A quantitative PCR approach for determining the ribosomal DNA copy number in the genome of Agave tequila Weber Electron. J. Biotechnol.
Rubio-Piña,Jorge; Quiroz-Moreno,Adriana; Sánchez-Teyer,L. Felipe.
Background: Agave tequilana has a great economic importance in Mexico in order to produce alcoholic beverages and bioenergy. However, in this species the structure and organization of the rDNAs in the genome are limited, and it represents an obstacle both in their genetic research and improvement as well. rDNA copy number variations per eukaryotic genome have been considered as a source of genetic rearrangements. In this study, the copy number of 18S and 5S rDNAs in the A. tequilana genome was estimated, and an absolute quantitative qPCR assay and genome size was used. In addition, an association between the rDNAs copy number and physical mapping was performed to confirm our results. Results: The analysis were successfully applied to determine copy number...

Restriction fragment analysis of the ribosomal DNA of Paratelmatobius and Scythrophrys species (Anura, Leptodactylidae) Genet. Mol. Biol.
Lourenço,Luciana B.; Garcia,Paulo C.A.; Recco-Pimentel,Shirlei M..
Physical maps of the ribosomal RNA gene 28S of species belonging to the genera Paratelmatobius and Scythrophrys were constructed, using five restriction endonucleases. The restriction sites for Bam HI, Bgl II, Bst EII, and Eco RI had similar positions in all species, although there were interspecific differences in the size of the restriction fragments obtained. An additional Pvu II site was found in Scythrophrys specimens from Piraquara (State of Paraná, Brazil) and from São Bento do Sul (State of Santa Catarina, Brazil), but not in the Scythrophrys specimens from Rancho Queimado (State of Santa Catarina, Brazil). This finding is in agreement with the hypothesis regarding the existence of two species in the genus Scythrophrys. On the other hand, the extra...

ITS2 variability of Biomphalaria (Mollusca, Planorbidae) species from the Paranapanema Valley (São Paulo State, Brazil): diversity patterns, population structure, and phylogenetic relationships Genet. Mol. Biol.
Tuan,Roseli; Santos,Priscila dos.
The ribosomal DNA internal transcribed spacer 2 (ITS2) has been shown to be a useful genetic marker for species identification and phylogenetic reconstruction in the genus of freshwater snails Biomphalaria (Preston 1910). Additionally, ITS2 studies in Biomphalaria have uncovered significant intra-specific genetic variability suggesting the presence of cryptic species complexes. We obtained ITS2 sequences for the Biomphalaria species B. glabrata, B. tenagophila, B. occidentalis and B. peregrina from the Paranapanema Valley (São Paulo State, Brazil) and compared them with a comprehensive set of published Biomphalaria ITS2 sequences using Bayesian inference of phylogeny. Analysis of the resulting trees showed that the newly obtained B. glabrata sequences did...

Comparative cytogenetic analysis of two grasshopper species of the tribe Abracrini (Ommatolampinae, Acrididae) Genet. Mol. Biol.
Rocha,Marília de França; Melo,Natoniel Franklin de; Souza,Maria José de.
The grasshopper species Orthoscapheus rufipes and Eujivarus fusiformis were analyzed using several cytogenetic techniques. The karyotype of O. rufipes, described here for the first time, had a diploid number of 2n = 23, whereas E. fusiformis had a karyotype with 2n = 21. The two species showed the same mechanism of sex determination (XO type) but differed in chromosome morphology. Pericentromeric blocks of constitutive heterochromatin (CH) were detected in the chromosome complement of both species. CMA3/DA/DAPI staining revealed CMA3-positive blocks in CH regions in four autosomal bivalents of O. rufipes and in two of E. fusiformis. The location of active NORs differed between the two species, occurring in bivalents M6 and S9 of O. rufipes and M6 and M7 of...

Pires,Larissa Bettin; Usso,Mariana Campaner; Giuliano-Caetano,Lucia; Dias,Ana Lúcia.
Abstract The genera Cichlasoma and Gymnogeophagus belong to the subfamily Cichlinae, the only one in Neotropical cichlids. Cichlasoma dimerus, C. paranaense, C. portalegrense, Gymnogeophagus rhabdotus, and G. lacustris were collected at different points in the Paranapanema and Paraguay basins and the Lagoon of Patos hydrographic system. In addition to conventional analysis, CMA3 fluorochrome staining, and FISH with 18S rDNA probe were performed. All species had a diploid number equal to 48, with interand intraspecific differences in karyotype formulae. All species presented a single AgNOR site, except G. rhabdotus and the C. paranaense population of the Paranapanema River, which revealed more than one pair of nucleolar chromosomes. AgNORs were coincident...

Karyotype diversity between species of Crenicichla (Perciformes, Cichlidae) from different Brazilian hydrographic basins Genet. Mol. Biol.
Pires,Larissa Bettin; Usso,Mariana Campaner; Giuliano-Caetano,Lucia; Dias,Ana Lúcia.
Abstract Crenicichla is the largest genus in the Cichlidae family in South America. The genus includes 100 valid species that are popularly known in Brazil as jacundás or joaninhas and are widely distributed in rivers east of the Andes. Cytogenetic analyses were carried out on seven species in this genus. All species showed a diploid number of 48 with interspecific differences in karyotype formulas and AgNORs located in interstitial position on the short arm of the largest metacentric pair, except for the two populations from C. britskii. Population A showed terminal markings on the long arm of the fifth pair of the complement, and population B showed up to two marked chromosome pairs. FISH with an 18S rDNA probe was coincident with AgNORs and CMA3, except...

Taxonomy and phylogenetic analysis of Aspergillus section nigri isolated from yerba mate in Misiones (Argentina) JBAG
Castrillo,M.L; Fonseca,M.I; Bich,G.A; Jerke,G; Horianski,M.A; Zapata,P.D.
Members of Aspergillus section Nigri are distributed worldwide and are potentially ochratoxin A producers; some of them are used at industrial level as source of extracellular enzymes and organic acids. The taxonomy of this section comprises one of the most complex and confusing of genus Aspergillus. The objectives of present work were to study the phylogeny of Aspergillus section Nigri and their taxonomy by amplifying ITS1, 5.8S and ITS2 regions. We analyzed 14 strains of Aspergillus section Nigri isolated from different commercial forms of yerba mate, classified according to morphology. Genomic DNA was extracted, the ITS regions were amplified with universal primers ITS1 and ITS4, and the obtained fragments were sequenced. The obtained sequences were...

Molecular identification of Trichogramma cacoeciae Marchal (Hymenoptera: Trichogrammatidae): a new record for Peru Neotropical Entomology
Almeida,Raul P. de; Stouthamer,Richard.
Discovery of male morphological characters for species identification was a great improvement in Trichogramma systematic. However, not all species could be easily identified. In some cases, the lack of males (thelytokous status of species that carry the Wolbachia symbiont) made Trichogramma identification impossible. This problem was solved via antibiotic and heating treatments for elimination of the bacteria and allowing the production of males. The only Trichogramma species reported in which thelytoky is not induced by bacterial infection is T. cacoeciae, so here another means of species identification is needed. This species was identified based on the ITS2 (Internal transcribed spacer 2) sequence, a modern technique that has been proved useful in...

Cytogenetics of two Farlowella species (Loricariidae: Loricariinae): implications on the taxonomic status of the species Neotropical Ichthyology
Marajó,Leandro; Viana,Patrik F.; Ferreira,Milena; Py-Daniel,Lúcia H. Rapp; Feldberg,Eliana.
ABSTRACT Farlowella is one of the most diverse genera of the Loricariinae, restricted to South America rivers. The taxonomic and phylogenetic relationships among its species are contentious and, while genetic studies would contribute to the understanding of their relationships, the only available datum refer to the karyotype description of only one species. In the present study two Amazonian species, Farlowella cf. amazonum and F. schreitmuelleri, were analyzed using conventional and molecular cytogenetic procedures. Both species had diploid chromosome number 58, but different fundamental numbers (NF) 116 and 112, respectively, indicative of chromosomal rearrangements. C-banding is almost poor, especially in F. cf. amazonum, and occurs predominantly in the...

Magnani,Marciane; Fernandes,Thiago; Prete,Cássio Egidio Cavenaghi; Homechim,Martin; Ono,Elisabete Yurie Sataque; Vilas-Boas,Laurival Antonio; Sartori,Daniele; Furlaneto,Márcia Cristina; Fungaro,Maria Helena Pelegrinelli.
Some species belonging to the genus Aspergillus are potential producers of ochratoxin A (OA), a mycotoxin with nephrotoxic, immunosuppressive, teratogenic and carcinogenic effects. The aim of the present study was to identify the species of Aspergillus that contaminate the inside of coffee beans collected in the stage of maturation and drying, from 16 producing areas located in the northern region of the State of Paraná, in the South of Brazil. A total of 108 isolates of Aspergillus spp. was identified at the species level, by sequencing the internal transcribed spacer (ITS1-5.8S-ITS2) of ribosomal DNA (rDNA). The results revealed the presence of potentially ochratoxigenic species in 82% of the geographic regions studied, among which Aspergillus niger was...

Molecular markers in studies on fish parasites (Platyhelminthes): Review Biological Sciences
Graça, Rodrigo Junio da; Fabrin, Thomaz Mansini Carrenho; Gasques, Luciano Seraphim; Prioli, Sônia Maria Alves Pinto; Prioli, Alberto José; Takemoto, Ricardo Massato.
Studies with molecular markers are currently more common for all groups of living organisms. Molecular techniques used in Platyhelminthes parasites of fishes do not merely reveal complex life cycles, but are important for species distinction and the elucidation of the phylogenetic hypothesis. Current research verified which molecular markers were mainly used phylogenetic studies on Platyhelminthes parasites of fish so that subsidies for further phylogenetic studies in Icthyoparasitology could be provided. Data base of CAPES Journals platform was employed for bibliometric analysis comprising the keywords “fish” and “phylogeny” associated with “Cestoda”, “Digenea” or “Monogenea”. Information retrieved was quantified and tabulated. Most studies were on...

Chromosome differentiation in three species of Leptostemonum (Solanum, Solanaceae) endemic to oceanic islands Boletín de la Sociedad
Chiarini,Franco E; Gauthier,Martha J.
The chromosomes of Solanum nelsonii, S. sandwicense (endemic to the Hawaii islands) and S. vespertilio (from the Canary islands) were studied by means of classical staining, CMA/DAPI banding and FISH with probes for the 18-5.8-26S and 5S rDNA genes. The aim of this study was to test for chromosomal changes (chromosome number, karyotype, heterochromatin pattern, rDNA loci) during the evolution of these taxa with respect to their continental relatives. An apparent chromosome stasis was confirmed in all three species, in regards to chromosome number and karyotype morphology. However, there was also evidence of cryptic, cumulative sequence changes. Speciation in these species is not likely associated with large, obvious chromosome rearrangements or polyploidy,...

Genetic variability within Fusarium solani specie as revealed by PCR-fingerprinting based on pcr markers BJM
Brasileiro,Bereneuza Tavares Ramos Valente; Coimbra,Maria Raquel Moura; Morais Jr,Marcos Antonio de; Oliveira,Neiva Tinti de.
Fusarium solani fungus (teleomorph Haematonectria haematococca) is of relevance for agriculture, producing a disease that causes significant losses for many cultivars. Moreover, F. solani is an opportunistic pathogen to animals and humans. The complexity associated to its correct identification by traditional methods justifies the efforts of using molecular markers for isolates characterization. In this work, three PCR-based methods (one PCR-ribotyping and two PCR-fingerprinting) were used to investigate the molecular variability of eighteen F. solani isolates from four Brazilian States, collected from different substrates. Genetic analysis revealed the intraspecific variability within the F. solani isolates, without any correlation to their geographical...

Reclassification of Candida guilliermondii FTI 20037 as Candida tropicalis based on molecular phylogenetic analysis BJM
Lima,Luanne Helena Augusto; Felipe,Maria das Graças de Almeida; Torres,Fernando Araripe Gonçalves.
Yeasts of the genus Candida are of clinical importance and also have many industrial applications, mainly in the food industry. The yeast Candida guilliermondii FTI 20037 has been extensively studied in order to establish a biotechnological process for the production of xylitol. The goal of this study was to verify the taxonomic classification of this strain based on the analysis of rDNA sequences and the xyl1 gene. DNA fragments from these sequences were amplified by PCR and BLAST analysis revealed strong identity with the corresponding sequences from Candida tropicalis. Based on these results, we propose that C. guilliermondii FTI 20037 must be reclassified as C. tropicalis.
