Pharmaceutical industries are among the major contributors to industrial waste. Their effluents when wrongly handled and disposed of endanger both human and environmental health. In this study, we investigated the potential genotoxicity of a pharmaceutical effluent, by using the Allium cepa, mouse- sperm morphology, bone marrow chromosome aberration (CA) and micronucleus (MN) assays. Some of the physico-chemical properties of the effluent were also determined. The A. cepa and the animal assays were respectively carried out at concentrations of 0.5, 1, 2.5, 5 and 10%; and 1, 5, 10, 25 and 50% of the effluent. There was a statistically different (p < 0.05), concentration-dependent inhibition of onion root growth and mitotic index, and induction of... |

Investigating the roles of chemical factors stimulating and inhibiting sperm motility is required to understand the mechanisms of spermatozoa movement. In this study, we described the composition of the seminal fluid (osmotic pressure, pH, and ions) and investigated the roles of these factors and salinity in initiating spermatozoa movement in the Pacific oyster. The acidic pH of the gonad (5.82 ± 0.22) maintained sperm in the quiescent stage and initiation of flagellar movement was triggered by a sudden increase of spermatozoa external pH (pHe) when released in seawater (SW). At pH 6.4, percentage of motile spermatozoa was three times higher when they were activated in SW containing 30 mM NH4Cl, which alkalinizes internal pH (pHi) of spermatozoa, compared... |

The development of hatchery production techniques for blue mussels is receiving increased research attention in Europe as it could offer an alternative source of spat for the industry ('Blue Seed' Project, Kamermans ICSR 2007). Among the necessary techniques are those of controlled spawning and fertilization, and techniques of triploidisation that can offer increased seasonal availability, yield and/or decreased growing time in the subsequent production stages. In several experiments, Mytilus edulis oocytes, obtained by thermally induced spawning, were fertilized with Mytilus edulis spermatozoa obtained in the same way or by stripping. Parallel treatments examined the effect of mixing Mytilus edulis oocytes with spermatozoa stripped from the Pacific... |

Sperm characteristics of scallops have not been well described in the scientific literature. The effects of sperm release technique (thermal shock versus serotonin injection), of sperm collection technique (testis sampling versus serotonin injection), of sperm sampling location along the genital tract, of in vitro sperm maturation, and of time post activation on scallop sperm characteristics were assessed in the present work. Whatever sperm release technique used, no significant differences were observed regarding the percentage of motile spermatozoa and the velocity of the average path (YAP). Compared to testicular sperm, a higher percentage of motile spermatozoa, YAP and intracellular adenosine triphosphate (ATP) content were observed for sperm shed... |

Cryopreservation is a valuable tool for genetic improvement programs. Several bivalve mollusc species have already been the subject of such programs and the Tahitian black pearl oyster industry is now planning the development of selective breeding for desirable traits in Pinctada margaritifera. The ability to cryopreserve spermatozoa would, therefore, offer significant benefits to the cultured black pearl industry. Spermatozoa were cryopreserved with CPA 0.7 M trehalose in 0.8 M Me2SO and a two-step freezing process was used: straws were first maintained in nitrogen vapour for 10 minutes, then directly plunged into liquid nitrogen and stored for one week before use. The viability of thawed sperm was 23 % lower than that of fresh sperm. When using thawed... |

The biology of cod reproduction is well described in the scientific literature. However, sperm biology and spermatozoa management are poorly studied in this species. Because of its recent farming expansion, a better knowledge of cod gametes is becoming especially useful. This work aimed at establishing tools to study sperm biology in cod, and also investigated the existence of changes in cod sperm quality during the spawning period. We showed that sperm concentration could be assessed using spectrophotometry at 260 nm. Sperm motility significantly decreased after a 168-h storage at 4 degrees C. A 1:9 dilution of sperm in a non-activating medium (1/3 seawater and 2/3 freshwater, osmotic pressure: 360 mOsm kg(-1)) improved sperm storage. Sperm concentration,... |

A sperm cryopreservation protocol adapted from turbot, was tested on sea bass using either 250-mu L straws or 1.5-mL cryovials. A dilution to 1/3 in Mounib's extender and a cooling rate of 65 degrees C.min(-1) allowed frozen sperm to recover an initial motility similar to that of fresh sperm at thawing; however, significant differences in motility (P < 0.001, n = 10 fish semen) were observed at further post-activation times, the motility decrease being faster in thawed sperm. At the experimental scale, triplicate inseminations of 2-mL aliquots (approximately 2 000 eggs) showed a significant fertility decay of thawed sperm compared to that of fresh sperm (P < 0.01, n = 12 fish semen) when a discriminating 35.10(3) spermatozoa to egg ratio was applied.... |

As in many marine fish, motility is triggered by osmotic pressure (OP) in cod and hake spermatozoa and motility ranges several minutes with a high velocity (above 50 mu m/s) period restricted to 100 s. A decrease in the flagellar beat frequency (BF) is partly responsible of this briefness. Detailed observations of the flagella during the motility period showed: 1) sea water provokes OP damages (blebs) which impair the correct wave propagation 2) waves become restricted to the proximal flagellum and tip becomes devoid of wave 3) the wave amplitude decreases. The combination of these factors drastically limits to the earliest period of motility the ability for spermatozoa to efficiently progress towards egg for fertilization. |

Fertilization of the oocyte is a critical process of sexual reproduction depending among other factors on spermatozoa functionality. Mitochondria participate in many crucial processes for spermatozoa motility and fertilizing ability. The Pacific oyster, Crassostrea gigas, is characterized by a high inter-individual variability of its reproductive success which has been shown to be partially due to the variability of gamete quality. The present study explored spermatozoa characteristics such as: i) relationships among mitochondrial functionality (ATP content, mitochondrial membrane potential (MMP), reactive oxygen species production); sperm viability (percentages of live, dying, and dead); and motility; and ii) their involvement in inter-oyster variability... |

The 2010 Deepwater Horizon (DWH) oil spill released millions of barrels of oil and dispersant into the Gulf of Mexico. The timing of the spill coincided with the spawning season of Crassostrea virginica. Consequently, gametes released in the water were likely exposed to oil and dispersant. This study aimed to (i) evaluate the cellular effects of acute exposure of spermatozoa and oocytes to surface slick oil, dispersed mechanically (HEWAF) and chemically (CEWAF), using flow-cytometric (FCM) analyses, and (ii) determine whether the observed cellular effects relate to impairments of fertilization and embryogenesis of gametes exposed to the same concentrations of CEWAF and HEWAF. Following a 30-min exposure, the number of spermatozoa and their viability were... |

Four healthy bucks of the West African Dwarf breed aged between 24 and 30 months were used for this study. The bucks were first used as control and later as experimental animals upon being fed daily with oiled pumpkin plant for the period of six months. The objective of the study was to investigate the effect of the pumpkin plant on the morphology of the spermatozoa of the bucks. There were significant differences (p< 0.05) between the control and experimental values for both primary and secondary morphological spermatozoa abnormalities: the pyriform head has a control value of 6 (0. 42%) and post-feeding value of 0 (0%), the beat tail; 14(0.97%) and 2 (0.16%) for the control and post feeding values respectively. The curved mid piece: 17 (1.18%) and 1... |

Spermatogenesis and the spermatozoon ultrastructure follow a common pattern in terrestrial oligochaetes. However, many of their characteristics are distinctive of each species and they are of great importance for phylogenetic and taxonomic studies. Germinal cells were studied through conventional electronic microscope techniques in order to go deep on spermatogenesis, especially on the mature spermatozoon ultrastructure of Eisenia foetida. Spermatogenesis occurs in seminal vesicles and follows a pattern comparable to that of some other oligachaetes species. Spermatozoon ultrastructural details revealed that it is a filiform cell, with the acrosome placed in anterior position, followed by the nucleus, the midpiece and the tail which flagellum has a 9+2... |

Ten healthy male (boar) of large white pigs aged between 6.0 and 18.0 months, were used for this study. Spermiogram and morphological characteristics were studied in testes and in the different parts of the epididymis of the boar. The objective was to study the changes that occur in testes and epididymis during epididymal transit. Progressive motile spermatozoa were first observed in the testes (6-10%) in the caput (20-30%), corpus (40-50%) and highest at the caudal epididymis (85%). Cytoplasmic droplets were lost as the spermatozoa moved up the epididymis. The caput epididymis had a value of 20.05% of abnormal sperm cell, which is different significantly (p<0.5) from 15 80 % of the caudal epididymis. The total abnormal sperm cells in corpus are also... |

Since the 1970s, There have been studies of the venom of Latrodectus sp. spiders, in particular the latrotoxin (LTX) of Latrodectus mactans. Many of the studies were aimed at understanding the action of the venom on the muscular system. Now accepted that LTX is able to generate a calcium-permeable membrane pore and modulate the release of synaptic vesicles that activate a receptor and induce cellular changes. Interestingly, when work began with venom obtained from the Latrodectus sp present in Chile, it generated clinical indications similar to the bite of this spider in another country, with some differences in intensity. The purpose of the first studies was to understand the systemic mechanisms of this venom, and other active compounds were studied for... |
