β-lactoglobulin (BLG), a dominant allergen in goat milk, is difficult to remove by traditional biochemical methods. Its elimination from goat milk by genetic modification therefore poses a major challenge for modern goat breeders. A shRNA targeting BLG mRNA with high interference efficiency was identified, with which lentiviral vectors were used for mediating stable shRNA interference in goat-fetal fibroblast cells. Apart from high efficiency in the knockdown of BLG expression in these cells, lentivector-mediated RNAi manifested stable integration into the goat genome itself. Consequently, an in vitro model for goat BLG-content control was compiled, and a goat-cell line for accompanying transgenetic goat production created. |

O presente trabalho teve como objetivo verificar a viabilidade do antígeno (Ag) para diagnóstico de lentivírus caprino por meio do teste de imunodifusão em gel de ágar (IDGA), em diferentes temperaturas, com e sem a adição do conservante Phenylmethylsulfonyl Fluoride (PMSF). A proteína do capsídeo viral - p28 presente no antígeno é bem estável, suportando a temperatura ambiente média de 25 °C por até 15 dias. A ação do conservante PMSF é efetiva para a manutenção dos níveis proteicos, além disso, é quase inerte na formação da linha de precipitação. O congelamento a -20 °C e, principalmente a -80 °C, com ou sem a adição de conservante, é muito eficaz para a manutenção da quantidade e qualidade da proteína viral p28 presentes no antígeno de Lentivírus... |

A caprino-ovinocultura é expressiva no nordeste brasileiro e de relevância socioeconômica para a região do semiárido. No entanto, as infecções por Toxoplasma gondii e Lentivirus podem estar presentes nos rebanhos e ocasionar problemas reprodutivos, articulares e neurológicos. Para verificar a circulação destes agentes em 37 criatórios de pecuaristas familiares do município de Cansanção, Bahia, foram avaliados caprinos e ovinos com idades superiores a um ano, criados extensivamente. BACKGROUND: Ubiquitin Specific Peptidase 39 (USP39) is a 65 kDa SR-related protein involved in RNA splicing. Previous studies showed that USP39 is related with tumorigenesis of human breast cancer cells. RESULTS: In the present study, we investigated the functions of USP39 in human hepatocellular carcinoma (HCC) cell line SMMC-7721. We knocked down the expression of USP39 through lentivirus mediated RNA interference. The results of qRT-PCR and western blotting assay showed that both the mRNA and protein levels were suppressed efficiently after USP39 specific shRNA was delivered into SMMC-7721 cells. Wound scarring remains a major challenge for plastic surgeons. Transforming growth factor (TGF)-β plays a key role in the process of scar formation. Previous studies have demonstrated that truncated TGF-β type II receptor (t-TGF-βRII) is unable to continue signal transduction but is still capable of binding to TGF-β, thereby blocking the TGF-β signaling pathway. Hepatocyte growth factor (HGF) is a multifunctional growth factor that promotes tissue regeneration and wound healing.
