Characterization of a multi-tolerant tannin acyl hydrolase II from Aspergillus carbonarius produced under solid-state fermentation Electron. J. Biotechnol.
Valera,Larissa Serrani; Jorge,João Atílio; Guimarães,Luis Henrique Souza.
Background Tannases are enzymes with biotechnological potential produced mainly by microorganisms as filamentous fungi. In this context, the production and characterization of a multi-tolerant tannase from Aspergillus carbonarius is described. Results The filamentous fungus A. carbonarius produced high levels of tannase when cultivated under solid-state fermentation using green tea leaves as substrate/carbon source and tap water at a 1:1 ratio as the moisture agent for 72 h at 30°C. Two tannase activity peaks were obtained during the purification step using DEAE-Cellulose. The second peak (peak II) was purified 11-fold with 14% recovery from a Sepharose CL-6B chromatographic column. The tannase from peak II (tannase II) was characterized as a heterodimeric...

Xylanolytic enzymes production by Aspergillus niger GS1 from solid-state fermentation on corn stover and their effect on ruminal digestibility Electron. J. Biotechnol.
Regalado,Carlos; Vázquez-Obregón,Immer; García-Almendárez,Blanca Estela; Domínguez-Domínguez,Jorge; Aguilera-Barreyro,Araceli; Amaro-Reyes,Aldo.
Hemicellulosic agricultural by-products such as corn stover (CS) are highly available materials which represent an opportunity to develop value added products. Native Aspergillus niger GS1 was used for solid-state fermentation (SSF) on alkali pre-treated CS (ACS) aimed to optimize xylanolytic enzymes production, and their effect on in vitro ruminal and true digestibility of ACS. Enzyme production was empirically modelled using a fractional factorial design 2(9-5), and the resulting significant factors were glucose, yeast extract and two mineral salts, which were arranged in a Draper-Lin optimization design at two levels. Predicted optimum xylanolytic activity of 33.6 U (mg protein)-1 was achieved at 48 hrs of SSF, and was validated by confirmatory...

APPLICATION OF SOLID-STATE FERMENTATION FOR CELLULASE ENZYME PRODUCTION USING TRICHODERMA VIRIDE AgEcon
Mojsov, Kiro.
The Solid-state fermentation (SSF) is alternative to submerged fermentation for production antibiotics, single cell protein, enzymes, organic acids, biofuel, etc. However, the advantages of SSF in various processes are found to be greater than in submerged fermentation. This technique not only decreases the cost of the process but also makes product cheaper for consumers. The paper describes experimental application of SSF on wheat straw for production of mycelia protein and cellulase enzymes by Trichoderma viride. This actual waste from agriculture industry was used as a nourishing base by Trichoderma viride in SSF for cellulase enzyme production. Growth and enzymes production by Trichoderma viride were evaluated on wheat straw and alkali treatment...

Ramachandran,Sumitra; Patel,Anil K.; Nampoothiri,Kesavan Madhavan; Chandran,Sandhya; Szakacs,George; Soccol,Carlos Ricardo; Pandey,Ashok.
Solid-state fermentation was carried out for the production of alpha-amylase using Aspergillus oryzae. Different oil cakes such as coconut oil cake (COC) sesame oil cake (SOC), groundnut oil cake (GOC), palm kernel cake (PKC) and olive oil cake (OOC) were screened to be used as substrate for the enzyme production and also compared with wheat bran (WB). GOC was found to be the best producer of the enzyme among these. Combination of WB and GOC (1:1) resulted higher enzyme titres than the individual substrates. Maximum amount of enzyme (9196 U/gds) was obtained when SSF was carried out using WB + GOC, having initial moisture of 64% and supplemented with lactose and ammonium nitrate (1% each) at 30ºC for 72h using 2 mL spore suspension (6x10(7)spores/ml)....

Nie,Guangjun; Zhu,Zhubin; Liu,Fang; Nie,Zhijie; Ye,Yuchao; Yue,Wenjin.
ABSTRACTThe aim of this work was to study the co-production of nattokinase and poly (γ-glutamic acid) by Bacillus subtilis natto with soybean and rice husk under solid-state fermentation (SSF). The results showed that the size of soybean particle and rice husk significantly improved the co-production of nattokinase and poly (γ-glutamic acid), yielding 2503.4 IU/gs and 320 mg/gs, respectively in the improved culture medium composed of 16.7% soybean flour and 13.3% rice husk with 70% water content. The yields increased by approximate 7- and 2-fold factor relative to their original ones. Thus, the co-production of nattokinase and poly (γ-glutamic acid) under SSF could be considered as an efficient method to exploit agro-residues for economical production of...

Biochemical characterisation of a glucoamylase from Aspergillus niger produced by solid-state fermentation BABT
Slivinski,Christiane Trevisan; Machado,Alex Vinicius Lopes; Iulek,Jorge; Ayub,Ricardo Antônio; Almeida,Mareci Mendes de.
In this work, glucoamylase was produced by Aspergillus niger in solid-state fermentation. The enzyme was partially purified by ammonium sulphate precipitation and ion exchange and gel filtration chromatographies. Its molecular mass was estimated as 118.17 kDa by electrophoresis. The partially purified enzyme had an optimum pH range of 4.5-5.0 and an optimum temperature of 60 °C, with average activity 152.85 U mL-1. Thermal and pH stability assays with the crude extract showed that more than 60 % of the activity remained at pH 4.6 and 60 °C, even after an exposition to these conditions longer than 24 h. Yet, after purification, the enzyme was stable at these for at least 4 h, which indicated that its purification for use in starch saccharification was...

Rice bran as a substrate for proteolytic enzyme production BABT
Sumantha,Alagarsamy; Deepa,Paul; Sandhya,Chandran; Szakacs,George; Soccol,Carlos Ricardo; Pandey,Ashok.
Rice bran was used as the substrate for screening nine strains of Rhizopus sp. for neutral protease production by solid-state fermentation. The best producer, Rhizopus microsporus NRRL 3671, was used for optimizing the process parameters for enzyme production. Fermentation carried out with 44.44 % initial moisture content at a temperature of 30 C for 72 h was found to be the optimum for enzyme secretion by the fermenting organism. While most of the carbon supplements favored enzyme production, addition of casein resulted in a marginal increase in protease yield. Fermentation was then carried out under optimized conditions to obtain the crude extract of the enzyme, which was partially purified by precipitation and dialysis. A 3-fold increase in the enzyme...
