Otolith growth in trout Oncorhynchus mykiss: supply of Ca2+ and Sr2+ to the saccular endolymph ArchiMer
Payan, P; Borelli, G; Priouzeau, F; De Pontual, Helene; Boeuf, G; Mayer Gostan, N.
Kinetic and pharmacological characteristics of Ca2+ fluxes across the saccular epithelium of trout were studied using a perfused isolated inner ear. Ca-45(2+) influx from the Ringer solution to the endolymph was 3-4 nmoles h(-1) mu(-1) endolymph, which corresponds to a global turnover rate of the endolymph calcium of 200% h(-1). Ca2+ entry into the proximal endolymph was faster than into the distal fluid. Net Ca2+ movement across the saccular epithelium depended on the direction and intensity of the chemical gradient of calcium between the Ringer solution and the endolymph. Increasing the calcium concentration in the Ringer solution up to 4.4 mmol l(-1) provoked an accumulation of Ca2+ in both proximal and distal endolymphs, and equilibrium was reached...

Sources de variation intra-populationelle de la morphologie des otolithes: asymétrie directionnelle et régime alimentaire ArchiMer
Mille, Tiphaine.
Otoliths are calcified structures located in Osteichthyes’ inner ear that are involved in audition and balance. Their morphology is used as an indicator of various ecological processes or properties. This application requires identifying the endogenous and exogenous factors that act simultaneously as sources of shape variation. This thesis aims at detecting and quantifying the relative contributions of directional asymmetry and diet to otolith shape variation at the intra-population level. Directional asymmetry between left and right otoliths was found in flat-fishes, the blind-side otolith being always longer and larger, whereas it was negligible in round-fishes. However, asymmetry amplitude never exceeded 18 %, which suggests evolutionary canalization of...

Ionic composition of endolymph in teleosts: Origin and importance of endolymph alkalinity ArchiMer
Payan, P; Kossmann, Hans; Watrin, A; Mayer-gostan, N; Boeuf, Gilles.
Ionic (Na+, K+, Cl-, PO43-, pH), total CO2, total calcium and protein concentrations in the plasma and endolymph of the inner ear were compared in trout Oncorhynchus mykiss and turbot Scophthalmus maximus. In both species, saccular endolymph was characterized by high levels of K+ and total CO2 and in trout by an alkaline pH, The kinetic characteristics of proton secretion across the saccular epithelium of trout were investigated using a titration technique in which isolated saccules were mounted as closed sacs. The rate of proton secretion depends strongly on the pH of the Ringer's solution and secretion stops at a pH below 7.2, Proton secretion is driven by an energy-dependent mechanism involving basolateral ouabain-sensitive Na+/K+ exchangers. Proton...

In vitro Study of Morphological Changes of the Cultured Otocyst Isolated from the Chick Embryo International Journal of Morphology
Intarapat,Sittipon; Gonmanee,Thanasup; Thonabulsombat,Charoensri.
The aim of this study was to observe morphological changes of the cultured otocysts isolated from various stages of the chick embryo. Isolated otocysts were dissected from embryonic day, E2.5-4.5 of incubation (HH stage 16-26) according to stages of developing inner ear. Morphology of the chick otocyst exhibited an ovoid shape. The width and height of the otocyst were 0.2 mm and 0.3 mm, respectively. Elongation of a tube-like structure, the endolymphatic duct, was found at the dorsal aspect of the otocyst. The cultured otocyst is lined by the otic epithelium and surrounding periotic mesenchymal cells started to migrate outwards the lateral aspect of such epithelium. Notably, the acoustic-vestibular ganglion (AVG) was observed at the ventrolateral aspect of...
