Oil palm (Elaeis guineensis Jacq.) is the highest yielding oil-bearing crop. However, being a perennial crop, genetic improvement of oil palm is extremely slow. Indeed, compared to other annual oil crops such as soybean and rapeseed, genetic manipulations remained less important. Therefore, to remain competitive, oil palm growers and breeders need new and novel approaches. In this report, the potential of immature embryos (IE) as a useful tool for oil palm genetic transformation studies was evaluated. It was evident that IEs were amenable to both direct and Agrobacterium-mediated gene transfer. Due to the abundant supply of IE, optimization of biolistic and Agrobacterium-mediated gene transfer into IEs were easily carried out. Transient transformation... |

A method for regeneration of the commercially important common bean (Phaseolus vulgaris ) using N6-benzylaminopurine(BAP) and adenine sulphate (AS) was established. Embryogenic axes of the Costa Rican common bean cultivars Bribrí, Brunca, Guaymí, Huetar and Telire were cultured on Murashige and Skoog medium supplemented with 100 mgl-1 myo-inositol, 1 mgl-1 thiamine, 30 gl-1 sucrose, BAP (0, 5 and 10 mgl-1), AS (0, 20 and 40 mgl-1) and 8 gl-1 agar. Regardless of the concentration of BAP and AS in the induction medium, the number of shoots and leaves differed significantly among the common bean cultivars evaluated. The higher average of shoots was obtained for Brunca > Telire > Bribrí > Guaymí > Huetar. Moreover, independently of the cultivar,... |

A protocol for the in vitro culture of Cucurbita pepo cv. Caserta was studied, using a cotyledon segment with an attached hypocotyl fragment as an explant. First, to determine the optimal seedling age, explants were collected from 4 to 6-day-old in vitro germinated seedlings and cultured in MS basal medium supplemented with benzylaminopurine (BAP, 4.5 µM), under a 16-h photoperiod at 27ºC. Based on the results obtained, the explants collected from the 4-day-old seedlings were then cultured in MS basal medium supplemented with different concentrations of BAP (0, 1.1, 2.2, 3.3, 4.5, or 5.5 µM) and incubated under a 16-h photoperiod at 27ºC. In vitro organogenesis was most efficient with explants collected from 4-day-old seedlings cultured in medium... |

An effective tissue culture is an indispensable basis for the production of transgenic plants. In this study, we investigated the induction of embryogenic callus production from the scutellum of spring wheat and subsequent regeneration of plants. Twenty-two Argentine spring wheat varieties from three breeders were screened to select promising genotypes for the optimization of the procedure. Experimental variables were the developmental stage of the immature embryos and the concentration of the growth regulator 2,4-dichlorophenoxyacetic acid (2,4-D). Two developmental stages, termed H and W, respectively (according to He et al., 1986), were selected, and their performance in culture media containing high and low concentrations of 2,4-D was examined.... |

In this paper we describe a new apochelan species, Milnesium variefidum sp. nov. from Scotland and provide novel morphological and molecular data for Milnesium berladnicorum Ciobanu et al., 2014. The new species differs from the most similar M. berladnicorum by the presence of developmental dimorphism in claw configuration, absent or weakly developed cuticular bars under claws I-III, a different arrangement of cuticular pseudoplates, and by differences in the sequences of three nuclear DNA fragments: 18S rRNA (p-distance: 0.6%), 28S rRNA (2.0%), ITS-2 (9.3%), and on mitochondrial gene COI (12.4%). Although ontogenetic claw configuration change was suspected to occur in some Milnesium species, we are the first to document it through the combined use of... |
