Association of a PIT1 gene polymorphism with growth hormone mRNA levels in pig pituitary glands Genet. Mol. Biol.
Franco,Maurício Machaim; Antunes,Robson Carlos; Oliveira,Katiana Mello de; Pereira,Cícero Donizete; Biase,Fernando Henrique; Nunes,Francis de Morais Franco; Goulart,Luiz Ricardo.
Fourty-six non-castrated, halothane-free, male Landrace pigs were genotyped by PCR-RFLP for the Rsa I polymorphism in the PIT1 gene and classified into AA and AB genotypes. Total RNA was extracted from the pituitaries and the relative quantities of growth hormone (GH) mRNA were determined by semi-quantitative RT-PCR. Pigs with the AB genotype had higher levels of GH mRNA than those with the AA genotype (p = 0.034; Kruskal-Wallis test). This result suggests that the Rsa I polymorphism may be involved in Pit-1 protein expression or function, which in turn may influence GH transcription and expression. Thus, the Rsa I PIT1 gene polymorphism in this pig line may be used as a molecular marker to identify higher GH expression and possibly select for carcass and...

Differential expression of the KLK2 and KLK3 genes in peripheral blood and tissues of patients with prostate cancer Genet. Mol. Biol.
Meola,Juliana; Goulart,Luiz R.; Oliveira,Jaqueline D.D.; Neves,Adriana F.; Oliveira Jr.,Waldesse P.; Saraiva,Ana C.M.; Capaneli,Andréia C.; Cardoso,Alexandra M.; Prado,Lindolfo D.; Borba,Sebastião A.; Silva,Heyder D..
We used the multiplex semi-quantitative reverse-transcriptase PCR (RT-PCR) to investigate kallikrein 2 and 3 (KLK2 and KLK3) mRNA levels in prostate tissue from 42 prostate cancer patients, 33 of whom were also assessed for peripheral blood KLK2 expression by qualitative semi-nested RT-PCR. We found that KLK2 was an important tissue biomarker for distinguishing between prostate cancer patients and those with benign prostatic hyperplasia, particularly when KLK2 expression was > 60% of that of the beta2-microglobulin constitutive gene. Patients with an average relative expression value > 0.6 (cutoff value) had an eleven-fold higher chance of having prostate cancer. When one or two tissues samples were evaluated for KLK2 expression using the cutoff...

Expression of NCED gene in colored cotton genotypes subjected to water stress AGRIAMBI
Souza,Alexandre M. S. de; Batista,Vandré G. L.; Pinheiro,Morganna P. N.; Suassuna,Janivan F.; Lima,Liziane M. de; Fernandes,Pedro D..
ABSTRACT Considering that the NCED gene acts on the biosynthetic cascade of ABA, a hormone involved in the functioning of stomata and consequently in the regulation of transpiration, the aim of this research was to analyze the expression of this gene in colored cotton genotypes subjected to water stress at the beginning of plant growth. Four colored cotton genotypes were used, subjected to two managements, with and without water stress, beginning the treatments when the blade of the first true leaves reached an area that allowed the evaluation of gas exchange. For the studies of the expression of the NCED gene, via RT-qPCR, leaves were collected on three distinct dates: at 4 and 6 days of water stress, and after the plants regained their turgor. The...

Molecular characteristics, recombinant expression and activity detection of OsGSTL1 from rice BABT
Hu,Tingzhang; Yang,Yongwei; Tan,Lili; Yang,Junnian; Wu,Yingmei.
The mRNA of OsGSTL1 was detected in the roots and leaves of rice plants at seedling and tillering stages, and their roots, leaves and panicles at the heading stage. The full-length open reading frame of OsGSTL1 cDNA was 732 bp and encoded a putative polypeptide of 243 amino acids with a calculated molecular mass of 27.30 kDa and a theoretical pI of 5.50. The protein sequences of OsGSTL1 exhibited typical feature of the lambda class GST, which contained the conserved domain "GST_C_Lambda" in C-terminal alpha helical domain and a highly conserved Cys42 in active center. In silico predictions showed that the OsGSTL1 protein was strongly hydrophilic. The phylogenetic analysis revealed OsGSTL1 belonged to monocots subgroup and was closer to IN2-1 of Z. may. The...

NaCl stress impact on the key enzymes in glycolysis from Lactobacillus bulgaricus during freeze-drying BJM
Li,Chun; Sun,Jinwei; Qi,Xiaoxi; Liu,Libo.
Abstract The viability of Lactobacillus bulgaricus in freeze-drying is of significant commercial interest to dairy industries. In the study, L.bulgaricus demonstrated a significantly improved (p < 0.05) survival rate during freeze-drying when subjected to a pre-stressed period under the conditions of 2% (w/v) NaCl for 2 h in the late growth phase. The main energy source for the life activity of lactic acid bacteria is related to the glycolytic pathway. To investigate the phenomenon of this stress-related viability improvement in L. bulgaricus, the activities and corresponding genes of key enzymes in glycolysis during 2% NaCl stress were studied. NaCl stress significantly enhanced (p < 0.05) glucose utilization. The activities of glycolytic enzymes...
