Transcriptome sequencing and expression profiling of genes involved in the response to abiotic stress in Medicago ruthenica Genet. Mol. Biol.
Shu,Yongjun; Li,Wei; Zhao,Jinyue; Liu,Ying; Guo,Changhong.
Abstract Medicago ruthenica is a perennial forage legume with the remarkable ability to survive under unfavorable environmental conditions. It has been identified as an excellent species of Medicago that can adapt to various environmental stresses including low temperature, drought, and salinity. To investigate its potential as a genetic resource, we performed transcriptome sequencing and analysis in M. ruthenica under abiotic stresses. We generated >120 million reads from six cDNA libraries, resulting in 79,249 unique transcripts, most of which were highly similar to transcripts from M. truncatula (44,608, 56.3%) and alfalfa (M. sativa, 48,023, 60.6%). Based on gene expression profiles, 2,721 transcripts were identified as abiotic stress responsive...

Infection with the protozoan parasite Bonamia ostreae modifies in vitro haemocyte activities of flat oyster Ostrea edulis ArchiMer
Morga, Benjamin; Arzul, Isabelle; Chollet, Bruno; Renault, Tristan.
Bonamia ostreae is an intracellular protozoan parasite, infecting haemocytes of the European flat oyster Ostrea edulis. Oyster defence mechanisms mainly rely on haemocytes. In the present study in vitro interactions between parasites and flat oyster haemocytes were investigated using flow cytometry and light microscopy. Haemocyte parameters including: non specific esterase activity, reactive oxygen species (ROS) production and phagocytosis were monitored using flow cytometry after 2 hour cell incubation with live and dead B. ostreae. Two ratios of parasites per haemocyte were tested (5:1 and 10:1), haemocytes alone were used as controls and the experiment was carried out three times. Flow cytometry revealed a decrease of non specific esterase activities...

Saxitoxin Modulates Immunological Parameters and Gene Transcription in Mytilus chilensis Hemocytes ArchiMer
Astuya, Allisson; Carrera, Crisleri; Ulloa, Viviana; Aballay, Ambbar; Nunez-acuna, Gustavo; Hegaret, Helene; Gallardo-escarate, Cristian.
Saxitoxin (STX) is a neurotoxin produced by dinoflagellates in diverse species, such as Alexandrium spp., and it causes paralytic shellfish poisoning (PSP) in humans after the ingestion of contaminated shellfish. Recent studies have suggested that the immune functions of bivalves could be affected by harmful algae and/or by their toxins. Herein, hemocytes are the main effector cells of the immune cellular response. In this study, we evaluated the response of hemocytes from the mussel Mytilus chilensis to STX exposure in a primary culture. Cell cultures were characterized according to size and complexity, while reactive oxygen species (ROS) production was evaluated using a dichlorofluorescein diacetate (DCFH-DA) assay. Finally, phagocytic activity was...

Interference of Volunteer Corn from Different Origins and Emergence Time on Soybean Yield and Stress Metabolism Planta Daninha
RIZZARDI,M.A.; PIASECKI,C.; SCHONS,J.; CAVERZAN,A.; LANGARO,C..
ABSTRACT: Volunteer corn occurrence with soybean is favored by the glyphosate-resistant (GR) corn cultivation preceding soybean and no-tillage systems. Volunteer corn interference causes significant losses in soybean grain yield. The levels of crop losses change with the corn density, origin, and time of emergence. High levels of weed interference in crops can result in the production of reactive oxygen species and lead to the occurrence of oxidative stress. The objectives of this study were to evaluate the effects of interference of (1) different origins (individual plants and clumps) and times of emergence of volunteer corn on soybean growth, yield components, and grain yield loss; and (2) if the volunteer corn interference causes oxidative stress in...

The compensatory effect of glutathione on alleviating salinity–induced modulations in growth and biochemical traits in maize irrigated with diluted seawater CIGR Journal
El-Shahawy, Tarek Abd El-Ghafar.
Salinity stress has recently received much attention as an object worthy of research and interest. It is a great challenge for the future global agricultural production that aspires to a large-scale conversion of raw seawater to irrigation use. Our study aims to investigate the antioxidant and free radical scavenging effect of glutathione (GSH) that would enhance maize tolerance to the destructive effect of salinity. A greenhouse trail was conducted in this context during the summer season of 2015 using two salinity (Mediterranean seawater: 3000 and 6000 ppm) and GSH (100 and 200 pm) levels. Tap water was used as a control. Individually, saline water acted in a distinctly different manner than GSH. Irrigation with diluted seawater caused morphological...

The antioxidative defense system is involved in the premature senescence in transgenic tobacco (Nicotiana tabacum NC89) Biol. Res.
Liu,Yu; Wang,Lu; Liu,Heng; Zhao,Rongrong; Liu,Bin; Fu,Quanjuan; Zhang,Yuanhu.
BACKGROUND: α-Farnesene is a volatile sesquiterpene synthesized by the plant mevalonate (MVA) pathway through the action of α-farnesene synthase. The α-farnesene synthase 1 (MdAFS1) gene was isolated from apple peel (var. white winterpearmain), and transformed into tobacco (Nicotiana tabacum NC89). The transgenic plants had faster stem elongation during vegetative growth and earlier flowering than wild type (WT). Our studies focused on the transgenic tobacco phenotype. RESULTS: The levels of chlorophyll and soluble protein decreased and a lower seed biomass and reduced net photosynthetic rate (Pn) in transgenic plants. Reactive oxygen species (ROS) such as hydrogen peroxide (H2O2) and superoxide radicals (O2._) had higher levels...

Passion fruit (Passiflora edulis) leaf extract modulates the oxidative metabolism of rat peritoneal neutrophils in a model of inflammation BJPS
Araújo,Cleiber Lucan Alves; Salles,Bruno Cesar Correa; Duarte,Stella Maris da Silveira; Rodrigues,Maria Rita; Paula,Fernanda Borges de Araújo.
This study was conducted to evaluate the effect of extracts of Passiflora edulis Sims leaves on the oxidative metabolism of rat peritoneal neutrophils using a model of acute inflammation. The extract was obtained by maceration in 70% ethanol, evaporation under reduced pressure and lyophilisation. Total phenolic content (TP) was determined by the Folin-Ciocalteu assay. The P. edulis extract, in different doses, was administered by gavage 1 h prior to inflammation induction by carrageenan (8 mg/kg, i.p.); five hours later, the neutrophils were obtained by intraperitoneal lavage. The tests performed in neutrophils were cytochrome C and chemiluminescence assay as well as myeloperoxidase (MPO), superoxide dismutase (SOD), glutathione peroxidase (GPx) and...
