The delay of the maize harvest may cause qualitative and quantitative grain losses. This work evaluated the effects of the harvest time on the agronomic performance of maize hybrids with contrasting growth cycles. The experiment was set in Lages, Santa Catarina State,South Brazil, during the 2012/2013 and 2013/2014 growing seasons. A randomized block design, arranged in split plots was used. Five single-cross hybrids were tested in the main plots: P1630H and P32R22H (hyper-early), P2530 (super early) and P30F53YH and P30R50YH (early). Five harvest times were assessed in the split plots: 0 (grain physiological maturity), 10, 20, 30 and 40 days after physiological maturity. The sowing was performed on 12/5/2012 and 10/5/2013. The percentage of lodged and... |

ABSTRACT The aim of this study was to evaluate somatic embryogenesis in juvenile explants of the THB papaya cultivar. Apical shoots and cotyledonary leaves were inoculated in an induction medium composed of different concentrations of 2,4-D (6, 9, 12, 15 and 18 µM) or 4-CPA (19, 22, 25, 28 and 31 µM). The embryogenic calluses were transferred to a maturation medium for 30 days. Histological analysis were done during the induction and scanning electron microscopy after maturing. For both types of auxin, embryogenesis was achieved at higher frequencies with cotyledonary leaves incubated in induction medium than with apical shoots; except for callogenesis. The early-stage embryos (e.g., globular or heart-shape) predominated. Among the auxins, best results... |

The present study analyzed the reproduction of wild-caught Penaeus paulensis in relation to its molt cycle. The experimental design consisted of two treatments (continuous water flow and batch renewal) with two replicates. The stocking density in experimental tanks (1.50 x 0.96m) was approximately 7 animals/m², resulting in 4 males (23.9 ± 2.3 g) and 6 females (52.0 ± 5.5 g) per tank. Shrimp had their uropods cut for individual marking and female maturation was induced through the unilateral eyestalk-ablation. The intermolt period of females and males (17.4 ± 3.2 and 17.8 ± 4.6 days, respectively), number of days between molting and the first spawn (6.9 ± 2.8 days) and number of spawns in the intermolt period (1.4 ± 0.5), presented no significant... |

A simple and efficient protocol for recurrent somatic embryogenesis and plant regeneration is one of the prerequisites for genetic improvement of guava. An efficient reproducible regeneration somatic embryogenesis protocol was developed in four genotypes of Psidium guajava L. using immature zygotic embryo as starter explant. Somatic embryogenesis induction was obtained on MS basal medium supplemented with 2.0 mgL-1 2, 4-D, 400 mgL-1 L-glutamine, 6% sucrose and 500 mgL-1 Malt extract. Following SE induction different developmental stages of somatic embryos (Globular, heart-shaped, torpedo, cotyledonary) was directly obtained and further recurrent embryogenesis also obtained upon prolonged incubation in induction media. Addition of polyethylene glycol (50... |

Hepatic progenitor cells (HPCs) are a potential cell source for liver cell transplantation but do not function like mature liver cells. We sought an effective and reliable method to induce HPC maturation. An immortalized HP14.5 albumin promoter-driven Gaussian luciferase (ALB-GLuc) cell line was established from HPCs isolated from fetal mouse liver of post coitus day 14.5 mice to investigate the effect of induction factors on ALB promoter. HP14.5 parental cells were cultured in DMEM with different combinations of 2% horse serum (HS), 0.1 µM dexamethasone (DEX), 10 ng/mL hepatic growth factor (HGF), and/or 20 ng/mL fibroblast growth factor 4 (FGF4). Trypan blue and crystal violet staining were used to assess cell proliferation with different induction... |

Most contacts with food protein and microbiota antigens occur at the level of the gut mucosa. In animal models where this natural stimulation is absent, such as germ-free and antigen-free mice, the gut-associated lymphoid tissue (GALT) and systemic immunological activities are underdeveloped. We have shown that food proteins play a critical role in the full development of the immune system. C57BL/6 mice weaned to a diet in which intact proteins are replaced by equivalent amounts of amino acids (Aa diet) have a poorly developed GALT as well as low levels of serum immunoglobulins (total Ig, IgG, and IgA, but not IgM). In the present study, we evaluated whether the introduction of a protein-containing diet in 10 adult Aa-fed C57BL/6 mice could restore their... |

The phytohormone ethylene is involved in several physiological and developmental processes in higher plants, including ripening of fruits, abscission of organs and tissues, senescence, wound response as well as in other abiotic stresses. The enzyme 1-aminocyclopropane-1-carboxylate oxidase (ACO) catalyzes the last step of ethylene biosynthesis. The production of ethylene and the expression of a Coffea arabica ACO gene during the last stages of fruit maturation were investigated. A rapid increase of ethylene production at the green-yellow stage of fruit ripening, after the end of endosperm formation, and a decrease of ethylene production at the cherry stage indicates a climacteric phase during ripening. An ACC oxidase (Ca-ACO) from coffee fruit cDNA was... |

Abscisic acid (ABA) and indole-3-acetic acid (IAA) participate in the propagation of plants by somatic embryogenesis, causing polar structural differentiation of the embryo. The goal of the assay was to compare endogenous levels of ABA and IAA between somatic embryos (SE) and zygotic embryos (ZE) of Nothofagus alpina (Poepp. & Endl.) Oerst. In this study, a somatic embryo maturation assay involving the addition of varying concentrations of exogenous ABA was performed on cotyledonary-stage of N. alpina. Furthermore, the endogenous levels of ABA and IAA were quantified in the immature ZE, the mature ZE, and the embryonic axis of a mature embryo of N. alpina. The current study utilized high performance liquid chromatography (HPLC) for quantification.... |

The efficiency of composting processes with and without the addition of Californian red worms (Eisenia foetida) was evaluated, using manure of dairy cows to generate organic fertilizer. Several parameters were assessed as physio-chemical indicators of maturity, such as temperature, pH, C/N ratio, phytotoxicity and macro and micronutrients at 25 and 54 ws (mature and stored products, respectively). A linear model was used in the statistical analysis, with four replicates that included as fixed effects, the composting systems and time. At 25 wk, both systems reduced the C/N ratio to values of 15.5 in vermicompost and 17.1 in semi-compost. The content of total N, N-NO3, Ca and some micronutrients also increased. K and Na concentrations were higher in the... |

The objective of this study was to investigate the mRNA expression and protein localization of Grb10 gene in bovine cumulus-oocyte complexes (COCs) from different follicle sizes. Firstly, it was investigated the mRNA expression to correlate with maturation rates. COCs from follicles at 1-3, 4-6, 6-8 and >8mm were used to evaluate Grb10 gene expression by qRT-PCR assay and nuclear maturation rates. It was observed that more competent oocytes (from follicles at 6-8 and >8mm; P>0.05), had lower Grb10 mRNA expression levels when compared to the oocytes from follicles at 1-3 and 4-6mm (P>0.05). After it was performed an immunofluorescence analysis in COCs from different follicle sizes (1-3, 4-6, 6-8 and >8mm) to investigate... |
