Variation in growth at different temperatures and production and size of conidia in hybrid strains of Verticillium lecanii (Lecanicillium spp.) (Deuteromycotina: Hyphomycetes) OAK

In a previous study, we confirmed alterations in several fungal traits in hybrid strains of Verticillium lecanii generated via protoplast fusion. In the present study, we surveyed radial colony growth in different temperature regimes as well as conidia productivity and conidia size of hybrid strains of V. lecanii to elucidate the various primary fungal properties of these strains and to determine the correlation between these properties and virulence to insect pests. Minimum temperature thresholds for all tested strains (except Vertalec®) were below 5°C. Optimal temperatures for colony growth were 20–25°C. Pathogenicity and its mode of action in different sedentary stages of Heterodera glycines (Tylenchida: Heteroderidae) by Verticillium lecanii hybrid strains OAK

The current study was conducted to investigate the pathogenicity and its mode of action by Verticillium lecanii hybrid strains in different sedentary stages of Heterodera glycines. Three different sedentary stages (pale yellow female, yellow brown cyst, and dark brown cyst) of H. glycines were treated with V. lecanii and incubated for 3 weeks on water agar. After 3 weeks of incubation, eggs were investigated for the following parameters: (i) the infection frequencies of eggs, (ii) the number of eggs laid, and (iii) the number of mature and healthy eggs. Screening of Verticillium lecanii (Lecanicillium spp.) Hybrid Strains Based on Evaluation of Pathogenicity against Cotton Aphid and Greenhouse Whitefly, and Viability on the Leaf Surface OAK

In a previous study, protoplast fusion was performed among 3 strains of the entomopathogenic fungi Verticillium lecanii, as a result of which 174 hybrid strains were obtained. The aim of the present study was to screen desirable hybrid strains that have a wide host range or durable effects. Initially, 43 hybrid strains were used in bioassays against the cotton aphid. Of these strains, 30 induced equal or higher mortality as compared to Vertalec (41.9%). Secondly, 50 hybrid strains were used in bioassays against the greenhouse whitefly. Of these strains, 37 exhibited an equal or higher infection rate as compared to that of Mycotal (36.2%). Somatic hybridization between Citrus sinensis (L.) Osbeck and C. grandis (L.) Osbeck PAB

This work had as objective to produce citrus somatic hybrids between sweet oranges and pummelos. After chemical fusion of sweet orange embryogenic protoplasts with pummelo mesophyll-derived protoplasts, plants were regenerated by somatic embryogenesis and acclimatized in a greenhouse. The hybrids of 'Hamlin' sweet orange + 'Indian Red' pummelo and 'Hamlin' sweet orange + 'Singapura' pummelo were confirmed by leaf morphology, chromosome counting and molecular analysis. These hybrids have potential to be used directly as rootstocks aiming blight, citrus tristeza virus, and Phytophthora-induced disease tolerance, as well as for rootstocks improvement programs.

Citrus asymmetric somatic hybrids produced via fusion of gamma-irradiated and iodoacetamide-treated protoplasts PAB

The objective of this study was to produce citrus somatic asymmetric hybrids by fusing gamma-irradiated protoplasts with iodoacetamide-treated protoplasts. Protoplasts were isolated from embryogenic suspension cells of grapefruit (Citrus paradisi Macfad.) cultivars Ruby Red and Flame, sweet oranges (C. sinensis Osbeck) 'Itaboraí', 'Natal', Valencia', and 'Succari', from 'Satsuma' (C. unshiu Marcow.) and 'Changsha' mandarin (C. reticulata Blanco) and 'Murcott' tangor (C. reticulata x C. sinensis). Characterization of fusion products from protoplasts of yeasts and their segregants by electrophoretic karyotyping and RAPD Rev. Microbiol.

In order to characterize fusion products from yeast protoplasts and their segregants, with important features to the wine making industry, electrophoretic karyotyping and RAPD (Random Amplified Polymorphic DNA) were utilized. Electrophoretic karyotyping was performed by the CHEF ("contour-clamped homogeneous electric field electrophoresis") method, which allowed the detection of chromosomal band complementation in fusion products and the presence of patterns of both parental and intermediary strains in segregants. By utilizing two primers, an amplification pattern of DNA fragments was obtained. Isolation of recombinant strains with enhanced pectinase production by protoplast fusion between Penicillium expansum and Penicillium griseoroseum BJM

Protoplast fusion between complementary auxotrophic and morphological mutant strains of Penicillium griseoroseum and P. expansum was induced by polyethylene glycol and calcium ions (Ca2+). Fusant strains were obtained in minimal medium and a prototrophic strain, possibly diploid, was chosen for haplodization with the fungicide benomyl. Different recombinant strains were isolated and characterized for occurrence of auxotrophic mutations and pectinolytic enzyme production. The fusant prototrophic did not present higher pectinase production than the parental strains, but among 29 recombinants analyzed, four presented enhanced enzyme activities.
