CitEST project resulted in the construction of cDNA libraries from different Citrus sp. tissues under various physiological conditions. Among them, plantlets of Rangpur lime were exposed to hydroponic conditions with and without water stress using PEG6000. RNA from roots was obtained and generated a total of 4,130 valid cDNA reads, with 2,020 from the non-stressed condition and 2,110 from the stressed set. Bioinformatic analyses measured the frequency of each read in the libraries and yielded an in silico transcriptional profile for each condition. A total of 40 contigs were differentially expressed and allowed to detect up-regulated homologue sequences to well known genes involved in stress response, such as aquaporins, dehydrin, sucrose synthase, and... |

A procedure is described to regenerate plants from protoplasts of Brazilian citrus cultivars, after isolation, fusion and culture. Protoplasts were isolated from embryogenic cell suspension cultures and from leaf mesophyll of seedlings germinated in vitro. The enzyme solution for protoplast isolation was composed of mannitol (0.7 M), CaCl2 (24.5 mM), NaH2PO4 (0.92 mM), MES (6.15 mM), cellulase (Onozuka RS - Yakult, 1%), macerase (Onozuka R10 - Yakult, 1%) and pectolyase Y-23 (Seishin, 0.2%). Protoplast culture in liquid medium after chemical fusion lead to the formation of callus colonies further adapted to solid medium. Somatic embryo formation occurred spontaneously after two subcultures, on modified MT medium supplemented with 500 mg/L of malt extract.... |

The objective of this work was to evaluate the effect of grafting (onto Solanum torvum Sw.) on plant growth, yield and fruit quality of the Pala and Faselis eggplant (Solanum melongena L.) cultivars, grown in a soil infested with Verticillium dahliae Kleb. and Meloidogyne incognita, or in noninfested soil. Soil infestation decreased yield, plant height, final above-ground biomass, and also reduced fruit mean weight and shoot dry weight depending on cultivar or grafting. Grafting decreased fruit oxalic acid and the soluble solid contents, and increased mean fruit weight, depending on cultivar and soil infestation. Grafting also reduced the negative effects of the pathogens on disease index, plant height and shoot dry weight. Cultivar Pala was more vigorous... |

The objective of this work was to evaluate the response of rangpur lime (Citrus limonia) to arbuscular mycorrhiza (Glomus intraradices), under P levels ranging from low to excessive. Plants were grown in three levels of soluble P (25, 200 and 1,000 mg kg-1), either inoculated with Glomus intraradices or left noninoculated, evaluated at 30, 60, 90, 120 and 150 days after transplanting (DAT). Total dry weight, shoot P concentration and specific P uptake by roots increased in mycorrhizal plants with the doses of 25 and 200 mg kg-1 P at 90 DAT. With 1,000 mg kg-1 P, mycorrhizal plants had a transient growth depression at 90 and 120 DAT, and nonmycorrhizal effects on P uptake at any harvesting period. Root colonization and total external mycelium correlated... |

Abstract: The objective of this work was to evaluate the effect of seed stratification on germination rate, germination speed, and initial development of seedlings of six pecan (Carya illinoinensis) cultivars under subtropical climatic conditions in southern Brazil. For stratification, the seeds were placed in boxes with moist sand, in a cold chamber at 4°C, for 90 days. In the fourteenth week after sowing, the emergence speed index, total emergence, plant height, stem diameter, and number of leaves were evaluated. Seed stratification significantly improves the germination potential and morphological traits of the evaluated cultivars. |

Abstract The Cabernet Sauvignon (CS) cultivar is one of the most widely planted wine grapes in Brazil. The final grape quality depends on the clone type, rootstock, and the terroir. This study aims to assess and characterize the effects of seven combinations of CS clones grafted onto two rootstocks on the physical–chemical parameters and productivity of the grapes in the highland of southern Brazil, during the 2012-2013, 2013-2014, 2014-2015, and 2015-2016 growing seasons. The experiment was carried out in a 14-year-old commercial vineyard, located in São Joaquim municipality, Santa Catarina State, trained on a vertical shoot position system on a double cordon with spur pruning. The treatments consisted in a completely randomized experimental block of the... |

Abstract Trifoliate orange is one of the rootstocks most used worldwide mainly because it provides excellent quality fruit production. Harvest at the correct time is among factors that determine the physiological quality of seeds. The aim of this study was to identify physicochemical changes in ‘Limeira-IAC382’ trifoliate orange fruits, searching for the ideal harvest moment to obtain high quality seeds. Fruits were harvested at 180, 210 and 240 days after anthesis, stored in cold chamber and evaluated at 0, 15, 30 and 45 days of storage. The following variables were determined: mass loss, fruit color index, soluble solids, total titratable acidity and fruit juice ratio. Seeds were manually extracted and dried until reaching 30% water content. Seeds were... |
