Physical maps of the ribosomal RNA gene 28S of species belonging to the genera Paratelmatobius and Scythrophrys were constructed, using five restriction endonucleases. The restriction sites for Bam HI, Bgl II, Bst EII, and Eco RI had similar positions in all species, although there were interspecific differences in the size of the restriction fragments obtained. An additional Pvu II site was found in Scythrophrys specimens from Piraquara (State of Paraná, Brazil) and from São Bento do Sul (State of Santa Catarina, Brazil), but not in the Scythrophrys specimens from Rancho Queimado (State of Santa Catarina, Brazil). Karyotypic characteristics of three species of the genus serrasalmus ( S. altispinnis , S. gouldingi and S. Serrulatus ) from the middle and lower Negro River, Amazon Basin, were investigated using different staining techniques and Fluorescent in situ hybridization with 5S and 18S rDNA probes. The diploid number was invariably 2n = 60 and the fundamental number was FN = 110. Nevertheless, the karyotypes differed from each other in composition: 24m, 20sm, 6st, 10a in S. altispinnis ; 22m, 22sm, 6st, 10a in S. gouldingi and 20m, 22sm, 8st, 10a in S. serrulatus. Abstract The genera Cichlasoma and Gymnogeophagus belong to the subfamily Cichlinae, the only one in Neotropical cichlids. Cichlasoma dimerus, C. paranaense, C. portalegrense, Gymnogeophagus rhabdotus, and G. lacustris were collected at different points in the Paranapanema and Paraguay basins and the Lagoon of Patos hydrographic system. In addition to conventional analysis, CMA3 fluorochrome staining, and FISH with 18S rDNA probe were performed. Members of Aspergillus section Nigri are distributed worldwide and are potentially ochratoxin A producers; some of them are used at industrial level as source of extracellular enzymes and organic acids. The taxonomy of this section comprises one of the most complex and confusing of genus Aspergillus. The objectives of present work were to study the phylogeny of Aspergillus section Nigri and their taxonomy by amplifying ITS1, 5.8S and ITS2 regions. Discovery of male morphological characters for species identification was a great improvement in Trichogramma systematic. However, not all species could be easily identified. In some cases, the lack of males (thelytokous status of species that carry the Wolbachia symbiont) made Trichogramma identification impossible. This problem was solved via antibiotic and heating treatments for elimination of the bacteria and allowing the production of males. ABSTRACT Farlowella is one of the most diverse genera of the Loricariinae, restricted to South America rivers. The taxonomic and phylogenetic relationships among its species are contentious and, while genetic studies would contribute to the understanding of their relationships, the only available datum refer to the karyotype description of only one species. Some species belonging to the genus Aspergillus are potential producers of ochratoxin A (OA), a mycotoxin with nephrotoxic, immunosuppressive, teratogenic and carcinogenic effects. The aim of the present study was to identify the species of Aspergillus that contaminate the inside of coffee beans collected in the stage of maturation and drying, from 16 producing areas located in the northern region of the State of Paraná, in the South of Brazil. The chromosomes of Solanum nelsonii, S. sandwicense (endemic to the Hawaii islands) and S. vespertilio (from the Canary islands) were studied by means of classical staining, CMA/DAPI banding and FISH with probes for the 18-5.8-26S and 5S rDNA genes. The aim of this study was to test for chromosomal changes (chromosome number, karyotype, heterochromatin pattern, rDNA loci) during the evolution of these taxa with respect to their continental relatives. Seven fungi were isolated from 50 samples of cosmetic powders. Morphological analyses and ribosomal DNA Internal Transcribed Spacers sequencing were performed which allowed the discrimination of the isolated fungi as Aspergillus fumigatus, Penicillium sp., and Cladosporium sp. which could have, among their species, potentially pathogenic microorganisms. |
