Some arachnidan peptides with potential medical application J. Venom. Anim. Toxins incl. Trop. Dis.
De Lima,ME; Borges,MH; Verano-Braga,T; Torres,FS; Montandon,GG; Cardoso,FL; Peixoto,KDBA; Cardoso-Jr,HC; Benelli,MT.
The search for new active drugs that can alleviate or cure different diseases is a constant challenge to researchers in the biological area and to the pharmaceutical industry. Historically, research has focused on the study of substances from plants. More recently, however, animal venoms have been attracting attention and studies have been successful in addressing treatment of accidents. Furthermore, venoms and their toxins have been considered good tools for prospecting for new active drugs or models for new therapeutic drugs. In this review, we discuss some possibilities of using different toxins, especially those from arachnid venoms, which have shown some potential application in diseases involving pain, hypertension, epilepsy and erectile dysfunction....

Spider venom peptides as potential drug candidates due to their anticancer and antinociceptive activities J. Venom. Anim. Toxins incl. Trop. Dis.
Wu,Ting; Wang,Meng; Wu,Wenfang; Luo,Qianxuan; Jiang,Liping; Tao,Huai; Deng,Meichun.
Abstract Spider venoms are known to contain proteins and polypeptides that perform various functions including antimicrobial, neurotoxic, analgesic, cytotoxic, necrotic, and hemagglutinic activities. Currently, several classes of natural molecules from spider venoms are potential sources of chemotherapeutics against tumor cells. Some of the spider peptide toxins produce lethal effects on tumor cells by regulating the cell cycle, activating caspase pathway or inactivating mitochondria. Some of them also target the various types of ion channels (including voltage-gated calcium channels, voltage-gated sodium channels, and acid-sensing ion channels) among other pain-related targets. Herein we review the structure and pharmacology of spider-venom peptides that...

Fish welfare: the state of science by scientometrical analysis Biological Sciences
Ghisi, Nédia de Castilhos; Oliveira, Elton Celton de.
In 2014, Brazil produced 474.33 thousand tons of captive-bred fish. In addition, regulatory agencies of animal ethics and welfare have recently encouraged experiments to be done using simpler vertebrates, such as fish. The aim of this article was to perform a scientometric analysis of scientific production that deals with fish welfare, in an attempt to find trends and gaps in this line of research. Our analyses showed a growing concern about fish welfare, although several questions remained inadequately covered. The most studied species was the Atlantic salmon, with Norway having the most publications on this theme. There are controversies among scientists about fish capacity for suffering and enjoyment (sentience). As regards slaughter or euthanasia,...

Subarachnoid injection of ifenprodil and ketamine association improves the anti-hyperalgesic action of ketamine in dogs Arq. Bras. Med. Vet. Zootec.
Rondon,E.S.; Valadão,C.A.A.; Parada,C.A..
To test clinically whether a small dose of ifenprodil can enhance the anti-hyperalgesic effect of ketamine in dogs, a prospective randomized cross-over study was done with eight mongrel dogs (weighing 16.9 ± 3.7kg). Animals received two distinct treatments: ketamine (0.3mg kg-1; KT) and an ifenprodil plus ketamine combination (0.03mg kg-1 and 0.3mg kg-1, respectively; IKT). Dogs were anesthetized with propofol (5mg kg-1 intravenously) and a subarachnoid needle was placed between the 5th and 6th lumbar vertebrae. Five minutes after subarachnoid injection of KT or IKT, an incision including cutaneous and subcutaneous tissues was made on the common pad of one hind limb and was immediately closed with a simple interrupted suture pattern. The dogs were treated...

Macêdo,Maíra Carvalho; Mota,Renata de Sousa; Patrício,Naiane Araújo; Santos,Ana Paula Campos dos; Mendes,Selena Márcia Dubois; Dias,Cristiane Maria Carvalho Costa; Baptista,Abrahão Fontes; Sá,Katia Nunes.
Abstract HTLV-1 creates a chronic health condition that involves moderate to severe pain with a negative impact on quality of life (QoL). There is no consensus on which attitudes to pain are more related to the worsening of QoL in HTLV-1 infected patients. The aim of this study was to investigate the correlation between QoL and multidimensional aspects of pain in patients with HTLV-1. A cross-sectional study was conducted in Salvador, Bahia, Brazil. The study included individuals diagnosed with HTLV-1. The Short Form 36 Questionnaire was used to analyze QoL, and the Brief Pain Inventory was used to assess multidimensional aspects of pain. The mean pain intensity was 4.88 ± 3.06 on the visual pain scale, and the average impact on QoL corresponded to a loss...

An interleukin-33/ST2 signaling deficiency reduces overt pain-like behaviors in mice BJMBR
Magro,D.A.C.; Hohmann,M.S.N.; Mizokami,S.S.; Cunha,T.M.; Alves-Filho,J.C.; Casagrande,R.; Ferreira,S.H.; Liew,F.Y.; Cunha,F.Q.; Verri Jr,W.A..
Interleukin (IL)-33, the most recent member of the IL family of cytokines, signals through the ST2 receptor. IL-33/ST2 signaling mediates antigen challenge-induced mechanical hyperalgesia in the joints and cutaneous tissues of immunized mice. The present study asked whether IL-33/ST2 signaling is relevant to overt pain-like behaviors in mice. Acetic acid and phenyl-p-benzoquinone induced significant writhing responses in wild-type (WT) mice; this overt nociceptive behavior was reduced in ST2-deficient mice. In an antigen-challenge model, ST2-deficient immunized mice had reduced induced flinch and licking overt pain-like behaviors. In the formalin test, ST2-deficient mice also presented reduced flinch and licking responses, compared with WT mice. Naive WT...

Infrared coagulation versus rubber band ligation in early stage hemorrhoids BJMBR
Gupta,P.J..
The ideal therapy for early stages of hemorrhoids is always debated. Some are more effective but are more painful, others are less painful but their efficacy is also lower. Thus, comfort or efficacy is a major concern. In the present randomized study, a comparison is made between infrared coagulation and rubber band ligation in terms of effectiveness and discomfort. One hundred patients with second degree bleeding piles were randomized prospectively to either rubber band ligation (N = 54) or infrared coagulation (N = 46). Parameters measured included postoperative discomfort and pain, time to return to work, relief in incidence of bleeding, and recurrence rate. The mean age was 38 years (range 19-68 years). The mean duration of disease was 17.5 months...

Central release of nitric oxide mediates antinociception induced by aerobic exercise BJMBR
Galdino,G.S.; Duarte,I.D.; Perez,A.C..
Nitric oxide (NO) is a soluble gas that participates in important functions of the central nervous system, such as cognitive function, maintenance of synaptic plasticity for the control of sleep, appetite, body temperature, neurosecretion, and antinociception. Furthermore, during exercise large amounts of NO are released that contribute to maintaining body homeostasis. Besides NO production, physical exercise has been shown to induce antinociception. Thus, the present study aimed to investigate the central involvement of NO in exercise-induced antinociception. In both mechanical and thermal nociceptive tests, central [intrathecal (it) and intracerebroventricular (icv)] pretreatment with inhibitors of the NO/cGMP/KATP pathway (L-NOArg, ODQ, and...

Relationship of inflammatory markers and pain in patients with head and neck cancer prior to anticancer therapy BJMBR
Oliveira,K.G.; von Zeidler,S.V.; Lamas,A.Z.; de Podestá,J.R.V.; Sena,A.; Souza,E.D.; Lenzi,J.; Lemos,E.M.; Gouvea,S.A.; Bissoli,N.S..
Pain is a common symptom in patients with cancer, including those with head and neck cancer (HNC). While studies suggest an association between chronic inflammation and pain, levels of inflammatory cytokines, such as C-reactive protein (CRP) and tumor necrosis factor-alpha (TNF-α), have not been correlated with pain in HNC patients who are not currently undergoing anticancer treatment. The purpose of this study was to examine the relationship between these inflammatory markers and perceived pain in HNC patients prior to anticancer therapy. The study group consisted of 127 HNC patients and 9 healthy controls. Pain was assessed using the Brief Pain Inventory (BPI), and serum levels of CRP and TNF-α were determined using the particle-enhanced turbidimetric...

An acid-sensing ion channel that detects ischemic pain BJMBR
Naves,L.A.; McCleskey,E.W..
Ischemic pain occurs when there is insufficient blood flow for the metabolic needs of an organ. The pain of a heart attack is the prototypical example. Multiple compounds released from ischemic muscle likely contribute to this pain by acting on sensory neurons that innervate muscle. One such compound is lactic acid. Here, we show that ASIC3 (acid-sensing ion channel #3) has the appropriate expression pattern and physical properties to be the detector of this lactic acid. In rats, it is expressed only in sensory neurons and then only on a minority (~40%) of these. Nevertheless, it is expressed at extremely high levels on virtually all dorsal root ganglion sensory neurons that innervate the heart. It is extraordinarily sensitive to protons (Hill slope 4,...
