Artificial reproduction involves collection and handling of gametes in a way that secures their quality and maximizes the fertilization outcome. In addition to initial sperm quality, numerous steps can affect the final result of fertilization, from the sperm collection process until gamete mixing (or co-incubation) when spermatozoon enters or fuses with the oocyte. In this review, we summarize the whole process of sperm handling, from collection until fertilization for fish, penaeid shrimp, bivalve mollusks and marine mammals. To obtain sperm from captive animals, techniques vary widely across taxa, and include stripping by abdominal massage or testis surgical removal in fish, spermatophore collection in penaeid shrimps, gonadal scarification or... |

This study evaluated the influence of follicular fluid (FF) added to the maturation medium on the quality of bovine embryos produced in vitro. In the first experiment, oocytes were matured in media containing five different FF concentrations with different maturation times and classified according to meiotic progression and migration of cortical granules. In the second experiment, oocytes matured in the same media were fertilized at three different maturation times; thereafter, cleavage and blastocyst rates were evaluated. In the third experiment, oocytes were matured in media containing three different FF concentrations at two different maturation times, and embryo quality, inferred by the ratio of inner cell mass and trophectoderm cells compared with... |

In vitro embryos production from prepubertal heifers can help contribute to breeding programs; however, strategies are necessary to increase their embryo production. The aim of this study was to investigate the effects of two nutritional plans on oocyte recovery, embryo production and growth performance of prepubertal Nelore heifers. Thirty-four Nelore heifers with age of 6.5 months were divided into two feeding treatments (NP1 and NP2). The NP1 diets served as the control and NP2 diets were formulated to contain an average of 1.22-fold more energy than NP1. After 3 months of supplementation, the animals underwent follicular aspiration (ovum pick-up, OPU) every 21 d for 3 months and embryos were produced in vitro.... |

The sirtuins are members of the histone deacetylase (HDAC) family and are involved in the regulation of transcription activation and apoptosis. In this regard, the aim of this study was to investigate the influence of different temperatures during in vitro maturation (IVM) on the developmental competence of bovine oocyte, as well as mRNA expression of sirtuins. |

Despite significant improvements in the in vitro production of cattle embryos, the suboptimal in vitro culture environment still limits the embryo quality and production. Techniques that associate the advantages of in vivo and in vitro systems, such as intrafollicular transfer of immature oocytes, have been proposed mainly to increase the embryo quality. In this context, we tried to use a goat as live incubator and associated nonsurgical embryo transfer techniques in small ruminants to perform ex situ (in vivo) maturation of bovine oocytes. For this, immature bovine cumulus-oocyte complexes (COCs) of grade 1 and 2 were randomly distributed into two groups for in vitro (IVM; n = 38) and ex situ (ESM; n = 40) maturation. The IVM was performed for a period of... |

Bos indicus cows usually have better reproductive performance in tropical and subtropical regions than Bos taurus cows, presumably due to their better adaptation to tropical environments. The aim of this study was to evaluate the developmental competence and expression of the Hsp 70.1 gene in immature oocytes from B. taurus (Holstein) and B. indicus (Gyr) dairy cows raised in a tropical region. Cumulus–oocyte complexes were obtained by transvaginal ultrasound-guided follicle aspiration between spring and early autumn, and subjected to in vitro maturation and fertilization. Presumptive zygotes were co-cultured with their own cumulus cells in CR2aa media with 10% fetal calf serum; Grade 1 blastocysts were transferred to synchronized crossbred recipients. The... |
