NEUROTOXIC POTENTIAL OF TRICHLORFON TO MULTIPLE SUBLETHAL DOSES IN WISTAR RATS Acta biol.Colomb.
TAPIERO HERNÁNDEZ,YACSON; RONDÓN BARRAGÁN,IANG; CÉSPEDES RUBIO,ANGEL.
The organophosphates used for pest control induce sensory, motor and psychiatric disturbances after chronic exposure. The ester type is the cause of the intermediate syndrome and delayed neuropathy, in which the white and gray matter in the brain are severely affected. The aim of this study was to evaluate the effect of multiple sublethal doses of Trichlorfon on neurons, astrocytes and myelinated tissue in a rat model of brain neurotoxicity. Trichlorfon (metrifonate) was administered to adult Wistar rats at doses of 11 or 22 μg/kg by oral gavage every seven days for four or eight weeks (four experimental groups) and a control group (placebo). One week after the last dose, animals were euthanized and the brains perfused, removed and cut into coronal...

GM1 improves neurofascin155 association with lipid rafts and prevents rat brain myelin injury after hypoxia-ischemia BJMBR
Zhang,Y.P.; Huang,Q.L.; Zhao,C.M.; Tang,J.L.; Wang,Y.L..
White matter injury characterized by damage to myelin is an important process in hypoxic-ischemic brain damage (HIBD). Because the oligodendrocyte-specific isoform of neurofascin, neurofascin 155 (NF155), and its association with lipid rafts are essential for the establishment and stabilization of the paranodal junction, which is required for tight interaction between myelin and axons, we analyzed the effect of monosialotetrahexosyl ganglioside (GM1) on NF155 expression and its association with lipid rafts after HIBD in Sprague-Dawley rats, weighing 12-15 g, on day 7 post-partum (P7; N = 20 per group). HIBD was induced on P7 and the rats were divided into two groups: one group received an intraperitoneal injection of 50 mg/kg GM1 three times and the other...

Chronic postnatal administration of methylmalonic acid provokes a decrease of myelin content and ganglioside N-acetylneuraminic acid concentration in cerebrum of young rats BJMBR
Brusque,A.M.; Rotta,L.; Pettenuzzo,L.F.; Junqueira,D.; Schwarzbold,C.V.; Wyse,A.T.; Wannmacher,C.M.D.; Dutra-Filho,C.S.; Wajner,M..
Levels of methylmalonic acid (MMA) comparable to those of human methylmalonic acidemia were achieved in blood (2-2.5 mmol/l) and brain (1.35 µmol/g) of rats by administering buffered MMA, pH 7.4, subcutaneously twice a day from the 5th to the 28th day of life. MMA doses ranged from 0.76 to 1.67 µmol/g as a function of animal age. Control rats were treated with saline in the same volumes. The animals were sacrificed by decapitation on the 28th day of age. Blood was taken and the brain was rapidly removed. Medulla, pons, the olfactory lobes and cerebellum were discarded and the rest of the brain ("cerebrum") was isolated. Body and "cerebrum" weight were measured, as well as the cholesterol and triglyceride concentrations in blood and the content of myelin,...
