Floridoside extracted from the red alga Mastocarpus stellatus is a potent activator of the classical complement pathway ArchiMer
Courtois, Anthony; Simon-colin, Christelle; Boisset, Claire; Berthou, Christian; Deslandes, Eric; Guezennec, Jean; Bordron, Anne.
Many biological properties of algae have been found to have useful applications in human health, particularly in the fields of oncology and immunology. Floridoside, extracted from the red alga Mastocarpus stellatus, has a structure similar to the xenoantigen Gal alpha 1-3 Gal. This xenoantigen has been described to induce a high immune response in human xenografts and is mediated by natural anti-gal antibodies that activate the classical complement pathway. Based on this property, we analyzed the potential activities of floridoside on the immune system. We demonstrated that floridoside activates a complement cascade via the classical complement pathway, through the recruitment and activation of natural IgM. This algal molecule could represent an important...

ABSENCE OF SEASONAL EFFECT ON THE IMMUNOMODULATORY ACTION OF BRAZILIAN PROPOLIS ON NATURAL KILLER ACTIVITY J. Venom. Anim. Toxins
SFORCIN,J. M.; KANENO,R.; FUNARI,S. R. C..
Propolis, a beehive product widely used in folk medicine as an antiinflammatory agent, has been attracting researchers' attention to scientifically elucidate its biological properties and therapeutic activities. The aim of this paper was to study the possible effect of propolis on natural killer activity, since propolis immunomodulatory action has been suggested, especially on non-specific immunity. Propolis was produced by africanized honeybees (Apis mellifera L.), collected throughout a whole year, and pooled by season. Hydroalcoholic solutions of propolis were prepared with each pool and administered to rats by gavage over three days. Natural killer activity of non-adherent spleen cells was evaluated by the 51Cr-release cytotoxicity assay against Yac-1...

Abstract Background Tityus serrulatus scorpion venom (TsV) contains toxins that act on K + and Na + channels and account for the venom’s toxic effects. TsV can activate murine peritoneal macrophages, but its effects on human lymphocytes have been poorly investigated. Considering that lymphocytes may play an important role in envenomation, we assessed whether TsV affects the expression of phenotypic (CD3, CD4, and CD8) and activation (CD69, CD25, and HLA-DR) markers, cell proliferation, and cytokine production in peripheral blood mononuclear cells. Methods Cytotoxicity of TsV was evaluated via the MTT assay. Cell proliferation, expression of phenotypic and activation markers, and release of cytokines were assessed using flow cytometry, after treatment...

Mechanism of action of probiotics BABT
Hemaiswarya,S; Raja,R.; Ravikumar,R.; Carvalho,Isabel S..
The modern diet doesn't provide the required amount of beneficial bacteria. Maintenance of a proper microbial ecology in the host is the main criteria to be met for a healthy growth. Probiotics are one such alternative that are supplemented to the host where by and large species of Lactobacillus, Bifidobacterium and Saccharomyces are considered as main probiotics. The field of probiotics has made stupendous strides though there is no major break through in the identification of their mechanism of action. They exert their activity primarily by strengthening the intestinal barrier and immunomodulation. The main objective of the study was to provide a deep insight into the effect of probiotics against the diseases, their applications and proposed mechanism of...

Galectins: a key intersection between glycobiology and immunology BJMBR
Rabinovich,G.A.; Riera,C.M.; Landa,C.A.; Sotomayor,C.E..
Galectins are a family of evolutionarily conserved animal lectins, widely distributed from lower invertebrates to mammals. They share sequence and structure similarities in the carbohydrate recognition domain and specificity for polylactosamine-enriched glycoconjugates. In the last few years significant experimental data have been accumulated concerning their participation in different biological processes requiring carbohydrate recognition such as cell adhesion, cell growth regulation, inflammation, immunomodulation, apoptosis and metastasis. In the present review we will discuss some exciting questions and advances in galectin research, highlighting the significance of these proteins in immunological processes and their implications in biomedical...

Immunomodulatory properties of Alternanthera tenella Colla aqueous extracts in mice BJMBR
Guerra,R.N.M.; Pereira,H.-A.W.; Silveira,L.M.S.; Olea,R.S.G..
Plants from the genus Alternanthera are thought to possess antimicrobial and antiviral properties. In Brazilian folk medicine, the aqueous extract of A. tenella Colla is used for its anti-inflammatory activity. The present study investigated the immunomodulatory property of A. tenella extract by evaluating the antibody production in male albino Swiss mice weighing 20-25 g (10 per group). The animals received standard laboratory diet and water ad libitum. The effect of A. tenella extract (5 and 50 mg/kg, ip) was evaluated in mice immunized with sheep red blood cells (SRBC 10%, ip) as T-dependent antigen, or in mice stimulated with mitogens (10 µg, Escherichia coli lipopolysaccharide, LPS, ip). The same doses (5 and 50 mg/kg, ip) of A. tenella extract were...

Comparison of different monoclonal antibodies against immunosuppressive proteins of Ascaris suum BJMBR
Oshiro,T.M.; Rafael,A.; Enobe,C.S.; Fernandes,I.; Macedo-Soares,M.F..
The extract of Ascaris suum suppresses the humoral and cellular immune responses to unrelated antigens in the mouse. In order to further characterize the suppressive components of A. suum, we produced specific monoclonal antibodies which can provide an important tool for the identification of these proteins. The A. suum immunosuppressive fractions isolated by gel filtration from an extract of adult worms were used to immunize BALB/c mice. Popliteal lymph node cells taken from the immunized animals were fused with SP2/O myeloma cells and the cloned hybrid cells obtained were screened to determine the specificity of secreted antibodies. Three monoclonal antibodies named MAIP-1, MAIP-2 and MAIP-3 were selected and were shown to react with different epitopes...

Galectin-1, an alternative signal for T cell death, is increased in activated macrophages BJMBR
Rabinovich,G.A.; Riera,C.M.; Sotomayor,C.E..
Galectin-1 belongs to an evolutionarily conserved family of animal ß-galactoside-binding proteins, which exert their functions by crosslinking the oligosaccharides of specific glycoconjugate ligands. During the past decade, attempts to identify the functional role of galectin-1 suggested participation in the regulation of the immune response. Only in the last few years has the molecular mechanism involved in these properties been clearly elucidated, revealing a critical role for galectin-1 as an alternative signal in the generation of T cell death. In the present study we will discuss the latest advances in galectin research in the context of the regulation of the immune response, not only at the central level but also at the periphery. Moreover, we will...

The role of the SLAM-SAP signaling pathway in the modulation of CD4+ T cell responses BJMBR
Vilar,M.L.L.V.; Frutuoso,M.S.; Arruda,S.M.; Lima,D.M.; Bezerra,C.S.; Pompeu,M.M.L..
The signaling lymphocytic activation molecule (SLAM), present on the surface of hematopoietic cells, can regulate some events of the immune responses. This modulatory action is associated with the capacity of SLAM to interact with an intracytoplasmic adapter, such as SLAM-associated protein (SAP). SLAM is constitutively expressed in most of these cells, is rapidly induced after antigenic or inflammatory stimuli, and participates in the immunological synapse. Defects in the function of the SLAM-SAP pathway contribute to immunological abnormalities, resulting in autoimmune diseases, tumors of the lymphoid tissues and inadequate responses to infectious agents. Initially, the role of SLAM was investigated using an anti-SLAM monoclonal antibody (α-SLAM mAb)...
