186 kJ ME/kg) or low (LE -12,139 kJ ME/kg) energy. At 21 and 51 days of age, the colonic temperature was measured at room temperature and liver samples were obtained for Hsp70 quantification by Western blotting analysis. It was also investigated at these ages the time course response of colonic temperature and hepatic Hsp70 level during heat stress (35º C/5 h). The data showed that at early age, at room temperature, colonic temperature or hepatic Hsp70 levels were not...

Hsp70 Genotypes and Heat Tolerance of Commercial and Native Chickens Reared in Hot and Humid Conditions Rev. Bras. Ciênc. Avic.
Duangjinda,M; Tunim,S; Duangdaen,C; Boonkum,W.
ABSTRACT Heat tolerance in poultry production was obtained attention due to the need for genetic lines that can withstand climate changes. This study aimed at investigating heat tolerance in commercial and native broiler genetics, as well as the physiological and growth performance responses of HSP70 genotypes submitted to heat stress. In Experiment I, heterophil:lymphocyte (H:L) ratio, as an indicator of heat tolerance, was compared between commercial broilers (n = 100) and Thai native chickens (n = 100). Growing chickens (with similar initial weight) of each genetic strain were randomly divided into two groups: 1) thermoneutral environment (26 oC ± 2 oC) and 2) heat stress (36 oC ± 2 oC). The results showed that native chickens originating from a...

Taurine Reduces Heat Stress by Regulating the Expression of Heat Shock Proteins in Broilers Exposed to Chronic Heat Rev. Bras. Ciênc. Avic.
Belal,SA; Kang,DR; Cho,ESR; Park,GH; Shim,KS.
ABSTRACT The aim of the current study was to investigate the effects of taurine on heat stress by evaluating them RNA and protein expressions of HSP90, 70 and 60in Ross broilers. Birds were distributed in a control group (CO) reared at 24ºC, a heat-stress group (HS) maintained at 34°C (weeks 3-5) and not supplemented with taurine, and a heat-stress group (HST) maintained at 34°C (weeks 3-5) and supplemented with 0.1% taurine from 2 weeks of age. The final body weight was significantly higher in the HST group than in the HS group (p<0.05). The mRNA expression of all three genes in the liver and of HSP90 in the muscle were significantly lower in the HST than in the HS group (p<0.05). In the liver, the expression of HSP70 and HSP60 proteins was...

Understanding the mechanisms of glutamine action in critically ill patients Anais da ABC (AABC)
Oliveira,Gisele P.; Dias,Cristina M.; Pelosi,Paolo; Rocco,Patricia R.M..
Glutamine (Gln) is an important energy source and has been used as a supplementary energy substrate. Furthermore, Gln is an essential component for numerous metabolic functions, including acid-base homeostasis, gluconeogenesis, nitrogen transport and synthesis of proteins and nucleic acids. Therefore, glutamine plays a significant role in cell homeostasis and organ metabolism. This article aims to review the mechanisms of glutamine action during severe illnesses. In critically ill patients, the increase in mortality was associated with a decreased plasma Gln concentration. During catabolic stress, Gln consumption rate exceeds the supply, and both plasma and skeletal muscle pools of free Gln are severely reduced. The dose and route of Gln administration...

Proline content and protein patterns in Eucalyptus grandis shoots submitted to high and low temperature shocks BABT
Souza,Gustavo M.; Cardoso,Victor J. M.; Gonçalves,Antonio N..
Proline content and protein patterns changes in response to temperature shocks of both acclimated and non acclimated E. grandis shoots cultivated in vitro were investigated. Analysis of soluble proteins through SDS-PAGE and proline were carried out after 12h at 12ºC (cold acclimation) or 33ºC (heat acclimation), and immediately after temperature shocks at 41ºC and 0ºC. Analyses were also performed 24h after the temperature shocks (recovery period). Temperature treatment at 0ºC did not change soluble protein patterns both in acclimatized and non acclimatized plants, whereas cold temperature induced high proline levels, which kept relatively high after recovery period. Three novel, probably HSPs, proteins (90.5, 75 and 39 kDa) were observed in both...

Hormone replacement therapy increases levels of antibodies against heat shock protein 65 and certain species of oxidized low density lipoprotein BJMBR
Uint,L.; Gebara,O.C.E.; Pinto,L.B.; Wajngarten,M.; Boschcov,P.; Luz,P.L. da; Gidlund,M..
Hormone replacement therapy (HRT) reduces cardiovascular risks, although the initiation of therapy may be associated with transient adverse ischemic and thrombotic events. Antibodies against heat shock protein (Hsp) and oxidized low density lipoprotein (LDL) have been found in atherosclerotic lesions and plasma of patients with coronary artery disease and may play an important role in the pathogenesis of atherosclerosis. The aim of the present study was to assess the effects of HRT on the immune response by measuring plasma levels of antibodies against Hsp 65 and LDL with a low and high degree of copper-mediated oxidative modification of 20 postmenopausal women before and 90 days after receiving orally 0.625 mg equine conjugate estrogen plus 2.5 mg...

Recombinant 60-kDa heat shock protein from Paracoccidioides brasiliensis: is it a good antigen for serological diagnosis of paracoccidioidomycosis? BJMBR
Peron,G.; Fernandes,F.F.; Landgraf,T.N.; Martinez,R.; Panunto-Castelo,A..
Paracoccidioides brasiliensis and P. lutzii are fungi that cause paracoccidioidomycosis (PCM), the most prevalent systemic mycosis in South America. For serological diagnosis, although 43-kDa glycoprotein (gp43) is regarded as highly specific for PCM, the occurrence of false negative reactions in sera from patients infected with P. lutzii suggests that preparation with only one antigen is not recommended. Heat shock proteins are feasible alternatives as a second antigen because they are often highly immunogenic. In this study, we evaluated the usefulness of recombinant 60-kDa heat shock protein from P. brasiliensis (rPbHsp60) for the serological diagnosis of PCM. Using western blotting assay, we observed that 77.3% of the sera from PCM patients were...
