Comparison of haploid and doubled haploid sugar beet clones in their ability to micropropagate and regenerate Electron. J. Biotechnol.
Klimek-Chodacka,Magdalena; Baranski,Rafal.
Background: Haploid plant material is considered as recalcitrant to organogenesis, propagation, and maintenance in vitro. However, sugar beet (Beta vulgaris L.) breeders utilizing doubled haploid (DH) technology in their breeding programs indicate that sugar beet haploids may be cultured in vitro as well as diploids. Thus in this paper the in vitro performance of haploid and the doubled haploid sugar beet of various origin was evaluated. The DHs were derived from haploids by diploidization and twelve such haploid and corresponding DH clone pairs were obtained thus the comparison included haploid and DH clones that had identical allelic composition and differed only in their ploidy level. Results: The genotypes differed in shoot morphology and...

Callus induction and plant regeneration of the endemic Astragalus nezaketae in Turkey Electron. J. Biotechnol.
Erisen,Semiha; Yorgancilar,Mustafa; Atalay,Emine; Babaoglu,Mehmet; Duran,Ahmet.
A callus induction and plant regeneration protocol was developed from leaf and petiole explants of the endemic Astragalus nezaketae. Explants were cultured on Murashige and Skoog medium (MS) supplemented with different plant growth regulators (PGRs) [?-naphthaleneacetic acid (NAA), benzyladenine (BA), 2,4-dichlorophenoxyacetic acid (2,4-D), kinetin (Kin), thidiazuron (TDZ)]. The combinations and concentrations of PGRs were shown significant variations for the frequency of callus formation, appearence of callus and the potential of callus differentiation. NAA x BA have been found highly affective in callusing and plant regeneration. Other PGRs have not resulted in callus differentiation for shoot formation. The highest number of shoots (6/explants) was...

NaCl effects in Zea mays L. x Tripsacum dactyloides (L.) L. hybrid calli and plants Electron. J. Biotechnol.
Pesqueira,Julieta; García,Maria Dina; Staltari,Sebastian; Molina,Maria del Carmen.
High salt concentrations in soils negatively affect maize growth. Techniques such as remote hybridization and in vitro selection have been extensively used to accelerate breeding processes. In order to determine the usefulness of Tripsacum to improve salt tolerance in maize, the effects of NaCl, in vitro and in vivo, were evaluated in an intergeneric hybrid (MT) obtained from crossing Zea mays with Tripsacum dactyloides. Organogenic calli, induced from immature MT hybrid embryos, were exposed to different NaCl concentrations and the survival and regeneration percentages were calculated. Plants of the MT hybrid, obtained from the organogenic calli, were exposed to NaCl concentrations considered harmful for maize. The shoot dry weights of plants exposed to...

Response of root explants to in vitro cultivation of marketable garlic cultivars Horticultura Brasileira
Scotton,Danielle C; Benedito,Vagner Augusto; Molfetta,Jeanne B de; Rodrigues,Benedita Inês FP; Tulmann-Neto,Augusto; Figueira,Antonio.
Garlic cultivars are sexually sterile under standard growth conditions, with direct implications for commercial production costs as well as breeding programs. Garlic is propagated commercially via bulblets, which facilitates disease transmission and virus load accumulation over vegetative generations. Tissue culture produces virus-free clones that are more productive, while keeping the desired traits of the cultivar. Consequently, this technique allows studies of garlic genetics as well as guarantees genetic conservation of varieties. We aimed at analyzing the in vitro regeneration of eight marketable cultivars of garlic using root segments as explants. For each genotype, bulblet-derived explants were isolated and introduced into MS medium supplemented...

Bellon, H.
The formation of the limbs, the digestive tract, the nervous system, the sensory and hormonal structures begins in this prawn during the embryonic stage C, according to Richard's definition (1974) and is performed at the stage H. The digestive tract differentiates with a caudo-cephalic gradient. The digestive gland is formed at least partially by vitellophages. The C.N.S. appears with a cephalo-caudal gradient and the regions of the eyestalk are quickly differentiated. The organ of Bellonci and the sensory pore appear independently during the stage F. The first neurosecretory cells are detected in the brain during the stage D.

Divergence and convergence in early embryonic stages of metazoans Naturalis
Galis, Frietson; Sinervo, Barry.
The early stages of organogenesis in metazoans differ drastically between higher order taxa such as phyla and classes. The segmented germ band stage in insects, the nauplius stage of crustaceans, and the neurula/pharyngula stage in vertebrates are examples of this diversification. In striking contrast with this divergence, is the similarity of these stages within these taxa, i.e., within insects, crustaceans, and vertebrates. The early stages of organogenesis, or phylotypic stages, have, thus, remained very similar in most species since the evolutionary origin of the taxa. These phylotypic stages are considerably more similar to each other than to the earlier stages of cleavage and gastrulation. Cleavage and gastrulation stages display not only great...

Physiological and biochemical changes during organogenesis and somatic embryogenesis of HBsAg-transgenic cherry tomato mutant Phyton
Guan,Z-J; Lu,S-B; Huo,Y-L; Guo,B; Guan,Z-P; Wei,Y-H.
Leaf explants of the HBsAg-transgenic cherry tomato (Solanum lycopersicum) mutant were cultured on Murashige and Skoog (MS) basal medium, supplemented with 1.0 mg/L 6-BA and 0.05 mg/L IAA for callus induction, to clarify the physiological and biochemical characteristics of morphogenesis development. Therefore, the physiological and biochemical changes during the development of organogenic shoots and somatic embryos in the mutant were studied. Superoxide dismutase (SOD) activities of the mutant had only one peak value on the 21st day. Peroxidase (POD) activities of the mutant declined less sharply since the explants were cultured. IAA oxidase activity of the mutant increased steadily until 42 days from culturing and then decreased sharply. Malondialdehyde...

Plant regeneration of Mullein Nightshade (Solanum donianum Walp.) from leaf explants Phyton
O'Connor-Sánchez,A; Domínguez-May,AV; Keb-Llanes,MA; Peña-Ramírez,YJ; Herrera-Valencia,VA.
A reliable protocol for plant regeneration of Mullein Nightshade (Solanum donianum Walp.) was developed from in vitro leaf explants. They were cultured on Murashige and Skoog semisolid medium, supplemented with several combinations of zeatin riboside and 1-naphthaleneacetic acid for shoot regeneration. The formulation that originated most shoots per explant was 8.5 µM zeatin riboside in absence of 1-naphthalene acetic acid. Explants with shoots were transferred to Murashige and Skoog medium, with half the normal salt concentration and without plant growth regulators for elongation. Elongated shoots were individualized, and they rooted readily in half-strength Murashige and Skoog medium without plant growth regulators. The regenerated plants, which were...

Biotechnology applied to Annona species: a review Rev. Bras. Frutic.
Encina,Carlos Lopez; Martin,Elisabeth Carmona; Lopez,Antonio Arana; Padilla,Isabel Maria Gonzalez.
Annonaceae is an ancient family of plants including approximately 50 genera growing worldwide in a quite restricted area with specific agroclimatic requirements. Only few species of this family has been cultivated and exploited commercially and most of them belonging to the genus Annona such as A. muricata, A. squamosa, the hybrid A. cherimola x A. squamosa and specially Annona cherimola: the cherimoya, commercially cultivated in Spain, Chile, California, Florida, México, Australia, Ecuador, Peru, Brazil, New Zealand and several countries in South and Central America. The cherimoya shows a high degree of heterozygosis, and to obtain homogeneous and productive orchards it is necessary to avoid the propagation by seeds of this species. Additionally, the...

Sour orange bud regeneration and in vitro plant development related to culture medium composition and explant type Rev. Bras. Frutic.
Silva,Rosely Pereira da; Souza,Amancio José de; Mendes,Beatriz Madalena Januzzi; Mourão Filho,Francisco de Assis Alves.
In order to evaluate the formation of adventitious buds and in vitro regeneration of sour orange plants (Citrus aurantium L.) two organogenesis-inducing experiments were conducted. In the first experiment, the induction and in vitro regeneration of adventitious buds were tested on epicotyl and internodal segments under the influence of BAP or KIN associated with NAA. The second experiment evaluated the in vitro regeneration of sour orange plants related to different explant types (epicotyls segments, internodal segments of in vitro germinated plantlets and internodal segments of greenhouse cultivated plants). Data collected on both experiments included the percentage of responsive explants (explants that formed buds), and the number of buds per explant....

Plant regeneration from cotyledonary explants of Eucalyptus camaldulensis Scientia Agricola
Dibax,Roberson; Eisfeld,Cristiane de Loyola; Cuquel,Francine Lorena; Koehler,Henrique; Quoirin,Marguerite.
Breeding methods based on genetic transformation techniques need to be implemented for Eucalyptus camaldulensis to shorten the long breeding cycles and avoid manipulation of adult trees; that requires the development of plant regeneration protocols enabling development of plants from transformed tissues. The present work aimed to optimise the regeneration process already established for the species. Cotyledonary leaves of E. camaldulensis were cultured in MS medium supplemented with naphthaleneacetic acid (NAA) and 6-benzylaminopurine (BAP) combinations. The most efficient treatment for bud indirect regeneration (2.7 µmol L-1 NAA and 4.44 µmol L-1 BAP) was used for further experiments. When explants were kept in the dark during the first 30 days, the...

It is possible to determine the optimum time for permanence of vegetative propagules (mini-cuttings) inside a greenhouse for rooting, and this value can be used to optimize the structure of the nursery. The aim of this study was to determine the dynamics of adventitious rooting in mini-cuttings of three clones of Eucalyptus benthamii x Eucalyptus dunnii. Sprouts of H12, H19 and H20 clones were collected from mini-stumps that were planted in gutters containing sand and grown in a semi-hydroponic system. The basal region of the mini-cuttings was immersed in 2,000 mg L-1 indole-3-butyric acid (IBA) solution for 10 seconds. The rooting percentage of the mini-cuttings, the total length of the root system and the rooting rate per mini-cutting were also evaluated...

Stable and efficient Agrobacterium tumefaciens mediated transformation of Phaseolus vulgaris Agrociencia
Espinosa-Huerta,Elsa; Quintero-Jiménez,Anareli; Cabrera-Becerra,K. Virginia; Mora-Avilés,M. Alejandra.
Common bean breeding requires novel techniques to incorporate genes that may not be available in bean genetic diversity. The objective of the present study was to analyze the main variables involved in Agrobacterium tumefaciens-mediated transformation of common bean towards providing an efficient system to introduce new agronomic traits. Bean cultivars Flor de Mayo Anita and Pinto Saltillo were inoculated with different A. tumefaciens strains, constructs and selection was done using two selection agents (kanamycin ot glufosinateammonium herbicide). Five-day old hypocotyls regenerated organogenic buds (cell clusters) 5 d after inoculation. A randomized complete blocks experimental design was used and the experimental unit was a petri dish with at least 10...

An improved method for in vitro regeneration of common bean (Phaseolus vulgaris L.) Agrociencia
Quintero-Jiménez,Anareli; Espinosa-Huerta,Elsa; Acosta-Gallegos,J. Alberto; Guzmán-Maldonado,H. Salvador; Mora-Avilés,M. Alejandra.
In vitro regeneration of common bean (Phaseolus vulgaris L.) is a requirement for genetic transformation which involves induction and development to the whole plant. Success in regeneration of common bean from tissue organ culture has been achieved to some extent in the last years. However, genotype effects in regeneration response as well as efficiency and reproducibility have been a limiting factor. Embryos, from four different varieties, were excised from sterilized mature seeds and cultured in Murashige and Skoog (MS) or Gamborg (GM) media containing 6-benzylaminopurine (BAP) (10 mg-1) and adenine (A) (0 or 20 mg-1). Efficient regeneration was achieved when inducing formation of differentiation of cells like bud clusters at the internodal segment of...
