Abstract: The objective of this work was to evaluate the microbicidal effect of silver nanoparticles (AgNPs) on potentially toxigenic fungi affecting cocoa (Theobroma cacao) crops. These fungi, isolated from diseased cocoa pods, were characterized phenotypically and genotypically. The microbicidal effect was assessed by measuring radial mycelial growth, in synthetic culture media, and at different AgNP concentrations in plant tissues. The inhibition effect was monitored in Petri dishes, and changes in fungal structures were observed through scanning electron microscopy. Two potentially toxigenic fungi were highly prevalent: Aspergillus flavus and Fusarium solani. The inhibition assays, performed in liquid and solid synthetic culture media, showed that... |

ABSTRACT Mycotoxins, which are produced by some fungi under improper storage conditions before or after harvesting in plant products, cause acute or chronic toxicities. Ochratoxin A (OA) which is also one of the harmful mycotoxins pose a threat to animal and human health. This study was carried out in order to determine OA levels in mixed feed and feedstuffs materials used in livestock enterprises throughout Sivas province. The 59 mixed feeds and 30 feedstuffs materials collected from different enterprises was analysed. Ochratoxin A concentrations were quantified using immunoassay (ELISA). In result, OA was found to be positive in 64 (71.91%) of the 89 samples analysed, whereas OA was not found in 25 samples (28.09%). |

The aim of this study was to determine the ochratoxin A (OTA) contamination of instant coffee samples collected in the market of the city of São Paulo, Brazil from August to December, 2004. The EN 14133/2003 method, originally developed to quantify OTA in wine, grape juice and beer samples, was evaluated and approved for analyzing OTA in instant coffee samples. OTA was isolated in an immunoaffinity column and quantified by HPLC with fluorescence detection. The established detection and quantification limits were 0.16 and 0.52 ng/g, respectively. The recoveries from spiked samples were 92.6 ± 1.7, 83.7 ± 0.8, and 91.0 ± 1.2 % at levels of 3.0, 5.0, and 8.0 ng/g, respectively. Of a total of 82 samples analised, 81 (98.8%) contained OTA at levels ranging from... |

The genera Aspergillus comprises species that produce mycotoxins such as aflatoxins, ochratoxins and patulin. These are cosmopolitan species, natural contaminants of agricultural products. In coffee grains, the most important Aspergillus species in terms of the risk of presenting mycotoxins belong to the genera Aspergillus Section Circumdati and Section Nigri. The purpose of this study was to assess the occurrence of isolated ochratoxigenic fungi of coffee grains from organic and conventional cultivation from the South of Minas Gerais, Brazil, as well as to evaluate which farming system presents higher contamination risk by ochratoxin A (OTA) produced by fungi. Thirty samples of coffee grains (Coffea arabica L.) were analysed, being 20 of them of... |

Toxigenic fungi were studied in beans (Phaseolus vulgaris L.) of Classes black and color, cultivated in different regions of the State of Santa Catarina, south region of Brazil. The mean counts of filamentous fungi were 2.8 x 103 and 6.7 x 103 CFU/g for beans Classes black and color, respectively. Penicillium spp., Aspergillus spp. and Phoma spp. were the most frequent genera isolated, followed by Ryzopus spp., Alternaria spp., Helminthosporium spp., Cladosporium spp., Botrytis spp., Fusarium spp., Trichoderma spp., Curvularia spp. and Dreschelera spp. Among beans Class black, 24.6% of the Aspergillus strains produced mycotoxins: 13.1% produced aflatoxins (AFs); 11.5% produced ochratoxin A (OTA) and 28.9% of Penicillium produced citrinin (CTR). On the... |

During the summer of 2005, a total of 101 samples of wines and grape juices purchased from supermarkets and retail stores in São Paulo city were analysed for the presence of Ochratoxin A (OTA). OTA was evaluated in 29 red wines and 38 grape juices produced in Brazil and in 34 imported red wines (from Argentina, Chile, Uruguay, France, Italy, Portugal, Spain and South Africa). OTA was extracted in an immunoaffinity column and detected by HPLC with fluorescence detection, according to EN 14133/2003. The detection and quantification limits established were 0.01 and 0.03 ng/mL, respectively. The recoveries for wine samples were 94.1, 82.5, 86.1% and the relative standard deviation were 6.10, 1.03, 4.11% at levels of 0.03, 2.0, 5.0 ng/mL, respectively. For... |

ABSTRACT: The grape juice was included in meals of public schools in the states of Rio Grande do Sul and Santa Catarina, Brazil. The juices intended for schools have been produced by small wineries through the steam distillation method. This method presents a particularity in relation to other juice production methods, since the grapes are not macerated to extract the juice. In literature is well established that maceration of grapes is the main critical point associated with the ochratoxin A (OTA) contamination in juices. OTA is a mycotoxin with nephrotoxic and immunosuppressives effects. The aim of this study was to verify the OTA occurrence in juices produced by steam distillation method, as grape juice may be a source of OTA exposure for children. No... |
