Macaw palm (Acrocomia aculeata (Jacq.) Lodd. ex Mart.) is a tropical species with multiple uses, including oil supply for biodiesel production. However, structural and physiological studies of the seed are still scarce, as well as in vitro propagation techniques. The aim of this study was to characterize the anatomy, histochemistry and ultrastructure of the seed and the in vitro somatic embryo of A. aculeata, gaining insight into relationships between the post-harvest seed behavior and the somatic embryo conversion to plant. Acca sellowiana has anatropous, bitegmic and crassinucellate ovules. The outer and inner integuments are double-layered except in the micropyle, where they are composed of more layers; the micropyle is zig-zag shaped. The egg apparatus lies at the micropylar pole, and the zynergids present a conspicuous filiform apparatus. The antipodal cells are present in the chalazal region, persisting before the occurrence of double fertilization. The zygote is visible 21 days after pollination; nuclear endosperm is already present. The first mitotic division of the zygote occurs at 24th day. A simple and efficient protocol for recurrent somatic embryogenesis and plant regeneration is one of the prerequisites for genetic improvement of guava. An efficient reproducible regeneration somatic embryogenesis protocol was developed in four genotypes of Psidium guajava L. using immature zygotic embryo as starter explant.
