Viability of probiotic bacteria and some chemical and sensory characteristics in cornelian cherry juice during cold storage Electron. J. Biotechnol.
Nematollahia,Amene; Sohrabvandi,Sara; Mortazavian,Amir Mohammad; Jazaeri,Sahar.
Background: Increased popularity of vegetarianism, lactose intolerance, and the high cholesterol content in dairy products, are all factors that have recently increased the demand for nondairy probiotic products. The objective of this study is to evaluate the effect of refrigeration on the viability of probiotics and asses some of the chemical and sensory characteristics in cornelian cherry juice. Results: The Iranian native probiotic strain (L. casei T4) showed greater viability compared to industrial types (viable count of 8.67 log cfu/mL versus <6.0 log cfu/mL at d 28). However, this most tolerant Iranian strain, could not withstand the conditions of 'Natural juice' at pH 2.6 for more than 7 d. Following a pH adjusted treatment (to pH ~3.5), the...

Effect of dietary probiotic Pediococcus acidilactici on antioxidant defences and oxidative stress status of shrimp Litopenaeus stylirostris ArchiMer
Castex, Mathieu; Lemaire, Pierrette; Wabete, Nelly; Chim, Liet.
in this study we evaluated the effects of the probiotic Pediococcus acidilactici MA18/5M on shrimp, Litopenaeus stylirostris (also called Penaeus stylirostris), first on antioxidant defences and secondly on the oxidative stress status in the shrimps' haemolymph and digestive gland. We conducted two experiments with the same protocol in which shrimps were fed two diets for three weeks: a control diet and a probiotic diet containing 1 g of live P. acidilactici MA18/5M kg(-1). In the first experiment, the shrimps were found to be healthy over the trial period; no mortalities and non-detectable signs of infection were recorded. These resulted in high final survival rates (above 90% in both treatments). On the other hand, during the second trial, carried out at...

Selection and characterization of potential probiotic bacteria for Litopenaeus stylirostris shrimp hatcheries in New Caledonia ArchiMer
Pham, Dominique; Ansquer, Dominique; Chevalier, Anne; Dauga, Clement; Peyramale, Aude; Wabete, Nelly; Labreuche, Yannick.
In New Caledonia, shrimp hatcheries are confronted with mass mortality in the larval stages, a phenomenon poorly understood as no specific causative agent has been identified. This has resulted in an excessive use of prophylactic antibiotics, although their adverse effects in aquaculture are notorious. The present work was thus aimed at selecting potential probiotic strains for penaeid hatcheries. From a pool of more than 400 marine bacterial isolates sampled from the local marine environment seven strains exhibited in vitro antagonistic activity towards Vibrio harveyi. These isolates were characterized both phenotypically and genotypically using a biochemical approach and 16S rDNA sequencing. Six out of these seven strains were found to belong to the...

Utilisation de bactéries lactiques probiotiques pour prémunir les poissons d'élevage contre des vibrions pathogènes ArchiMer
Lamari, Faouzi.
Diseases cause major production losses in fish farms. Bacteria of the genus Vibrio are known as opportunistic pathogens, and have been associated with mass-mortality episodes. To cope with these disease outbreaks, fish farmers often resort to the use of antibiotics. However, preventive overuse has led to the emergence of resistant strains and frequent treatment failures. Probiotic lactic acid bacteria are increasingly used to improve the quality of seed production in aquaculture, while limiting antibiotic treatments. Microbial strains must fulfil several criteria to be evaluated as potential probiotics. In a first step, we selected and characterized 55 strains of lactic acid bacteria isolated from fish hatchery. The first selection criterion focused on the...

Probiotic effect of FLOC on Vibrios in the pacific white shrimp Litopenaeus Vannamei ArchiMer
Aguilera-rivera, Diana; Prieto-davo, Alejandra; Escalante, Karla; Chavez, Cristina; Cuzon, Gerard; Gaxiola, Gabriela.
The advantages of FLOC over clear water (CW) in rearing juvenile L. vannamei and its effects on Vibrio communities were evaluated. Survival rate in FLOC and clear water were recorded and a probiotic was tested under both conditions. Daily growth rate (DGR) was higher in FLOC (p < 0.05) than in CW. Survival in each system increased significantly when a probiotic was included in the diet (p < 0.05). The Vibrionaceae community from the hepatopancreas (HP) and the culture medium did not differ between the two culture media. Nevertheless, a novel group of Vibrio strains was found to be unique to FLOC. No high level of lesions was observed in shrimp tissues from the FLOC + probiotic treatment; it suggests that the probiotic contributed to homeostasis and...

Probiotic P-acidilactici application in shrimp Litopenaeus stylirostris culture subject to vibriosis in New Caledonia ArchiMer
Castex, Mathieu; Chim, Liet; Pham, Dominique; Lemaire, Pierrette; Wabete, Nelly; Nicolas, Jean-louis; Schmidely, P; Mariojouls, C.
We studied the effects of a lactic acid bacterium, Pediococcus acidilactici (strain MA 18/5M, CNCM), as a dietary probiotic on growth performance and some nutritional and microbiological aspects of the shrimp Litopenaeus stylirostris. Experimental shrimp culture was carried out over 10 weeks, using floating cages of 14 m(2) each set in earthen ponds, in a New Caledonia farm affected by "Summer syndrome", a septicaemic vibriosis caused by Vibrio nigripulchritudo. The experiment design testing probiotic-coated pellets against control was replicated in two earthen ponds. High mortalities, characteristic of Summer syndrome, were observed during the first 2 weeks of the trial. The probiotic improved production in the treated cages from both ponds with increases...

Effect of probiotic Pediococcus acidilactici on antioxidant defences and oxidative stress of Litopenaeus stylirostris under Vibrio nigripulchritudo challenge ArchiMer
Castex, Mathieu; Lemaire, Pierrette; Wabete, Nelly; Chim, Liet.
Antioxidant defences and induced oxidative stress tissue damage of the blue shrimp Litopenaeus stylirostris, under challenge with Vibrio nigriputchritudo, were investigated for a 72-h period. For this purpose, L stylirostris were first infected by immersion with pathogenic V. nigripulchritudo strain SFn1 and then antioxidant defences: superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (Gpx), Total antioxidant status (TAS), glutathiones and induced tissue damage (MDA and carbonyl proteins) were determined in the digestive gland at 0, 12, 24, 48 and 72 h post-infection (h.p.i.). In the meantime, TAS was also measured in the blood. Infection level of the shrimps during the challenge was followed by determining V. nigripulchritudo prevalence...

Effects of a mix of Bacillus sp. as a potential probiotic for Florida pompano, common snook and red drum larvae performances and digestive enzyme activities ArchiMer
Hauville, Marion; Zambonino-infante, J. L.; Bell, J. Gordon; Migaud, H.; Main, K. L..
This study examined the effect of a commercial mix of Bacillus sp. on survival, growth and digestive enzyme activities of Florida pompano, red drum and common snook. Larvae were fed either live feed enriched with Algamac 3050 (Control), Algamac 3050 and probiotics (PB), or the previous diet combined with a daily addition of probiotics to the tank water (PB+). Survival was not affected by the treatments for any of the species. At the end of the pompano and snook trial, standard lengths of larvae from the PB and PB+ treatments were significantly greater than for the control larvae. Microbiological analyses were performed at the end of the pompano trial, and numbers of presumptive Vibrio were not a concern in the system. For both pompano and snook,...

Probiotic and formaldehyde treatments of Artemia nauplii as food for larval pollack, Pollachius pollachius ArchiMer
Gatesoupe, Joel.
Formaldehyde was used to disinfect Artemia cysts and nauplii, while introducing two probiotics in the enrichment process: Bactocell (Pediococcus acidilactici) and Levucell (Saccharomyces cerevisiae). The disinfectant was selected due to its potential compatibility with probiotics, since it was more effective against Gram-negative bacteria than against lactic acid bacteria and yeast. However, the presence of formaldehyde reduced the intake of P. acidilactici in Artemia. Consequently, the disinfection was stopped before Bactocell supplementation to the nauplii fed to pollack (Pollachius pollachius) larvae. The mean weight of pollack was higher with this probiotic treatment. Growth was even better with the combination of Levucell and Bactocell, but the yeast...

Updating the importance of lactic acid bacteria in fish farming: natural occurrence and probiotic treatments ArchiMer
Gatesoupe, Joel.
Many recent papers have deepened the state of knowledge about lactic acid bacteria (LAB) in fish gut. In spite of high variability in fish microbiota, LAB are sometimes abundant in the intestine, notably in freshwater fish. Several strains of Streptococcus are pathogenic to fish. S. iniae and Lactococcus garvieae are major fish pathogens, against which commercial vaccines are available. Fortunately, most LAB are harmless, and some strains have been reported for beneficent effects on fish health. A major step forward of the recent years was those converging evidences that LAB can stimulate the immune system in fish. An open question is whether viability can affect immunostimulation. The issue is crucial to commercialise live probiotics rather than...

Occurrence and role of lactic acid bacteria in seafood products ArchiMer
Leroi, Francoise.
Lactic acid bacteria (LAB) in fish flesh has long been disregarded because the high post-mortem pH, the low percentage of sugars, the high content of low molecular weight nitrogenous molecules and the low temperature of temperate waters favor the rapid growth of pH-sensitive psychrotolerant marine Gram-negative bacteria like Pseudomonas, Shewanella and Photobacterium. In seafood packed in both vacuum (VP) and modified atmosphere (MAP) packaging commonly CO2 enriched, the growth of the Gram-negative aerobic bacteria group (predominantly pseudomonads) is effectively inhibited and the number reached by LAB during storage is higher than that achieved in air but always several log units lower than the trimethylamine oxide (TMA-O) reducing and CO2-resistant...

Evaluation du probiotique bactérien Pediococcus acidilactici MA18/5M chez la crevette pénéide Litopenaeus stylirostris en Nouvelle- Calédonie ArchiMer
Castex, Mathieu.
P. acidilactici MA 18/5M is a dietary probiotic (BACTOCELL®) which has shown its efficiency on several terrestrial and aquatic species. The present thesis aimed to investigate more fully the effects of this probiotic on cultured penaeid shrimps. For this study, we chose a global approach by replacing the problem studied in a model of interactions between three compartments: the shrimp, its intestinal microflora and the probiotic. Having first defined how the probiotic should be administrated, we studied its effects under controlled laboratory conditions at several levels of Litopenaeus stylirostris biology: (i) Nutritional status and growth, (ii) associated bacterial microflora, and (iii) health status with respect to oxidative stress. Then the probiotic...

The objective of this work was to assess the time-related action of probiotic Lactobacillus plantarum in the bacterial microbiota of the digestive tract of Litopenaeus vannamei, and the relation of total haemocyte count and serum phenol oxidase activity of shrimp challenged with Vibrio harveyi. Shrimps were fed with a probiotic-supplemented diet, for eight days, then shifted to a commercial diet. Shrimps fed only with the commercial diet served as control. Evaluations were made on the 8th day of experiment and repeated two, four, six and eight days later. Total lactic bacteria in the digestive tract was higher until the 4th day of evaluation in the probiotic-supplemented group. Vibrio spp. counts were higher in the control at days zero and two. Until the...
