Genotoxicity assessment of a pharmaceutical effluent using four bioassays Genet. Mol. Biol.

Pharmaceutical industries are among the major contributors to industrial waste. Their effluents when wrongly handled and disposed of endanger both human and environmental health. In this study, we investigated the potential genotoxicity of a pharmaceutical effluent, by using the Allium cepa, mouse- sperm morphology, bone marrow chromosome aberration (CA) and micronucleus (MN) assays. Some of the physico-chemical properties of the effluent were also determined. Comportamento sexual do macho caprino da raça Moxotó às variações estacionais no Nordeste do Brasil. Infoteca-e

Resumo: Dez bodes da raca moxoto em idade de 24 a 26 meses foram submetidos durante dois anos a duas colheitas de ejaculados espermaticos semanais em vagina artificial. Os animais foram mantidos em pastagem nativa de caatinga raleada. Na avaliacao quantitativa do semen, encontrou-se maior volume na estacao chuvosa do que na seca. A concentracao de espermatozoides no ejaculado foi inversamente proporcional ao volume, portanto variando com as epocas do ano. Na estacao seca a porcentagem de espermatozoides vivos foi menor do que na estacao chuvosa. Estacionalidade na atividade sexual e qualidade do sêmen nos ovinos deslanados das raças Santa Inês e Somalis brasileira. Infoteca-e

Resumo: Em dez ovinos deslanados das racas Santa Ines e Somalis brasileira, sendo cinco de cada, foi coletado e ejaculado espermatico, atraves da vagina artificial, durante um ano. O objetivo foi avaliar a atividade sexual do macho ovino, nas condicoes do Nordeste brasileiro, no periodo seco (agosto a dezembro de 1983) e chuvoso (janeiro a maio de 1984) e as diferencas entre racas. Na avaliacao do esperma houve uma diferenca (P<0,001) entre racas, a Santa Ines mostrando maior volume e concentracao do que a Somalis brasileira. Investigating the roles of chemical factors stimulating and inhibiting sperm motility is required to understand the mechanisms of spermatozoa movement. In this study, we described the composition of the seminal fluid (osmotic pressure, pH, and ions) and investigated the roles of these factors and salinity in initiating spermatozoa movement in the Pacific oyster. The acidic pH of the gonad (5.82 ± 0.22) maintained sperm in the quiescent stage and initiation of flagellar movement was triggered by a sudden increase of spermatozoa external pH (pHe) when released in seawater (SW). Cryopreservation is a valuable tool for genetic improvement programs. Several bivalve mollusc species have already been the subject of such programs and the Tahitian black pearl oyster industry is now planning the development of selective breeding for desirable traits in Pinctada margaritifera. The ability to cryopreserve spermatozoa would, therefore, offer significant benefits to the cultured black pearl industry. Changes in Atlantic cod (Gadus morhua L.) sperm quality during the spawning season ArchiMer

The biology of cod reproduction is well described in the scientific literature. However, sperm biology and spermatozoa management are poorly studied in this species. Because of its recent farming expansion, a better knowledge of cod gametes is becoming especially useful. This work aimed at establishing tools to study sperm biology in cod, and also investigated the existence of changes in cod sperm quality during the spawning period. We showed that sperm concentration could be assessed using spectrophotometry at 260 nm. Sperm motility significantly decreased after a 168-h storage at 4 degrees C. Cryopreservation of sea bass (Dicentrarchus labrax) spermatozoa in experimental and production simulating conditions ArchiMer

A sperm cryopreservation protocol adapted from turbot, was tested on sea bass using either 250-mu L straws or 1.5-mL cryovials. A dilution to 1/3 in Mounib's extender and a cooling rate of 65 degrees C.min(-1) allowed frozen sperm to recover an initial motility similar to that of fresh sperm at thawing; however, significant differences in motility (P < 0.001, n = 10 fish semen) were observed at further post-activation times, the motility decrease being faster in thawed sperm. At the experimental scale, triplicate inseminations of 2-mL aliquots (approximately 2 000 eggs) showed a significant fertility decay of thawed sperm compared to that of fresh sperm (P < 0.01, n = 12 fish semen) when a discriminating 35.10(3) spermatozoa to egg ratio was applied....

Motility characteristics of spermatozoa in cod (Gadus morhua) and hake (Merluccius merluccius) ArchiMer

As in many marine fish, motility is triggered by osmotic pressure (OP) in cod and hake spermatozoa and motility ranges several minutes with a high velocity (above 50 mu m/s) period restricted to 100 s. A decrease in the flagellar beat frequency (BF) is partly responsible of this briefness. Detailed observations of the flagella during the motility period showed: 1) sea water provokes OP damages (blebs) which impair the correct wave propagation 2) waves become restricted to the proximal flagellum and tip becomes devoid of wave 3) the wave amplitude decreases. The combination of these factors drastically limits to the earliest period of motility the ability for spermatozoa to efficiently progress towards egg for fertilization.

The bucks were first used as control and later as experimental animals upon being fed daily with oiled pumpkin plant for the period of six months. Spermatogenesis and the spermatozoon ultrastructure follow a common pattern in terrestrial oligochaetes. However, many of their characteristics are distinctive of each species and they are of great importance for phylogenetic and taxonomic studies. Germinal cells were studied through conventional electronic microscope techniques in order to go deep on spermatogenesis, especially on the mature spermatozoon ultrastructure of Eisenia foetida. Spermiogram and Morphological Characteristics in Testicular and Epididymal Spermatozoa of Large White Boar in Nigeria International Journal of Morphology

Ten healthy male (boar) of large white pigs aged between 6.0 and 18.0 months, were used for this study. Spermiogram and morphological characteristics were studied in testes and in the different parts of the epididymis of the boar. The objective was to study the changes that occur in testes and epididymis during epididymal transit. Progressive motile spermatozoa were first observed in the testes (6-10%) in the caput (20-30%), corpus (40-50%) and highest at the caudal epididymis (85%). Cytoplasmic droplets were lost as the spermatozoa moved up the epididymis. Many of the studies were aimed at understanding the action of the venom on the muscular system. Now accepted that LTX is able to generate a calcium-permeable membrane pore and modulate the release of synaptic vesicles that activate a receptor and induce cellular changes. Morphological Study of the Cytoplasmic Droplets as an Index of Sperm Maturation in Black Bengal Buck (Capra hircus) International Journal of Morphology

Location of the cytoplasmic droplets (CD) and their dimensions varied significantly (p<0.01) when sperm cells traverse through the regions of caput, corpus and cauda epididymis and vasdeferens respectively. The gradual diminution in the morphology of CD between the epididymal regions were related significantly (p<0.01, p<0.05). Caudal shift of the CD, along with regression in size and finally their exclusion from the sperm cells reflected one of the most important events in the maturation process of male gametes in Black Bengal buck.

Testicular Parameters and Sperm Morphology of Chinchilla Rabbit Fed With Different Planes of Soymeal International Journal of Morphology

The testicular parameters and sperm morphology were studied in twenty healthy Chinchilla breed bucks of rabbits fed with different planes of soybean meal. Their ages were between 6 to 10 months with their weights ranging from 1.4 to 1.9kg.The same quality of feeds was fed to each group throughout the study and at the rate of 0.5kg/head/day. The feeds contained graded percentage of soybean meal and growers mash. Semen used in this study was from the caudal epididymides while morphological aberrations were determined from a total count of 400 spermatozoa in smears obtained with Wells and Awa stain.
