Surimi de chinchard ( Trachurus trachurus L. ) - Influence de conditions technologiques sur des caractéristiques de texture du produit après traitement thermique ArchiMer
Verrez-bagnis, Veronique.
Surimi, washed fish mince after grinding with salt, has a firm and elastic gel texture or “ashi” after thermal processing. The major objectives of this study on horse mackerel (Trachurus trachurus) surimi are to optimize the technological conditions in the aim to obtain the best ashi, to determine the influence of frozen storage and of egg white and potata starch addition, and to establish the preventive effect of incorporation into surimi of different combinations of cryoprotectants and NaC1. Gel texture is, by order importance, dependent on cooking temperature, length of setting and cooking and length of grinding of surimi with texture additives after thawing. Changes of gelling properties after 6 months at - 30°C of frozen st orage are limited. The...

Redescription and phylogenetic analyses of Durchoniella spp. (Ciliophora, Astomatida) associated with the polychaete Cirriformia tentaculata (Montagu, 1808) ArchiMer
Sauvadet, Anne-laure; Lynn, Denis H.; Roussel, Erwan; Le Panse, Sophie; Bigeard, Estelle; Schrevel, Joseph; Guillou, Laure.
Microscopic and phylogenetic analyses were performed on endocommensal astome ciliates retrieved from the middle intestine of a marine cirratulid polychaete, Cirriformia tentaculata, collected in the bay of Roscoff (English Channel, Northwest French coast) and on the Southwest English coast. Three morphotypes of the astome genus Durchoniella were identified, two corresponding to described species (the type species Durchoniella brasili (Léger and Duboscq, 1904) de Puytorac, 1954 and Durchoniella legeriduboscqui de Puytorac, 1954) while a third morphotype remains undescribed. Their small subunit (SSU) rRNA gene sequences showed at least 97.2% identity and phylogenetic analyses grouped them at the base of the subclass Scuticociliatia (Oligohymenophorea), as a...

Induction of apoptosis by UV in the flat oyster, Ostrea edulis ArchiMer
Gervais, Ophelie; Renault, Tristan; Arzul, Isabelle.
Apoptosis is a fundamental feature in the development of many organisms and tissue systems. It is also a mechanism of host defense against environmental stress factors or pathogens by contributing to the elimination of infected cells. Hemocytes play a key role in defense mechanisms in invertebrates and previous studies have shown that physical or chemical stress can increase apoptosis in hemocytes in mollusks. However this phenomenon has rarely been investigated in bivalves especially in the flat oyster Ostrea edulis. The apoptotic response of hemocytes from flat oysters, O. edulis, was investigated after exposure to UV and dexamethasone, two agents known to induce apoptosis in vertebrates. Flow cytometry and microscopy were combined to demonstrate that...

Ostreopsis lenticularis Y. Fukuyo (Dinophyceae, Gonyaulacales) from French Polynesia (South Pacific Ocean): A revisit of its morphology, molecular phylogeny and toxicity ArchiMer
Chomérat, Nicolas; Bilien, Gwenael; Derrien, Amelie; Henry, Kévin; Ung, André; Viallon, Jérôme; Darius, Hélène Taiana; Mahana Iti Gatti, Clémence; Roué, Mélanie; Hervé, Fabienne; Réveillon, Damien; Amzil, Zouher; Chinain, Mireille.
To date, the genus Ostreopsis comprises eleven described species, of which seven are toxigenic and produce various compounds presenting a major threat to human and environmental health. The taxonomy of several of these species however remains controversial, as it was based mostly on morphological descriptions leading, in some cases, to ambiguous interpretations and even possible misidentifications. The species Ostreopsis lenticularis was first described by Y. Fukuyo from French Polynesia using light microscopy observations, but without genetic information associated. The present study aims at revisiting the morphology, molecular phylogeny and toxicity of O. lenticularis based on the analysis of 47 strains isolated from 4 distinct locales of French...

Cellular Changes in the Placenta in Pregnancies Complicated with Diabetes International Journal of Morphology
Verma,Ranjana; Mishra,Sabita; Kaul,Jagat Mohini.
Placenta is the most accurate record of the infant prenatal experience. After delivery if the placenta is examined minutely, it provides much insight into the prenatal health of the baby and the mother. In diabetic pregnancy, placental weight is higher in comparison to normal pregnancy. To study the cellular differences that might contribute to larger size of placenta, light microscopic analysis was performed on 25 full term placentas, out of which 20 were of Gestational diabetes mellitus (12 controlled on diet, 8 controlled on insulin) and 5 control group. Tissue sections were processed and analyzed. Birth weight of neonate, placental weight, vascular pattern of chorionic blood vessels and site of attachment of umbilical cord were recorded. In the...

Electron Microscopic Evaluation of the Secretory Mechanisms of Renin from Juxtaglomerular Cells International Journal of Morphology
Aktas,Ranan Gulhan; Karabay,Gulten; Taskinalp,Oguz; Kutlu,A. Kemal.
Although the structure and the functions of juxtaglomerular cells (JG) have been well defined, there is still a controversy about the secretory mechanisms of renin from these cells. It has been assumed that exocytosis is the main secretory mechanism in these cells in many studies, while others suggest that secretion occurs in a quite different way in these cells. There are several studies suggesting that diacrine secretion, which is very difficult to visualize, might be the other mechanism for secretion of renin. This study is an attempt to find the answers of these questions by identifying the fine structural features of the secretory granules in juxtaglomerular cells. Cyclosporin A (CyA) has been used in the current experimental study since it has...

The hammerhead sharks shows a head laterally expanded with eyes and nostrils on the edges, which gives the species a hammer appearance. Another strand of studies indicates that the hypothesis of head shape alterations for better adaptions in the sensorial system with the development of structures associated to binocular sight and others sensorial organs. Given the variety of characteristics, the study aim was to describe the anatomical constitution of the hammerhead shark eye bulb. The bulb and its annexes exenteration was performed, and further dissection; the morphological description of the muscle insertions and eye bulb components were based on direct observation and were further photographed and catalogued. The eye bulb fragments and its annexes were...

Estrogen Receptors in Immunohistochemically Stained Slides: Comparing Subjective and Computerized Evaluation Methods International Journal of Morphology
Millas,Ieda; Barros,Mirna Duarte; Curcio,Daniella Franco; Buck,Hudson Sousa; Liquidato,Bianca Maria.
Immunohistochemistry is a specific and sensitive staining method for detection of several proteins. One important function of this method is to help us on the diagnosis of the presence of specific receptors as the estrogen receptors. The aim of this study was to compare two different methods to evaluate immunohistochemical staining intensity to detect the presence of ER in the nasal mucosa tissue: one using a digital system and the other through conventional direct microscopy observation. Sixty two stained samples were observed and analyzed under optic microscopy by three specialized professionals, who have graded intensity from: absent, mild, moderate and intense staining. Afterwards, an objective measurement was obtained by a relative optical density...

Tegumentary epithelial glands in the abdomen of virgin and physogastric queens of the stingless bee Scaptotrigona postica Latreille (Meliponini: Trigonina) Neotropical Entomology
Abdalla,Fábio C.; Cruz-Landim,Carminda da; Gracioli,Luciana F..
In the eusocial worker bees, portions of the epithelium of the abdominal epidermis may become hypertrophied and glandular, and produce the wax used to build the brood alveoli. In meliponine bees, the presence of similar epithelium was already observed in the queens, but its function is still unknown. Would this epithelium have homologous function in the workers? May the queens display other functions, besides reproduction, related to those considered exclusive of workers? In order to contribute to solve these questions, a histological and ultrastructural study of the tegumentary epithelial glands of the third tergite was carried out in virgins and physogastric queens of Scaptotrigona postica Latreille. The glandular tegumentary epithelium was easily...

Microstructure and flow properties of lyophilized mango pulp with maltodextrin AGRIAMBI
Afonso,Marcos R. A.; Rodrigues,Brena K. M.; Costa,José M. C. da; Rybka,Ana C. P.; Wurlitzer,Nedio J..
ABSTRACT The present study had as objective to determine the flow properties and behavior of lyophilized mango pulp powder as a function of different concentrations of maltodextrin. In the samples of the powders contain 5, 10 and 15% of maltodextrin the angle of effective internal friction, wall friction angle, flow index and bulk density were determined. The microstructure was evaluated by scanning electron microscopy. The freeze-dried samples are characterized as having an amorphous structure, and the drying aid used modified the surface of the particles. The flow index was 3.19, 4.28 and 4.53 for samples containing, respectively, 5, 10 and 15% maltodextrin. Increasing the concentration of maltodextrin in the mango pulp decreased the effective angles of...

A universal microscope manipulator Rev. Bras. entomol.
Boyadzhiev,Peter S.; Gechev,Tsanko S.; Donev,Atanas D..
A modified and improved model of a mechanical manipulator for observation of pinned and mounted insects is described. This device allows movement of the observed object around three perpendicular axes in the field of vision at all magnifications of stereomicroscopes. The main improvement of this new model is positioning of the guiding knobs for rotating around two of the axes next to each other, allowing faster and easier manipulation of the studied object. Thus, one of the main advantages of this device is the possibility to rotate the specimen without the need to refocus. The device enables easily reaching a precession deviation in the intersection point of axes up to 0.5 mm in the process of assembling.

Closely-related Xanthomonas citri subsp. citri isolates trigger distinct histological and transcriptional responses in Citrus limon Scientia Agricola
Orce,Ingrid Georgina; Debes,Mario; Sendín,Lorena; Luque,Ana; Arias,Marta; Vojnov,Adrián; Marano,María; Castagnaro,Atilio; Filippone,María Paula.
ABSTRACT Citrus canker, caused by Xanthomonas citri subsp. citri (Xcc), has an important economic impact on the citrus industry. Extensive information is available about the disease but, nevertheless, the study of plant-pathogen interactions could provide new information in the understanding of citrus canker disease. A new isolate has been identified, Xcc AT, which has a high genetic similarity (> 90 %) to the virulent Xcc T strain based on genetic clustering analyses of the rep-PCR fingerprinting patterns, but it does not produce cankerous lesions in Citrus limon. In this study, we compared C. limon responses to Xcc AT and to the virulent Xcc T strain at both histological and transcriptional levels. Histologically, leaves inoculated with Xcc AT...

Interspecific xenia and metaxenia in seeds and fruits of tomato Scientia Agricola
Piotto,Fernando Angelo; Batagin-Piotto,Katherine Derlene; Almeida,Marcílio de; Oliveira,Giancarlo Conde Xavier.
Xenia, the transmission of traits from the pollinizer to the female's tissues, is a phenomenon hitherto unknown in tomatoes. Here, we describe xenia effects on the seeds and fruits of Solanum lycopersicum, the tomato, elicited by S. galapagense. The wild tomatoes, such as S. galapagense, have highly pilose fruit surface and minute seeds, unlike the domesticated species. Crossings between S. galapagense (pollinizer) and two large-seeded, glabrous cultivars of S. lycopersicum (females) tested the former's ability to raise the trichome density and trichome-to-1000-cell ratio and to reduce the seed weight in the latter's fruits. Selfed fruits of the two cultivars, Micro-Tom and Pusa Ruby, were compared to the crossed fruits. The pollen of S. galapagense was...

Photoperiod and feeding behavior of the Antarctic fish Notothenia rossii (Perciformes: Nototheniidae) and functional morphology of chemical and visual sensory structures used in foraging Zoologia
Donatti,Lucélia; Zaleski,Tânia; Calil,Patrícia; Fanta,Edith.
The influence of photoperiod on the ability of Notothenia rossii Richardson, 1844 to detect prey was studied experimentally. In 12L/12D there was the highest number of individuals stimulated visually and chemically while in 24D chemical perception was the same as that observed in 12L/12D, the latter being lower than in 24L. The ultrastructure of chemo- and photo-sensory structures, involved in the detection of food, was studied. The photoreceptor cells of the retina are single and double cones, and rods. Taste buds were concentrated in the central part of upper and lower lips. Their shape is even, but the size is variable. The olfactory rosette has a sensory and a non-sensory epithelium, being rich in different mucosubstances. The conclusion was that in...

Effects of salinity on the physiology of the red macroalga, Acanthophora spicifera (Rhodophyta, Ceramiales) Acta Botanica
Pereira,Débora Tomazi; Simioni,Carmen; Filipin,Elisa Poltronieri; Bouvie,Fernanda; Ramlov,Fernanda; Maraschin,Marcelo; Bouzon,Zenilda Laurita; Schmidt,Éder Carlos.
ABSTRACT Salinity is an important abiotic factor since it is responsible for the local and/or regional distribution of algae. In coastal regions, salinity changes with prevailing winds, precipitation and tide, and particularly in extreme intertidal conditions. Acanthophora spicifera is a red seaweed that occurs in the supratidal region in which changes in abiotic conditions occur frequently. This study evaluated the effects of salinity on the metabolism and morphology of A. spicifera. Algae were acclimatized under culture conditions with sterilized seawater for seven days. Experiments used different salinities (15 to 50 psu) for seven days, followed by metabolic analyses. This study demonstrates that extreme salinities affect physiological parameters of A....
