Genetic diversity analysis of rice cultivars (Oryza sativa L.) differing in salinity tolerance based on RAPD and SSR markers Electron. J. Biotechnol.

Thirty rice cultivars were evaluated for salinity tolerance during the seedling stage and were divided into five tolerance groups including tolerant (T), moderately tolerant (MT), moderately susceptible (MS), susceptible (S) and highly susceptible (HS) which comprised 5, 10, 9, 4 and 2 cultivars respectively. Genetic diversity of all rice cultivars was evaluated using random amplified polymorphic DNA (RAPD) and simple sequence repeats (SSR) markers. The cultivars were evaluated for polymorphisms after amplification with 20 random decamer primers and 20 SSR primer pairs. The RAPD technique associated with restriction digestion of genomic DNA was used to assess the genetic variability within and among nine populations of Coffea arabica, including six progenies belonging to the Sarchimor germplasm, the progeny PR 77054-40-10 (Catuaí Vermelho IAC 81 x Icatu), and two commercial cultivars (IAPAR 59 and Catuaí Vermelho IAC-81). These populations were evaluated using analysis of molecular variance (AMOVA), genetic similarity among progenies, and percentage of polymorphic loci. A total of 99 RAPD markers were evaluated of which 67 (67.67%) were polymorphic. AMOVA showed that 38.5% and 61.5% of the genetic variation was distributed among and within populations, respectively. Genetic variation is a key component for improving a stock through selective breeding programs. Randomly amplified polymorphic DNA (RAPD) markers were used to assess genetic variation in three wild population of the catla carp (Catla catla Hamilton 1822) in the Halda, Jamuna and Padma rivers and one hatchery population in Bangladesh. Five decamer random primers were used to amplify RAPD markers from 30 fish from each population. Thirty of the 55 scorable bands were polymorphic, indicating some degree of genetic variation in all the populations. Population structure analysis of Pimelodus maculatus (Pisces, Siluriformes) from the Tietê and Paranapanema Rivers (Brazil) Genet. Mol. Biol.

Pimelodus maculatus populations from the Tietê and Paranapanema rivers were sampled and had their genetic structure analyzed by using RAPD markers, with the aim of contributing to future conservation studies. The proportion of polymorphic loci was greater than 50% in the populations of both rivers. Genetic diversity data showed that, in spite of its nine hydroelectric plants, the Tietê river population was genetically homogeneous, whereas the Paranapanema river population was structured. This might be due to the presence of high waterfalls distributed all along its course. These data may serve as indicators for future conservation studies on the Tietê and Paranapanema rivers.

This study reports on 156 specimens of the amphibian Eupemphix nattereri, a widely distributed leiuperid, obtained from 11 municipalities of central Brazil. The extent of genetic variation was quantified by determining the mean number of alleles per locus and the proportion of polymorphic loci. An analysis of molecular variance (AMOVA) was performed on the random amplified polymorphic DNA (RAPD) haplotypes. To assess genetic structure and phenotypic diversity of Eupemphix nattereri Steindachner, 1863, morphometric and molecular analyses were carried out for nine populations from the State of Goiás. A total of 11 morphometric traits were evaluated and genetic information was estimated using RAPD markers. Genetic and phenotypic distances were determined as a function of geographical origin. Correlation among genetic, morphometric, micro, and macroenviromental were analyzed by the Mantel test. Genetic data indicated high levels of genetic diversity (Φst= 0.3) among the nine populations. Somaclonal variation in Cereus peruvianus Mill. (Cactaceae): its potential to generate new varieties and broaden the species's genetic basis JBAG

In current study Random Amplified DNA-Polymorphism markers were used to detect genetic variations in C. peruvianus somaclones and to examine relationships among somaclones and plants grown from seeds in cultivated populations. The possible association of DNA fragments with morphological traits of somaclones was also investigated. Polymorphism in somaclones (62.24%) was higher than that detected in cultivated plants (42.57%). The casual grouping of somaclones presented atypical and typical shoot phenotypes showed that the random amplifi ed DNA-fragment polymorphisms detected in present study were not associated with shoot morphologies. Abstract: The objective of this work was to evaluate the use of RAPD and ISSR molecular markers to determine the genetic variability among cashew (Anacardium spp.) genotypes, and to indicate possible promising crosses based on cashew genetic variability and phenotypic performance. Ten hybrids - derived from the crosses CCP 76 x BGC 589, CCP 76 x BRS 226, CCP 76 x HAC 276-1, CCP 76 x Embrapa 51, CCP 76 x BRS 253, CCP 76 x HAC222-4, and BRS226 x Embrapa 51 - and their parents were assessed at the molecular level. Genetic distances in soybean based on RAPD markers Bragantia

Four methods were applied to determine pairwise genetic distances among five soybean genotypes which are potential genitors for a mapping population. Additionally, individual plants from the most divergent pair of genotypes were evaluated by the RAPD technique to determine their degree of homozygosity. Genetic distances based on RAPD data were calculated by the modified Rogers' distance, and also by the following arithmetical complements of similarity: simple match, Nei and Li, and Gower. These genetic distances were similar, presenting a correlation coefficient ranging from 0.99 to 1.00. In this work, the part of the squash core collection, maintained in the Greek Gene Bank, was assessed using the morphological and molecular data. Sixteen incompletely classified accessions of the squash were characterized along with an evaluation of their resistance against two isolates of Fusarium oxysporum. Presence of the yeast Candida tropicalis in figs infected by the fruit fly Zaprionus indianus (dip.: Drosophilidae) BJM

In the last years, the fruit fly Zapronius indianus became the most important plague of Brazilian fig production. A fermentation process is associated with infection of the fruit by this fly. A single yeast species, Candida tropicalis, was identified in all the infected figs. The presence of one species and the low genetic variability (RAPD) of the isolates indicates an uncommonly strict interaction between C. tropicalis and Z. indianus.

Random Amplified Polymorphic DNA(RAPD)markers were used to assess the genetic diversity of 587 individuals, belonging to 22 populations of Nothofagus nervosa that were distributed through the Coastal (38°S to 41 °S) and Andes Mountains in Central-Southern Chile (36°S to 40°S). The objective of this study was to complement the genetic inferences previously determined by isozyme analysis, in order to obtain more accurate genetic diversity estimations. We scored 81.8% of the polymorphic loci of the samples tested. The average incidence of genetic polymorphism within populations was high, with values ranging between 33% and 63%.
