Differential Effect of the Route of Inoculation of Rabies Virus on NeuN Immunoreactivity in the Cerebral Cortex of Mice International Journal of Morphology

Rabies is a lethal disease caused by a neurotropic virus that produces inconspicuous morphological changes hardly observable with conventional histopathology. The fatal outcome caused by rabies could be attributed to specific biochemical changes that severely impact neuronal function. The neuronal nuclear protein (NeuN) has become a widely used neuronal marker for the research and the histopathological diagnosis of nervous system diseases. To evaluate the distribution of the protein NeuN in the motor cortex of normal and rabies-infected mice adult ICR mice were inoculated with rabies virus either intramuscularly or intracerebrally. Current knowledge on bat lyssavirus infections in their native hosts is limited and little is known about the virulence, virus dissemination and transmission among free-living insectivorous bats. The present study is a brief description of rabies virus (RABV) dissemination in tissues of a naturally infected pregnant southern yellow bat (Lasiurus ega) and its fetuses, obtained by reverse-transcriptase polymerase chain reaction (RT-PCR). The RT-PCR was positive in samples from the brain, salivary gland, tongue, lungs, heart, kidneys and liver. Bats are very interesting animals: they are the unique flying mammals, have developed a highly sophisticated echolocation system, and have become specialized to eat different types of diets. Hematophagous (vampire) bats are those specialized to feed solely on blood and have served as a source of inspiration for researchers as well as for writers. Vampire bat attacks on humans have moved from the realm of science fiction to reality in Latin America and bats (including non-hematophagous ones) have assumed an important role in the transmission of rabies virus to humans. Abstract Background Rabies is very common in People’s Republic of China. Each year thousands of people die because of this disease, but rabies diagnosed in pregnancy is very rare. Case Presentation In this study, we report the case of a pregnant woman who was infected with the rabies virus after a dog bite. The symptoms of rabies appeared in labor and she died after pregnancy. Her baby and husband did not develop the disease. Conclusion The phenomenon that the newborn infant was healthy may be related to the protective role of placenta in resisting the invasion of the rabies virus or the absence of systemic viremia. The prompt administration of vaccines and anti-rabies immunoglobulin to the infant may have also contributed to his survival.

The aim of this study was to evaluate epidemiological aspects and immune response in patients attacked by domestic and wild animals and subjected to post-exposure rabies treatment with equine serum and associated vaccine. Background Brazil holds annual nationwide public campaigns to vaccinate dogs and cats against rabies. The presence of rabies antibodies in these animals, which are among the main transmitters of rabies to humans, is a good indicator that they are immunized and protected. Methods In the present study we analyzed 834 serum samples from dogs and cats from the Southeast of Brazil (Presidente Prudente and Dracena cities), 12 months after the 2009 vaccination campaign. Rabies is a severe and lethal disease that produces a slight inflammatory response during the infection process. We analyzed the immunopathological mechanisms that occur in the central nervous system (CNS) using mice genetically selected for maximal or minimal acute inflammatory reaction (AIRmax or AIRmin). As viral samples, we adopted the antigenic variant 3 (AgV3) of rabies virus from hematophagous bats and a fixed virus strain (PV1 43/3). Titration of specific antibodies was performed using enzyme-linked immunosorbent assay (ELISA). We observed a slight increase in IgG and IgG1 isotypes in infected AIRmax mice. In this study, we compared the levels of neutralizing antibodies induced by inactivated rabies vaccine in cattle by using three alternative immunization procedures. Forty-five bovines (breed nelore) were then organized in three groups (A, B and C, with 15 animals/group). Group A received only one vaccine dose at day zero and Group B received the first dose at day zero and then another dose at day 30 (early booster). A plasmid named pSH-G was constructed with the rabies-virus G-gene insert. This plasmid was transfected into eukaryotic BHK-21 cells and its stability tested. The presence of the pSH-G plasmid was confirmed by means of polymerase chain reaction (PCR) after each of ten cell passages, and the results were positive. The stable BHK-21/pSH-G+ clone obtained can be used in the study of rabies as well as in the production of vaccines.

This analysis aimed to identify characteristics of accidents that would, probably, provoke rabies infection. A total of 14,409 survey questionnaires for surveillance of human rabies from the Brazilian Information System for Disease Notification (SINAN), from 2000 to 2005, were analyzed. Regarding demographics, it was observed that 7,377 (51.5%) of the victims were white, 4,458 (30.93%) were children and 8,008 (55.58%) were males. Urban cases were prevalent (88.10%) while dogs were the animals most frequently involved in accidents, in 11,700 cases (81.19%). Official reporting of rabies in Nigeria started in 1912. The National Veterinary Research Institute (NVRI), Vom, within the last 77 years, confirmed 4809 cases of animal rabies in Nigeria. Rabies control through immunization programs has crashed woefully and consequently, the rabies situation has become chaotic and confounding. Locally, only 2,137,615 doses of dog anti-rabies vaccines were produced between 1956 and 2005 (average 43,625 per year) by NVRI, Vom. Dog population in Nigeria is currently estimated at 8 million. Immune response after rabies oral immunization in mice BABT

The objective of this study was to formulate an anti-rabies oral vaccine from the cell culture vaccine PV TECPAR to elicit the production of antibodies against the rabies in mice. A vaccine was developed using 10(7.5) DL50/0.03 ml viral antigens homogenised in lanovaseline to facilitate oral administration. Mice were vaccinated two times for seroconversion. Sera of the vaccinated mice showed a higher level of antibody production than the control group. These results could be used to direct the development of an anti-rabies oral vaccine.
